Cocoa fermentation is the first step in the post-harvest processing chain of cocoa and is important for the removal of the cocoa pulp surrounding the beans and the development of flavor and color precursors. In the present study, metagenomic and metatranscriptomic sequencing were applied to Costa Rican cocoa fermentation processes to unravel the microbial diversity and assess the function and transcription of their genes, thereby increasing the knowledge of this spontaneous fermentation process. Among 97 genera found in these fermentation processes, the major ones were Acetobacter, Komagataeibacter, Limosilactobacillus, Liquorilactobacillus, Lactiplantibacillus, Leuconostoc, Paucilactobacillus, Hanseniaspora, and Saccharomyces. The most prominent species were Limosilactobacillus fermentum, Liquorilactobacillus cacaonum, and Lactiplantibacillus plantarum among the LAB, Acetobacter pasteurianus and Acetobacter ghanensis among the AAB, and Hanseniaspora opuntiae and Saccharomyces cerevisiae among the yeasts. Consumption of glucose, fructose, and citric acid, and the production of ethanol, lactic acid, acetic acid, and mannitol were linked to the major species through metagenomic binning and the application of metatranscriptomic sequencing. By using this approach, it was also found that Lacp. plantarum consumed mannitol and oxidized lactic acid, that A. pasteurianus degraded oxalate, and that species such as Cellvibrio sp., Pectobacterium spp., and Paucilactobacillus vaccinostercus could contribute to pectin degradation. The data generated and results presented in this study could enhance the ability to select and develop appropriate starter cultures to steer the cocoa fermentation process toward a desired course.
Introducción. Las levaduras, las bacterias ácido lácticas y ácido acéticas, involucradas en la fermentación de cacao, son las responsables del consumo de los compuestos en la pulpa para desarrollar los precursores de sabor dentro del grano de cacao. Sin embargo, la fermentación es difícil de controlar por la complejidad de este proceso y las múltiples variables que la afectan, entre ellas la genética del cultivo. Objetivo. Analizar las transformaciones bioquímicas de diferentes grupos genéticos del cacao (Theobroma cacao L.) durante una micro fermentación controlada. Materiales y métodos. Se estableció una metodología de micro fermentación, con un cultivo iniciador de H. opuntiae y Acetobacter spp. en 4 grupos genéticos, obtenidos de clones comerciales del CATIE e híbridos de fincas de Upala. A lo largo de la fermentación se analizaron glucosa, fructosa, etanol, ácido láctico y acético en pulpa y metilxantinas (teobromina y cafeína) y (-)-epicatequina en grano. Resultados. No se encontraron diferencias significativas en el contenido y comportamiento de azúcares, ácidos orgánicos o etanol en pulpa a lo largo de la fermentación, pero sí en el tiempo de fermentación. Se observó una disminución de metilxantinas y epicatequinas, debido a reacciones enzimáticas y no enzimáticas. El contenido promedio de epicatequina resultó ser el compuesto distintivo entre los grupos genéticos del CATIE y cafeína en los cacaos comerciales. Conclusión. La metodología empleada posee un efecto regulador en el comportamiento de algunos metabolitos estudiados en pulpa de diferentes genéticas a lo largo de la fermentación, así como epicatequina y metilxantinas en grano.
Debido a los cambios en patrones de consumo orientados a reconocer atributos de productos locales y diferenciados, el presente estudio tuvo como objetivo evaluar variedades de frijol costarricense con la finalidad de identificar sus propiedades que permitan una mejor valoración y diferenciación dentro del mercado nacional. Se realizó un análisis proximal, fenólico, físico y de calidad e impurezas. Los análisis fueron realizados en el año 2018 en laboratorios de la Universidad Nacional, Costa Rica y el Laboratorio de Calidad e Inocuidad del Consejo Nacional de Producción (CNP). Entre los resultados obtenidos sobresalió la variedad Guaimí (proveniente de Upala) la cual presentó menor dureza, por tanto, podría necesitar menor tiempo de cocción para alcanzar la textura óptima, así como conservar mejor valor textural como sus propiedades funcionales. De forma general, se obtuvieron valores relevantes de proteína en rangos de 16 a 27 %, de fibra de 3 a 7 %, de extracto etéreo de 3 a 7 % y de cenizas de 4 a 6 %. Los resultados del estudio podrían ser un punto de partida para definir la funcionalidad de cada variedad de frijol de con sus propiedades físicas y químicas.
