ABSTRACTQueima da vagem e podridão de raízes da soja causadas por linhagens de Fusarium graminearum e produção de micotoxinas Amostras de sementes de soja (Glycine max) produzidas no Sul do Brasil estavam infetadas com Fusarium graminearum. Para determinar se membros desse complexo de espécies eram patogênicos à soja, seis isolados do fungo, originados de sementes soja, foram adicionados ao solo numa densidade de 10 3 macroconídios/ml ou vagens foram inoculadas individualmente com 10 4 macroconídios/ml. Plântulas crescidas em solo infestado desenvolveram pequenas lesões necróticas na coroa e na parte superior das raízes. Vagens inoculadas com conídios desenvolveram lesões necróticas marrom-escuro grandes (> 1 cm). Vagens jovens inoculadas com o fungo secaram e caíram da planta. Isolados de F. graminearum recuperados das lesões das coroas, raízes e vagens das plantas de soja foram identificadas como pertencentes aos grupos de compatibilidade 1, 2 ou 8, através da análise da seqüência de DNA do gene EF1-alfa, quando comparados com as linhagens conhecidas. Dois isolados de F. graminearum dos Estados Unidos, pertencentes ao grupo de compatibilidade 7, causaram sintomas semelhantes em soja. Testes das micotoxinas produzidas em soja e trigo (Triticum aestivum) indicaram que a maioria dos isolados Brasileiros produz Nivalenol, como a principal toxina dos tricotecenos, ao invés de Deoxinivalenol. Em adição, os isolados dos grupos 2 e 8 produziram uma nova toxina, o 3,AcetilNivalenol. RESUMOSurveys of soybean (Glycine max) seed grown in South Brazil revealed infection with Fusarium graminearum. To determine if members of this complex were pathogenic to soybean, six strains derived from soybean were added to soil at a rate of 10 3 macroconidia/ ml or individual pods were inoculated with 10 4 macroconidia/ml. Seedlings grown in infested soil developed small necrotic lesions in the crown and upper roots. Pods inoculated with conidia developed large (>1 cm), dark brown, necrotic lesions. Younger pods inoculated with the fungus blighted and dropped from the plant. Strains of the F. graminearum complex recovered from lesions on the crown, roots and pods of soybean plants were identified as lineage 1, 2 or 8 by obtaining the DNA sequence from the EF1-alpha gene and comparing it to strains of the known lineage. Two strains of F. graminearum lineage 7 from the U.S. caused similar symptoms of the disease on soybean. Mycotoxin tests on soybean and wheat (Triticum aestivum) indicate that most Brazilian strains produce nivalenol as the major trichothecene mycotoxin rather than deoxynivalenol. In addition, strains from lineages 2 and 8 produce the novel trichothecene, 3-acetylnivalenol.
A complexidade da população de Pyrenophora chaetomioides, principal agente causal da mancha da folha e do grão de aveia (Avena sativa) no Sul do Brasil, é pouco conhecida. Deste modo, estudos envolvendo a variabilidade da população do patógeno embasarão o desenvolvimento de variedades resistentes. Para a realização deste trabalho, foram selecionados oito isolados de P. chaetomioides a partir de sementes de aveia dos três estados do sul do Brasil. Para testar a virulência, os isolados foram inoculados em seis variedades de aveia, avaliando-se a severidade e o tipo de lesão. Todas as variedades de aveia testadas foram suscetíveis aos isolados, embora variações na intensidade de doença tenham sido observadas. Os isolados foram avaliados quanto às suas características amilolíticas, proteolíticas e lipolíticas, utilizando-se meios sólidos para análise destes complexos enzimáticos. O estudo da caracterização enzimática dos isolados revelou a associação de uma alta atividade enzimática com os isolados mais virulentos. Já a análise dos padrões isoenzimáticos de alfa e beta esterases mostrou alta variabilidade entre os isolados com sete perfis distintos identificados mas sem relação com a virulência em plântulas de aveia.
Manchas nos grãos de aveia (Avena sativa) é limitante à sua comercialização por tornar o produto escuro e não permitir seu uso pela indústria alimentícia. A localização do micélio de Pyrenophora avenae nos grãos de aveia e sua atividade enzimática podem esclarecer a causa das manchas. O objetivo deste trabalho foi determinar a localização de P. avenae, na cariopse de aveia, avaliar a sua atividade enzimática e seu efeito sobre proteínas e lipídios dos grãos de aveia. A localização do micélio nos tecidos da cariopse foi determinada após hidratação e cortes da mesma, seguido da análise dos tecidos sob lupa e microscópio. Para avaliação da atividade enzimática foram utilizados 18 isolados de P. avenae obtidos das principais regiões produtoras de aveia do Brasil, avaliando-os quanto às suas atividades amilolítica, proteolítica e lipolítica, sendo realizada por plaqueamento das estruturas vegetativas em meio sólido específico para as enzimas testadas. As determinações do percentual de proteínas e lipídios foram obtidas pelos métodos de Kjeldahl e Bligh & Dyer, respectivamente. O micélio de P. avenae é a principal causa da mancha nos grãos de aveia, localizando-se nos três tecidos do pericarpo. O fitopatógeno apresenta boa atividade enzimática para lipase e protease porém insignificante para a amilase. Os grãos de aveia manchados e sadios não diferiram nos teores de proteínas e de lipídios. Esses teores foram mais elevados nos tecidos superficiais do pericarpo e aleurona independente da presença ou não de manchas, justificando o crescimento superficial de P. avenae sobre os grãos de aveia.
RESUMOA mancha foliar e do grão da aveia (Avena sativa), denominada "helmintosporiose da aveia", causada pelo fungo Pyrenophora chaetomioides é uma doença de importância para o Brasil. Com o objetivo de identificar em que fase do desenvolvimento as cariopses de aveia branca apresentam-se mais suscetíveis à infecção por P. chaetomioides, foram conduzidos três experimentos de campo. Panículas de plantas de aveia foram expostas ao inóculo por períodos determinados de tempo desde a sua emergência. Através da utilização de sacos confeccionados com papel celofane, foram aplicados tratamentos que consistiam na proteção ou na exposição das panículas durante o desenvolvimento das cariopses. No final do ciclo, as panículas foram colhidas individualmente e a porcentagem de cariopses infetadas foi determinada através de plaqueamento em meio de cultura. As cariopses em formação, expostas ao inóculo durante os estágios de grão leitoso e massa mole, apresentaram as maiores incidências do patógeno. A temperatura e o nível de precipitação não influenciaram a porcentagem de cariopses infetadas pelo patógeno, no entanto, temperaturas altas, precipitação elevada e a ocorrência de outras moléstias, anteciparam a senescência das folhas basais, favorecendo a produção de inóculo pelo patógeno.Palavras-chave adicionais: Avena sativa, epidemiologia, helmintosporiose. ABSTRACT Establishment of Pyricularia chaetomioides in white barley caryopsis under field conditionsOat (Avena sativa) is one of the most important crops in South Brazil. As acreage increases, particularly in the no-till system, some diseases became more significant. In order to identify which growth stage of the kernel is more susceptible to the establishment of the fungus Pyrenophora chaetomioides, three experiments were conducted under field conditions. Oat panicles were exposed to natural inoculum during specific periods of time from their extrusion. To protect the panicles from the inoculum, cellophane bags were used to cover them. At the end of the cycle the panicles were individually harvested and the percentage of infected kernels was calculated after plating them in an artificial medium. The kernels exposed to the inoculum during the milk and dough stages showed the highest percentages of infection. RESUMOA mancha-negra dos grãos de aveia branca, cujo principal agente causal é Pyrenophora chaetomioides, é caracterizada pelo escurecimento de seus tecidos superficiais e pode ocasionar redução na qualidade e valor dos mesmos para a indústria alimentícia. Para melhor entender o processo de infecção e formação de manchas os objetivos deste trabalho foram i) Determinar o período de maior suscetibilidade dos grãos à formação de manchas por P. chaetomioides nos componentes florais da aveia; ii) determinar os diferentes níveis de resistência dos genótipos de aveia OR 2, ER 20877-2, UFRGS ��5004-2, UPF �5204-2, UFRGS 16, UFRGS ��12002-2, UFRGS ���007-5, UFRGS ��801�-�, UPF 16, UFRGS ���022-1, UFRGS ��5058-�, UFRGS 15, UFRGS 17, UFRGS 1� e UFRGS ��7005-2 à mancha-negra e sua correlação quanto à infecção dos grãos; iii) avaliar o efeito do controle químico na expressão da doença durante a fase de formação de grãos. Em laboratório, avaliou-se o percentual de grãos manchados e a incidência de P. chaetomioides nos componentes florais, enquanto que, a campo, avaliaram-se o rendimento e peso do hectolitro dos grãos. Nos estádios de grão leitoso e massa é quando ocorre a maior incidência de P. chaetomioides nos componentes florais. Os genótipos diferiram com relação ao sintoma da mancha-negra e incidência, mas não houve correlação entre estas duas variáveis. O controle químico na floração reduziu apenas 21% da incidência do fungo nas sementes, em relação à testemunha. Apesar disso, não foi suficiente para reduzir a formação das manchas e nem aumentar o rendimento que compensasse o seu custo.Palavras-chave adicionais: controle químico, fungicida, mancha-negra. ABSTRACTInfection process and spot development on kernels of white oats with differentiated levels of resistance against Pyrenophora chaetomioides. The black spots of oat (Avena sativa) grains, caused mainly by Pyrenophora chaetomioides, is characterized by the darkening of their superficial tissues, with reduction in their quality and value for the industry. In order to understand the infection process and formation of kernel spots the objectives of this work were i) to determine the period of kernel's susceptibility during their spot formation by P. chaetomioides on the floral components; ii) to determine the different levels of resistance of 15 oat genotypes to black spot and its correlation with kernel infection; iii) to assess the effect of chemical control on the expression of the disease during the period of kernel formation. In the laboratory, the percentage of spotted grains and the incidence of P. chaetomioides in the floral components was evaluated, whereas in the field, yield and industrial weight were measured.
