DORMANCY BREAK IN SEEDS OF SamaneatubulosaBenthan -(LeguminoseaeMimosoideae)ABSTRACT
-The Caatinga has a wide diversity of species with potential for exploitation, among which is the catingueira (Poincianella pyramidalis (Tul.) L. P. Queiroz), which is notable for its importance in the areas of timber and forestry. The use of rapid tests in quality control programs for seeds is an essential tool for evaluating physiological quality. The aim of this study was therefore to develop procedures for a tetrazolium test to evaluate viability and vigour in seeds of P. pyramidalis. Two procedures for preconditioning the seeds were tested: between moist paper towels and directly in water. In preparing the seeds, both manual scarification and cutting the seed at the opposite end to the hilum were tested. For staining, two concentrations of tetrazolium solution were used (0.075% and 0.15%) and three staining periods (30, 60 and 90 minutes), with the seeds being sorted into classes of viability and vigour. The experimental design was completely randomised in a 3 x 3 x 2 factorial scheme (harvest year x staining time x tetrazolium concentration), with four replications of 25 seeds. The tetrazolium test was efficient in assessing viability and vigour in seeds of P. pyramidalis. After cutting the opposite end to the hilum, the seeds were preconditioned directly in water for 24 hours at 25 °C, when the integument could be removed. Staining was achieved by immersing the seeds in a 0.075% tetrazolium solution for 90 minutes at 41 °C.
Bidens subalternans DC. is a weed found in several tropical countries such as Brazil. Large number of produced seeds and easy dispersion favor the colonization of agricultural fields by this species. To know the factors that affect the germination of B. subalternans can help to understand its ecology, permitting to develop control strategies. Laboratory experiments were carried out to evaluate how the temperature, photoperiod, burial depth, water deficit, and salt stress affect the seed germination of B. subalternans. The means of the treatments of each experiment were shown in scatter plots with the bars indicating the least significant difference (LSD, p0.05). The results showed a germination percentage above 77% for a wide alternating temperature (15/20 C to 30/35 C night/day). The highest germination and uniformity occurred at 25/30˚C night/day. Only 11% of the seeds germinated at a temperature of 35/40˚C night/day. The deeper burial of seeds reduced their germination. Only 17% of the seeds germinated in darkness conditions. However, in constant light and 12 hours of light/dark conditions the germination percentage was over 96%, confirming the light dependence of the B. subalternans during germination. In constant light and 12 hours of light/dark, the germination was over 96%. B. subalternans seeds showed sensitivity to water and salt stress, and their germination was inhibited under a water potential of-0.4 MPa and 100.09 mM, respectively. The sensitivity of B. subalternans seeds to high temperatures, water stress, and salt stress explains the high frequency of this weed in south-central Brazil. The light and sowing depth showed that burial of seeds by mechanical control is a strategy to reduce the high infestation of B. subalternans.
Germination test for papaya seeds demands thirty days to obtain the results, which is a long period for decision-making. Thus, rapid tests become essential tools to evaluate seed viability, and the tetrazolium test is an alternative for this purpose. Hence, this study aimed to establish the adequate procedure to apply this test, in order to reduce the time of evaluation of papaya seeds. For that, four tetrazolium solution concentrations (0.05, 0.1, 0.5 and 1%) and three staining times (3, 6 and 9 hours) were tested at temperatures of 35 and 40 ºC. The experimental design was completely randomized in 4 × 3 + 1 factorial scheme (concentrations × staining times + one control = germination test). It was found that the tetrazolium test allows to evaluate the physiological quality of papaya seeds, thus reducing the time for decision-making. For the test, we recommend using the concentrations of 0.1% for 9 hours or 1% for 6 hours of staining, at temperature of 40 ºC.
A B S T R A C TSeed germination of species that develop in arid and semi-arid soils can be altered by water deficit and abundance of salts in the soil. Knowledge on tolerance limits is fundamental to understand the adaptive capacity of species under such conditions. The objective of this study was to evaluate the physiological changes (germination and vigor) of Mimosa caesalpiniifolia seeds from different regions and submitted to water and saline stresses. To simulate water stress, the osmotic agent polyethylene glycol (PEG 6000) was adjusted to the potentials of 0, -0.2, -0.4, -0.6 and -0.8 MPa, whereas for the saline stress, NaCl solutions were adjusted to the potentials of 0, -0.1, -0.3, -0.5, -0.7; -0.9 and -1.1 MPa. The experimental design was completely randomized, in a 3 x 5 factorial scheme for water stress and 3 x 7 factorial scheme for saline stress, with four replicates of 25 seeds. The variables analysed were: germination, germination speed index and seedling shoot length, root length, shoot dry matter and root dry matter. M. caesalpiniifolia seeds, coming from different geographic regions, showed similar physiological responses to water stress; with respect to salt stress, their tolerance varies according to the region of harvest, and those from Vazante, MG, Brazil were more tolerant during germination and initial growth of seedlings.
