RESUMOO araça-boi (Eugenia stipitata Mc. Vaung) é uma frutífera nativa da Amazônia, pertencente a família Myrtaceae. Sua polpa é consumida na forma de sucos, geleias e sorvetes devido ao aroma e sabor agradáveis. Estudos de diversidade genética através de técnicas moleculares vêm sendo cada vez mais utilizados, mas a primeira etapa é a obtenção de DNA íntegro por meio de técnicas adequadas de isolamento. Este trabalho objetivou estabelecer um protocolo de extração de DNA para Eugenia stipitata visando estudos moleculares. Foram coletadas folhas de E. stipitata, para isolamento do DNA. A extração de DNA foi com base no protocolo CTAB com modificações nas concentrações de CTAB (2 e 5%), β-mercaptoetanol (0, 0,2 e 2 %), PVP (1 e 2 %) e presença e ausência de acetato de amônio (7,5 M). O DNA extraído foi amplificado com dois primers ISSR. A extração de DNA foi possível apenas com CTAB 5%, a quantidade de β-mercaptoetanol interferiu na quantidade de DNA, sendo melhor nas concentrações maiores. Não houve diferenças significativas em relação a concentração de PVP e presença e ausência de acetato de amônio. Os dois primers ISSR amplificaram regiões do genoma de E. stipitata. A extração de DNA de E. stipitata deve ser realizada com CTAB 5% e β-mercaptoetanol a 2%. O acetato de amônio pode ser retirado do protocolo sem prejuízo da qualidade do DNA. Os marcadores ISSR foram eficientes na amplificação e são recomendados para estudos de diversidade genética de E. stipitata.
Genipa americana (Rubiaceae) is an endemic Amazon-region species that may be subject to inbreeding, since it grows in fragmented environments. To examine this possibility, we examined the genetic diversity and population structure of three G. americana populations naturally grown in Northern Mato Grosso State using SSR markers. Sixty-four individuals were sampled from the three populations: 20 in AFL, 20 in MTP, and 24 in NBD. DNA extraction was performed according to the CTAB method, with modifications. Six SSR primers developed for the species were used. The allele frequency, the observed and expected heterozygosity, polymorphism information content), and the fixation Index were estimated. Molecular variance and principal coordinates analyses were conducted, and the most likely number of groups was inferred using the Structure software to help understand the genetic structure of the populations. Ruzza et al 2 in total, ranging from 2 to 4 alleles per locus, with a mean of 2.83. The expected heterozygosity ranged from 0.35 to 0.67 and remained higher than the observed heterozygosity for all loci. The three populations showed genetic diversity, shared genetic material and presented high inbreeding indexes. Cluster analysis results showed genetic material sharing between populations, as well as a lack of genetic structuring among individuals according to their geographical origins. The molecular characterization revealed that the genetic diversity is higher within than between populations. The three populations had shared genetic material and a high inbreeding index due to low observed heterozygosity. This could be a consequence of the fragmented environment where these populations currently live in, since it reduces the number of G. americana individuals and can increase inbreeding.
RESUMO Genipa americana é conhecida popularmente por jenipapo. Objetivamos, com este trabalho, realizar o estudo etnobotânico do jenipapo com agricultores no município de Carlinda, Mato Grosso, almejando conhecer como os agricultores identificam a espécie, quais os tipos de usos, bem como os usos com maior concordância entre os informantes. Vinte moradores de comunidades rurais do município de Carlinda participaram do estudo, selecionados pelo método de "Bola-de-Neve". Os dados foram analisados quali e quantitativamente com a concordância de uso entre os informantes, método adaptado de Amorozo e Gély (1988). Os agricultores relataram reconhecer o jenipapo principalmente pela folha, fruto e porte da árvore. Foram obtidas 30 formas de uso, nas categorias alimentar, tecnologia/artesanal, medicinal e outros. As principais partes utilizadas são folhas, frutos e a casca do tronco para fins medicinais, culinária e tecnologia. As maiores concordâncias de uso entre os agricultores foram para o preparo do licor (CUPc=29,17), suco (CUPc=16,67) e uso do fruto, folha e casca para tratamento dos rins (CUPc = 16,67). Os agricultores não precisam derrubar as árvores para fazer o uso dos recursos da espécie, não põem em risco a diversidade genética, nem a distribuição da espécie na região estudada.
Genipa americana L., commonly known as genipap, belongs to the Rubiaceae family. This study aimed to describe the pollen morphology of the species, evaluate its meiotic behavior and pollen viability, and provide information to help the maintenance and conservation of the species in its natural habitat. Flower buds were collected from 20 individuals in Alta Floresta and Matupá municipalities, Mato Grosso, Brazil. Pollen morphology was characterized using acetolysis and compared to existing literature. Meiotic and post-meiotic phases were analyzed using 2% acetocarmine stain, and pollen viability was estimated using Sudan IV, Alexander's stain, Lugol's solution (1%), and 2% acetocarmine stain. G. americana has medium-sized, 3-colporate pollen with reticulated exine and few meiotic irregularities. Acetocarmine stain showed the highest mean percentage of pollen viability (97.96%). Stain tests revealed significant differences, indicating high pollen viability and meiotic regularity. However, conservation and recovery of degraded areas are still necessary as there is no guarantee of successful reproduction due to factors associated with fragmentation, genetic drift, reduced gene flow, and inbreeding.
