Lipases são enzimas que são capazes de catalisar reações químicas e, em específico, catalisam ligações lipídicas. Essas enzimas podem ser obtidas a partir de fungos lipolíticos que por meio de uma fonte de carbono conseguem produzir altos níveis de lipases. Fungos que foram previamente selecionados do bagaço de cana-de-açúcar foram colocados para realizar fermentação submersa utilizando como única fonte de carbono o azeite de oliva extra-virgem. O caldo produzido desses fungos foi analisado por meio de espectrofotometria utilizando como substrato o p-NPB, os resultados obtidos foram analisados por meio de estatística com 5% de significância. Com a presente pesquisa foi possível identificar fungos capazes de crescerem em meio mineral com uma única fonte lipídica de carbono e quantificar atividades específicas de lipases utilizando o substrato p-NPB, foi possível então obter valores promissores de atividade específica lipolítica de três fungos 8,004 U mg-1, 5,897 U mg-1, 5,077 U mg-1, para os fungos Paecilomyces formosus (primeiro isolado), Paecilomyces formosus (segundo isolado) e Aspergillus tubingensis, que são valores citados pela primeira vez para esses três fungos e se comparados a outros microrganismos da literatura são mais altos e ativos para serem utilizados em processos industriais.
Spiranthera odoratissima A. St.-Hil (Rutaceae), a shrub whose common name is manacá do Cerrado in Brazilian Portuguese, is about 1-m high and has been used by folk medicine to treat stomachache, kidney and liver infections, headache, rheumatism and as a blood purifier. This study aimed at preparing hexane, ethyl acetate, methanolic, hydroethanolic and aqueous extracts from S. odoratissima leaves, at carrying out preliminary phytochemical screening and at evaluating their in vitro antioxidant and anti-Listeria monocytogenes activities. Antioxidant activity was evaluated by the DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2-azinobis-3-ethybenzothiazoline-6-sulfonate) and FRAP (ferric reducing antioxidant power) methods. Antibacterial activity was investigated against L. monocytogenes and Minimum Inhibitory Concentration (MIC) values of plant extracts were calculated by the broth microdilution method with the use of 96-well plates. In aqueous, methanolic, hydroethanolic, ethyl acetate and hexane extracts from S. odoratissima leaves, the following classes of compounds were investigated: organic acids, reducing sugars, flavonoids, saponin compounds, coumarin compounds, phenolics, tannins, purine compounds, catechins, flavonol derivatives, sesquiterpene lactones and anthraquinones. All plant extracts, except the hexane one, exhibited high antioxidant activity. Regarding antibacterial activity, the most polar extracts showed high activity against L. monocytogenes; their MIC values ranged between 12.5 and 62.5 µg mL-1, while the hexane one exhibited low activity (MIC = 1000 µg mL-1). In short, extracts from S. odoratissima leaves may be considered promising sources of secondary metabolites with relevant antioxidant and antibacterial activities.
Proveniente de matéria-prima vegetal ou animal, o biodiesel é um combustível renovável que nos últimos anos tem ganhado destaque como um possível substituto dos combustíveis fósseis, principalmente o diesel. O biodiesel é obtido mediante a transesterificação ou esterificação, em que as lipases que podem ser sintetizadas por meio de fungos lipolíticos são usadas como catalisadores. Assim sendo, a presente pesquisa teve por objetivo realizar a prospecção de enzimas lipolíticas em fungos e utilizar o caldo enzimático bruto para produzir biodiesel. O crescimento dos fungos foi realizado em meio mineral contendo uma única fonte de carbono: o azeite de oliva extra-virgem. O caldo bruto foi analisado por espectrofotometria, utilizando o p-NPB como substrato para quantificar as lipases. O caldo bruto produzido foi utilizado sem nenhuma modificação na presença de diferentes proporções de um planejamento central composto de óleo de soja e metanol para a produção de ésteres metílicos que foram analisados em cromatografia gasosa. Os fungos Paecilomyces formosus (Sakag. & Tada & Samson) I, Paecilomyces formosus II e Aspergillus tubingensis Mos. Produziram, respectivamente, uma atividade lipolítica específica de 9,268 U g-1, 7,032 U g-1 e 6,447 U g-1 e, a partir do caldo bruto usado diretamente para a produção de biodiesel, foi possível alcançar valores de 41,60% de ésteres metílicos, concluindo que as lipases desses fungos são ativas e que quando aplicadas diretamente por meio do caldo bruto é possível realizar a transesterificação obtendo valores promissores até maiores que alguns encontrados na literatura, sendo uma lipase de grande potencial industrial.
