Vildagliptin and sitagliptin are commonly used antidiabetic drugs. Chromosomal aberration (CA), sister chromatid exchange (SCE) and cytokinesis-block micronucleus (CBMN) assays were employed to assess and compare cytotoxic and genotoxic effects of these drugs. Peripheral lymphocytes were exposed to 125 lg/ml, 250 lg/ml and 500 lg/ml of vildagliptin and 250 lg/ml, 500 lg/ml and 1000 lg/ml of sitagliptin for 24 h and 48 h with and without exogenous metabolic activation. At the end of the study, it was determined that these drugs and their metabolites had no genotoxic effects on CA, SCE and CBMN. On the other hand, parallel to the increase in dose, vildagliptin showed weak cytotoxicity on the mitotic index, and depending on its increase in dose; sitagliptin caused potential cytotoxicity and cytostatic effect on the mitotic index, nuclear division index and proliferation index. Due to their cytotoxic and cytostatic potential, these drugs inhibit cell proliferation.
Although Melica uniflora is part of the omnivore and herbivore animal diets, there are not enough studies. The aim of the study is to determine the antimicrobial and antioxidant activity also total phenolic and total flavonoid amount of ethanolic extract of M. uniflora leaves from Yenice Forest, Karabük province, Turkey. In the study, we used the disk diffusion method to evaluate the antimicrobial activity with eight bacteria and two yeast strains; DPPH free radical scavenging activity and ferric reducing antioxidant power analysis were used determine antioxidant activity. Folin-Ciocalteu method for determining the total phenolic amount and AlCl3 method for the total flavonoid content of the extract were tested. Mean diameters of inhibition zones (IZD) of the bacteria were found in the range of 19.02 mm to 26.32 mm. This value was measured as 16.43 mm and 21.38mm for yeasts. The total antioxidant activity value of the extract was calculated at 4.54 mg AAE/g. The IC50 value was calculated 18.798 mg/mL for DPPH free radical scavenging activity. The FRAP value indicated that the reducing power of 1 gram of sample was equivalent to 3.33 µmol of Trolox. The total phenolic content of ethanol extract of M. uniflora leaves was determined as 0.466 mg GAE/g, while the flavonoid content was calculated as 4.44 mgQE/g. According to the obtained results, the analyzed M. uniflora leaves ethanol extracts demonstrated that the biological activity level could be considered significant.

Aynı zamanlarda yayınlanan, "Toksikoloji molekülere kaymaktadır" başlıklı bir makale, toksikolojinin yaşanan çağdaki gelişmelere uyum sağlamakta olduğunu göstermektedir. Bu gelişmelerin ardından toksikologlar, zehirli bir maddenin DNA'nın yapısı, fonksiyonu ve (hedef) gen ekspresyonu üzerindeki etkilerini araştırabilir, toksik cevabı düzenleyen genleri klonlayabilir ve "omik" teknolojilerden (genomik, proteomik, metabolomik, interaktomik), epigenetikten, sistem biyolojisinden, hesaplamalı bilimlerden ve in vitro biyolojiden elde edilen bilgileri harmanlayabilir hale gelmiştir. Günümüzde kullanılmakta olan 100.000'e yakın kimyasal madde mevcuttur ve bunların sadece küçük bir kısmının toksikolojik değerlendirmesi yapılmış durumdadır. Dolayısıyla, bilim adamları bu maddeleri tespit etmek ve etkilerini belirlemek için alternatif yaklaşımlara ihtiyaç duymaktadırlar. Sonuç olarak toksikoloji, kendisini hızla daha bütünsel bir bilimsel disipline dönüştürmektedir. Bu derlemenin amacı, teknolojik gelişmeler ışığında toksikolojik çalışmalara genel bir bakış sunmaktır.
