We report a kind of gas sensor using ZnO nanowires as the field ionization anode. The sharp tips of nanowires generate very high electric fields at relatively low voltages. The sensors show good sensitivity and selectivity. Moreover, the detection limitation of the field ionization based ZnO nanowire gas sensors is about 5%. More importantly, a sensor with ZnO nanowires as the anode exhibits an impressive performance with respect to stability and anti-oxidation behavior, which are significantly better than those of carbon nanotubes (CNTs) as electrodes. Therefore, the simple, low-cost, sensors described here could be deployed for a variety of applications.
Toll-like receptor-4 (TLR4) plays a critical role in innate and acquired immunity, but its role in radio-resistance is unknown. We used TLR4 knockout (KO,−/−) mice and gut commensal depletion methods, to test the influence of TLR4 and its' in vivo agonist on basal radio-resistance. We found that mice deficient in TLR4 were more susceptible to IR-induced mortality and morbidity. Mortality of TLR4-deficient mice after IR was associated with a severe and persistent bone marrow cell loss. Injection of lipopolysaccharide into normal mice, which is known to activate TLR4 in vivo, induced radio-resistance. Moreover, TLR4 in vivo ligands are required for basal radio-resistance. We found that exposure to radiation leads to significant endotoxemia that also confers endogenous protection from irradiation. The circulating endotoxins appear to originate from the gut, as sterilization of the gut with antibiotics lead to increased mortality from radiation. Further data indicated that Myd88, but not TRIF, may be the critical adaptor in TLR4-induced radio-resistance. Taken together, these data strongly suggest that TLR4 plays a critical role in basal radio-resistance. Our data suggest, it is important not to give antibiotics that may sterilize the gut before the whole body irradiation. Further, these data also suggest that management of gut flora through antibiotic or possibly probiotic therapy may alter the innate response to the total body irradiation.
Certain predisposition factors such as BRCA1/2 and CHEK2 mutations cause familial breast cancers that occur early. In China, breast cancers are diagnosed at relatively younger age, and higher percentage of patients are diagnosed before 40 years, than that in Caucasians. However, the prevalence for BRCA1/2 mutations and reported CHEK2 germline mutations is much lower or absent in Chinese population, arguing for the need to study other novel risk alleles among Chinese breast cancer patients. In this study, we searched for CHEK2 mutations in young, high-risk breast cancer patients in China and detected a missense variant Y390C (1169A > G) in 12 of 150 patients (8.0%) and 2 in 250 healthy controls (0.8%, P = 0.0002). Four of the Y390C carriers have family history of breast and/or ovarian cancer. In patients without family history, Y390C carriers tend to develop breast cancer early, before 35 years of age. The codon change at Y390, a highly conserved residue located in CHEK2's kinase domain, appeared to significantly impair CHEK2 activity. Functional analysis suggested that the CHEK2 Y390C mutation is deleterious as judged by the mutant protein's inability to inactivate CDC25A or to activate p53 after DNA damage. Cells expressing the CHEK2 Y390C variant showed impaired p21 and Puma expression after DNA damage, and the deregulated cell cycle checkpoint and apoptotic response may help conserve mutations and therefore contribute to tumorigeneisis. Taken together, our results not only identified a novel CHEK2 allele that is associated with cancer families and confers increased breast cancer risk, but also showed that this allele significantly impairs CHEK2 function during DNA damage response. Our results provide further insight on how the function of such an important cancer gene may be impaired by existing mutations to facilitate tumorigenesis. It also offers a new subject for breast cancer monitoring, prevention and management.
Abstract. We use the stratospheric/tropospheric chemical transport model MOZART-3 to study the distribution and transport of stratospheric O 3 during the remarkable stratospheric sudden warming event observed in January 2004 in the northern polar region. A comparison between observations by the MIPAS instrument on board the ENVISAT spacecraft and model simulations shows that the evolution of the polar vortex and of planetary waves during the warming event plays an important role in controlling the spatial distribution of stratospheric ozone and the downward ozone flux in the lower stratosphere and upper troposphere (UTLS) region. Compared to the situation during the winter of 2002-2003, lower ozone concentrations were transported from the polar region to mid-latitudes, leading to exceptional large areas of low ozone concentrations outside the polar vortex and "low-ozone pockets" in the middle stratosphere. The unusually long-lasting stratospheric westward winds (easterlies) during the 2003-2004 event greatly restricted the upward propagation of planetary waves, causing the weak transport of ozone-rich air originated from low latitudes to the middle polar stratosphere (30 km). The restricted wave activities led to a reduced extratropical downward ozone flux from the lower stratosphere to the lowermost stratosphere (or from the "overworld" into the "middleworld"), especially over East Asia. Consequently, during wintertime (15 December∼15 February), the total downward ozone transport on 100 hPa surface by the descending branches of Brewer-Dobson cir- Moreover, during the two warming winters, the meridional CTOF caused by the isentropic transport associating with the enhanced wave activity also increased and played an important role in the total extratropical CTOF budget.
We analyze the Sun's shadow observed with the Tibet-III air shower array and find that the shadow's center deviates northward (southward) from the optical solar disc center in the "Away" ("Toward") IMF sector. By comparing with numerical simulations based on the solar magnetic field model, we find that the average IMF strength in the "Away" ("Toward") sector is 1.54 ± 0.21stat ± 0.20syst (1.62±0.15stat ±0.22syst) times larger than the model prediction. These demonstrate that the observed Sun's shadow is a useful tool for the quantitative evaluation of the average solar magnetic field.b Deceased.
Epitaxial ferroelectric YMnO3 (YMO) thin films with high crystalline quality were fabricated on (0 0 0 1) GaN substrates by pulsed laser deposition followed by rapid thermal annealing. X-ray diffraction and transmission electron microscopy measurements reveal that YMO layers are of (0 0 0 1) orientation with an in-plane epitaxial relation (0 0 0 1)YMO‖(0 0 0 1)GaN,
. The orientation of the lattice of YMO was found to be controlled by the annealing conditions. Y2O3 nanocrystallites observed near the interface were possible seeds to stabilize the orientation of the epitaxial YMO thin films. The appropriate annealing treatment could improve the crystalline quality and reduce the interface leakage current.
