Constraints of the physical environment affect forest growth and forest operations. At a local scale, these constraints are generally considered during forest operations. At regional or continental scales, they are often integrated to larger assessments of the potential for a given land unit to be managed. In this study, we propose an approach to analyze the integration of physicalenvironment constraints in forest management activities at the regional scale (482 000 km 2 ). The land features that pose constraints to forest management (i.e., hydromorphic organic deposits, dead-ice moraines, washed till, glacial block fields, talus, and active aeolian deposits, slopes > 40%) were evaluated within 1114 land districts. To distinguish land districts that can be suitably managed from those where constraints are too important for sustainable timber production, we carried out a sensitivity analysis of physical constraints for the 1114 land districts. After analysis of two portions of the study area under management, a land district was considered suitable for management when more than 20% of its land area consists of features imposing few constraints or, for mountain-type relief districts, when more than 40% of the land area consists of features imposing few constraints. These cutoff values were defined by expert opinion, based on sensitivity analyses performed on the entire study area, on analyses of two different sectors with different types of constraints and on a strong understanding of the study area. Our results show that land districts where the physical environment posed significant constraints covered 7.5% of the study area (36 000 km 2 ). This study shows that doing an a priori classification of land units based on permanent environmental features could facilitate the identification of areas that are not suitable for forest management activities.Résumé : Les contraintes du milieu physique influencent la croissance des forêts et les opérations forestières. Aux échelles régionale ou continentale, elles sont souvent intégrées aux évaluations plus générales du potentiel d'aménagement d'une portion de territoire. Question Recent studies revealed by indirect evidence that lichen–spruce woodlands in the closed‐crown boreal forest are an alternative stable state of black spruce–feathermoss stands. This paper aims to demonstrate and quantify black spruce forest cover regression following successive wildfire events in the closed‐crown forest zone during the 20th century. A case study in north‐eastern North American closed‐crown forest zone is presented. Location The study area is at the centre of the closed‐crown boreal forest zone of eastern Canada (49°59′ N, 71°59′ W). Methods A burned area of 25 ha was selected in which three tree cohorts were visible. The first cohort contains partly buried trunks and stumps of a stand destroyed by a forest fire in 1920, the second contains standing dead trees from a 1995 forest fire and the third contains seedlings established after the 1995 fire. Fires were dated by scar analyses of living trees at the edge of the site. An array of sample plots was established in autumn 1997 to evaluate each cohort in terms of stocking and tree density. Results Black spruce stocking decreased significantly between cohorts, from 43.5% in Cohort 1 (all coniferous trees) to 25.6% in Cohort 2 and 11.5% in Cohort 3 (2011). Jack pine showed an inverse tendency, from 0% in Cohort 1 (not determined) to 0% in Cohort 2 and 3.8% in Cohort 3. Black spruce stems per hectare also dropped significantly from a minimum of 1322 in Cohort 1 to 1088 in Cohort 2 and 976 in Cohort 3. Conclusions This study is the first quantitative demonstration that lichen–spruce woodlands in the closed‐crown boreal forest zone are an alternative stable state of black spruce–feathermoss stands. Stocking levels and densities markedly declined after each of the two fires (1920 and 1995) that both occurred after spruce budworm outbreaks (1910 and 1980) and are most likely responsible for the shift from a closed‐crown to an open woodland.
Breast cancer is the leading cause of cancer related deaths in women worldwide and is classified into subtypes based on the cancer’s receptor status. Of these subtypes, those expressing the human epidermal growth factor receptor 2 (HER2) receptor were traditionally associated with poor prognosis. Several advances have been made in the treatment of HER2-positive breast cancer, yet issues of resistance and poor response to therapy remains prevalent. In this study we explored the impact of HER-family and homologous recombination deficiency SNPs on response to patients who received TCH-based (docetaxel (T), carboplatin (C), and trastuzumab (H)) treatment versus those who received other treatment regimens. Using Cox regression analysis, we identified 6 SNPs that correlate with recurrence free survival in our patients and supported our findings using support vector machines. We also used reverse phase protein array analysis to examine the impact ERBB3 SNPs may have on both the PI3K/AKT and MAPK/ERK signaling pathways. Finally, using cell line models, we correlated SNP status with sensitivity to platinum based drugs and docetaxel. We found that patients on a TCH based regimen with the minor allele of the ERBB3 (rs2229046 and rs773123) and BARD1 (rs2070096) SNPs, were significantly more likely to relapse than those women who were not. Additionally, we observed that patients with these ERBB3 SNPs had shown elevated protein expression/phosphorylation of Src kinase, c-MET (Y1234/1235), GSK-3β (S9) and p27, indicating that these SNPs are associated with non-PI3K/AKT signaling. Finally, using cell line models, we demonstrate that the BARD1 SNP (rs2229571) is associated with greater sensitivity to both carboplatin and cisplatin. The BARD1 and ERBB3 SNPs can potentially be used to determine those patients that will have a worse response to TCH based treatment, an effect that may arise from the SNPs impact on altered cellular signaling.
