2012Macroscopic and microscopic analysis of the tongue of the common opossum (Didelphis marsupialis) MICROSCOPY RESEARCH AND TECHNIQUE, HOBOKEN, v. 75, n. 10, pp. 1329-1333 ABSTRACTWe performed a macroscopic and microscopic study of the tongues of common opossums, Didelphis marsupialis, from South America. We studied two males and two females. We collected morphometric data on the tongue with precision calipers. For the light microscopy and scanning electron microscopy analyses, we fixed tissue fragments in 10% formaldehyde and 2.5% glutaraldehyde, respectively. The opossum tongues averaged 5.87 6 0.20 cm in length, 3.27 6 0.15 cm in width at the lingual body, and 3.82 6 0.15 cm in width at the root. The mean thickness of the lingual body was 1.8 6 0.1 cm, and the thickness of the root was 3.82 6 0.15 cm. Sharp filiform papillae were scattered across the entire tongue; conical filiform papillae occurred on the lingual body and tongue tip; fungiform papillae were scattered among the filiform papillae on the lingual body and tongue tip; and there were three vallate papillae at the root of the tongue. We found two strands of papillary projections in the tongue root. Despite the low variability observed in the lingual papillae, the morphological data obtained in this study may be related to the opossum's diverse food habits and the extensive geographic distribution of the species throughout America. The yolk sac is an extra-embryonic membrane that plays an important role in early embryonic survival. It is the production site for blood cells during embryonic mammalian development and is a likely source of stem cells. The aim of this study was to identify and characterize the putative haematopoietic cells from the yolk sac of bovine embryos at different stages of gestation. The yolk sac regresses according to gestational age and embryos are characterized into groups (I-V) according to the crown-rump measurement. Groups I-III survived in culture longer and exhibited the formation of cell clusters, whereas groups IV and V could not be maintained in culture for an extended period of time. Flow-cytometry analysis revealed that groups I-III had similar characteristics, including high expression levels of the haematopoietic markers CD34, CD90 and CD117. In groups IV and V, decreases were observed in the expression levels of CD117 and CD34. Cells were found to be capable of survival post-cryopreservation and exhibited varying abilities to form colonies in a methylcellulose matrix, depending on gestational age. Cytological analysis revealed the presence of blood cells (lymphocytes and monocytes). Quantitative PCR analysis demonstrated the presence of the haematopoietic progenitor genes GATA3 and LMO2, but not RUNX1. Thus, we have successfully isolated and characterized haematopoietic cells from the bovine embryo yolk sac at varying gestational ages. O quati (Nasua nasua, Linnaeus, 1766), de hábito diurno, e o mão-pelada (Procyon cancrivorus, G. Cuvier, 1798), de hábito crepuscular e noturno, são espécies de mamíferos terrestres, pertencentes à família dos procionídeos, que são amplamente encontrados no território brasileiro. Para este trabalho foram utilizados três quatis e dois mãos-peladas adultos provenientes do Criatório Científico-Cecrimpas do Centro Universitário da Fundação de Ensino Octávio Bastos (Ibama 02027.003731/04-76) dos quais foram obtidas amostras de tecido das glândulas salivares parótidas coradas em H.E. para microscopia de luz. Constatou-se que a glândula parótida dos quatis e mãos-peladas possui posicionamento semelhante à dos demais carnívoros domésticos, com seus ductos se abrindo no vestíbulo oral, próximo à altura do quarto dente pré-molar superior, e que histologicamente as glândulas parótidas dos quatis e mão-peladas são constituídas por ácinos serosos, assemelhando-se à maioria dos animais domésticos, homens e roedores e diferindo dos resultados encontrados nos cães jovens, cordeiros e outros carnívoros como o furão que possuem ácinos mistos, e de alguns carnívoros, que apresentam ácinos serosos e mucosos. Conclui-se que o fato da constituição serosa dos ácinos das glândulas parótidas dos quatis e mão-peladas se assemelhar pode ser devido ao hábito alimentar onívoro compartilhado pelas duas espécies.
Gracilinanus microtarsus is one of the smallest marsupials on earth. Since it spreads seeds, it has great ecological relevance. However, its reproduction data, especially those related to the anatomy of its reproduction apparatus, are scarce in the literature. Current analysis describes the male genital organs of six adult specimens of G. microtarsus. Macroscopic studies were undertaken on dissected organs, whereas histological studies were performed by inclusion technique in paraffin and by hematoxylin and eosin, and Masson trichrome staining. The male genital organs and accessory glands of G. microtarsus consist of two testicles within a pendular scrotum, placed cranially to the penis, featuring a histology consisting of seminiferous tubules with spermatogenic cells, spermatozoa and Sertoli cells, and a peritubular region with Leydig cells; testicles closely associated with epididymis with head, body and tail, with histological differences between the different regions; deferent ducts; funiculus spermaticus; accessory glands composed of prostate, divided into three distinct segments and bulbourethral glands; pelvic and penile urethra and penis with bifid glans. Results show that the male genital organs and accessory glands of G. Microtarsus are anatomically similar to that found in Didelphis sp. and other marsupials groups, with slight details such as the site of each organ.Keywords: Didelphimorphia. Marsupials. Possui grande potencial dispersor de sementes e por esta razão apresenta grande importância ecológica. No entanto, dados em relação à reprodução desses animais são escassos na literatura, principalmente, aqueles relacionados à anatomia dos órgãos do aparelho reprodutor. Por essa razão, objetivamos com este estudo realizar uma descrição dos órgãos genitais masculinos em seis exemplares de G. microtarsus adultos. Para tanto, realizamos estudos macroscópicos por dissecação dos órgãos e estudos histológicos por técnica de inclusão em parafina e coloração em hematoxilina e eosina, e tricromo de Masson. Encontramos, que os órgãos genitais masculinos e glândulas acessórias em G. microtarsus são representados por dois testículos localizados dentro de um escroto pendular, posicionados cranialmente ao pênis, cuja histologia revelou túbulos seminíferos com espermatogônias, espermatozoides e células de Sertoli e região peritubular com células de Leydig; testículos em íntima associação com os epidídimos, divididos em cabeça, corpo e cauda, com diferenças histológicas entre as diferentes regiões; ductos deferentes; funículo espermático; glândulas anexas compostas pelas glândulas prostáticas divididas em três distintos segmentos e glândulas bulbouretrais; uretra pélvica e peniana e um pênis com glande bífida. Resumo: A formalização é a principal forma de conservação de corpos e peças anatômicas, devido principalmente ao seu baixo custo. A técnica baseia-se na utilização do formaldeído (5-20%) como fixador e conservador. Entretanto existem vários fatores negativos à sua utilização, como odor desagradável, escurecimento, aumento de peso e rigidez das peças e sérios problemas ambientais quando descartada de forma incorreta, além de ser um produto classificado pela Agência Internacional de Pesquisas em Câncer como altamente cancerígeno. Existem várias opções para substituir o formaldeído, a glicerinação é uma delas, cujo será tratada neste trabalho. A glicerina atua como antifúngico e bactericida, além de ter muitas vantagens em relação ao formol, tratando-se de odor, textura e coloração, além de não ser prejudicial a saúde. Porém o seu custo ainda é elevado e isso explica sua pouca utilização em laboratórios de anatomia. Este trabalho tem como objetivo mostrar as vantagens do uso da glicerina em relação ao formol e propor uma possível substituição deste produto cancerígeno para preservar a saúde dos alunos, funcionários e professores dentro do laboratório, além de permitir um ambiente mais agradável para o aprendizado.
