RESUMOA cagaiteira é uma espécie nativa do Cerrado com elevado potencial frutífero. Como suas sementes apresentam elevada variabilidade, sendo ainda recalcitrantes e dormentes, a germinação in vitro apresenta-se como uma alternativa para a propagação da espécie, além de permitir a obtenção de explantes juvenis para a micropropagação. Com presente trabalho, objetivou-se avaliar o efeito da escarificação e da luminosidade na germinação in vitro de sementes de cagaiteira. Sementes desprovidas do tegumento e sementes intactas foram inoculadas em meio MS e mantidas na ausência e presença de luz. Sementes desprovidas de tegumento germinadas tanto na presença quanto na ausência de luz apresentaram 86,25% e 88,25% de germinação aos 31 e 71 dias da inoculação, respectivamente. Após 150 dias de inoculação, plântulas provenientes de sementes desprovidas de tegumento, germinadas na presença e ausência de luminosidade, apresentaram 10% e 12% de anomalias, respectivamente. Because it seeds show high variability, it is yet considered recalcitrant and dormant, the germination in vitro appears as an alternative for specie propagation, in addition to obtain juvenile explant for micropropagation. The present research aimed to evaluate the effect of scarification and ligth on germination in vitro of cagaiteira seeds. Seeds without teguments and intact seeds were inoculated in MS medium and kept in abscence and presence of radiation. Seeds without tegument germinated in the presence or abscence of ligth showed 86.25% and 88.25% germination at 31 and 71 days of inoculation, respectively. After 150 days of inoculation, plantlets from seeds without tegument germinated in the presence and abscence of ligth, showed 10% and 12% of abnormalities, respectively.
Por apresentar dormência em suas sementes, a propagação do pequizeiro necessita de estudos para a obtenção de mudas por via assexuada. Este trabalho teve como objetivo contribuir para o estabelecimento de um protocolo de micropropagação de pequizeiro. Para indução de brotações, os explantes foram inoculados em meio WPM com diferentes concentrações de BAP e ANA. Para a indução de raízes, brotações obtidas in vitro foram transferidas para meio WPM suplementado com diferentes concentrações de AIB e na presença ou ausência de carvão ativado. As plantas obtidas foram transferidas para sacos de polietileno contendo o substrato, cobertas com uma garrafa tipo pet para auxiliar no processo de aclimatização. O melhor tratamento para a indução de brotações foi aquele que utilizou 0,05mg L-1 de ANA combinado com 0,75mg L-1 de BAP, obtendo-se uma média de 6 brotações por explante, induzindo maior número de gemas por explante (17,4), o que proporcionou a maior taxa de multiplicação (8,7). A utilização de 3mg L-1 de AIB proporcionou a indução de raízes em 100% dos explantes, gerando um número médio de 12,87 raízes por brotação. As raízes desenvolvidas no meio de cultivo contendo carvão ativado foram maiores (33,16 mm), com maior número de raízes secundárias (19,53) e com maior taxa de sobrevivência na fase de aclimatização.
Some macrophytes species show a high growth potential, colonizing large areas on aquatic environments. Cattail (Typha angustifolia L.) uncontrolled growth causes several problems to human activities and local biodiversity, but this also may lead to competition and further problems for this species itself. Thus, the objective of this study was to investigate anatomical modifications on T. angustifolia plants from different population densities, once it can help to understand its biology. Roots and leaves were collected from natural populations growing under high and low densities. These plant materials were fixed and submitted to usual plant microtechnique procedures. Slides were observed and photographed under light microscopy and images were analyzed in the UTHSCSA-Imagetool software. The experimental design was completely randomized with two treatments and ten replicates, data were submitted to one-way ANOVA and Scott-Knott test at p<0.05. Leaves from low density populations showed higher stomatal density and index. These modifications on stomatal characteristics were more evident on the leaf abaxial surface. Plants from low density populations showed thicker mesophyll and higher proportion of aerenchymal area. Roots from low density populations showed a higher proportion of the vascular cylinder. Whereas, plants from higher density populations showed greater thickness of the endodermis, exodermis, phloem and root cortex. Higher density populations showed a higher proportion of aerenchymal gaps in the root cortex. Therefore, cattail plants from populations growing under high density population show anatomical traits typical of plants under stress, which promotes the development of less functional anatomical modifications to aquatic environments.Keywords: anatomical plasticity, cattail, macrophytes, plant handling, eutrophication, stress. Sewage sludge is the solid residue obtained from urban sewage treatment plants. It is possible to use the sludge in a sustainable way as fertilizer and as soil conditioner due to its high levels of organic matter and nutrients. Besides pathogens and volatile organic compounds, the residue may also contain heavy metals which may accumulate and contaminate crops and the food chain. The aim of this study was evaluates the changes in the fertility of dystrophic Red Latosol and in the availability of heavy metals following application of sewage sludge. It was assessed whether organic matter supplied to the soil as large amounts of sewage sludge would decrease availability of heavy metals in the soil due to of insoluble compounds formation. From this, an experiment was carried out in polyethylene pots using lettuce plant for test. Sewage sludge were applied to the soil in concentrations equivalent to 60, 120 and 180 t ha -1 , and a control without sludge, in four replicates, in a completely randomized design. The results show that sewage sludge led to an increase of organic matter contents, of the cation exchange capacity (CEC) and of nutrients found in the soil. It also improved plant growth up to a concentration of 120 t ha -1 . Availability of heavy metals, however, was reduced in sludge concentrations starting with 120 t ha -1 .Index terms: Biosolids, soil pollution, nutrients, Lactuca sativa. RESUMOLodo de esgoto é o resíduo obtido após o tratamento dos esgotos municipais. Em decorrência dos elevados teores de matéria orgânica e de nutrientes, é possível utilizar o lodo, de forma sustentável, como fertilizante agrícola e condicionador de solos. Todavia, além de patógenos e de compostos orgânicos voláteis, esse resíduo pode conter ainda metais pesados que podem ser acumulados e contaminar as culturas agrícolas e a cadeia trófica. Assim, neste trabalho, objetivou-se avaliar as alterações na fertilidade de um Latossolo Vermelho distrófico e na disponibilidade de metais pesados decorrentes da aplicação de lodo de esgoto. Foi avaliado se a matéria orgânica fornecida ao solo com aplicações elevadas de lodo deverá, em razão da formação de compostos insolúveis, diminuir a disponibilidade dos metais pesados do solo. Para tanto, foi conduzido experimento em vasos de polietileno, com a aplicação do lodo no solo nas concentrações equivalentes a 60, 120 e 180 t ha -1 , e um tratamento controle sem lodo, com quatro repetições, em delineamento inteiramente casualizado, utilizando alface como planta teste. Os resultados indicam que o lodo de esgoto promoveu o aumento dos teores de matéria orgânica, da capacidade de troca catiônica (CTC) e de nutrientes no solo e ainda melhorou o desenvolvimento das plantas até a concentração de 120 t ha -1 . A disponibilidade dos metais pesados, no entanto, foi reduzida nos tratamentos a partir de 120 t ha -1 de lodo.Termos para indexação: Biossólidos, poluição do solo, nutrientes, Lactuca sativa.
The plant micro-propagation in bioreactor systems is regarded as one way to reduce cost by automation and production scheduling. This research was carried out in order to obtain an efficient procedure for clone production of Eucalyptus camaldulensis on different types of bioreactor including continuous and temporary immersion bioreactor. To do so, the apical meristems (1 mm) and the apical meristems with adjacent tissue (2,5 mm) were used as initial explants. These tissues were cultured, for 60 days, in semisolid culture medium supplemented with 1 mg L -1 indole acetic acid (IAA) and 0.32 mg L -1 benzylaminopurine (BA). After 60 days, the meristems with adjacent tissue were transferred to a continuous immersion bioreactor and maintained in dark or light conditions. In order to verify the effect of the explant source on bioreactor multiplication, the explants subcultured from meristems multiplied in semisolid culture medium and the meristems multiplied in continuous immersion bioreactor were tested and maintained in dark conditions. After establishing this parameters, the multiplication experiments were carried out in continuous and temporary immersion and the multiplied explants were then rooted in MS medium supplemented with 0, 2, 4, 8 and 20 mg L -1 indole butyric acid (IBA) and kept in the dark or under controlled lighting conditions. After that, the rooting the plants were acclimatized in mist chamber. The meristem with adjacent tissue favored a greater number of buds/explants. The continuous immersion bioreactor in the dark provided higher shoots number and multiplication rate. The rooting was better on culture medium without auxin and kept in the dark for 15 days or the culture medium supplemented with auxin and maintained under light with 100% plantlet rooting. The Eucalyptus camaldulensis acclimatization was efficient, with high survival rate (76%). It was possible to establish the procedure for bioreactor micro-propagation of Eucalyptus camaldulensis large-scale clones.
RESUMO:O pequizeiro é uma frutífera nativa dos cerrados brasileiros com grande potencial econômico. Neste trabalho, objetivouse avaliar o efeito de substâncias potencialmente estimuladoras da germinação. As sementes foram extraídas dos caroços e colocadas para germinar em rolos de papel embebidos com as seguintes soluções: água destilada (Controle); 2mmol L -1 KNO 3 (Nitrato); 2mmol L -1 Etephon (ET); 1mmol L -1 GA 3 (GA); 1mmol L -1 GA 3 + 2mmol L -1 Etephon (GA + ET). Os resultados de percentagem de germinação e o tempo médio para germinação foram, respectivamente, de 54,0% e 9,3 dias no tratamento GA; 47,3% e 11,0 dias no tratamento GA + ET; 32,0% e 12,2 dias no tratamento Controle; 30,7% e 13,1 dias no tratamento Etephon e 20,1% e 13,1 dias no tratamento Nitrato. No tratamento GA + ET houve queda da taxa de germinação até os nove dias da semeadura, com relação ao tratamento GA, o que indica uma possível inibição da germinação de sementes de pequizeiros pela presença de etileno.Palavras-chave: Caryocaraceae, quebra de dormência, tempo médio para germinação. Resumo -O objetivo deste trabalho foi avaliar o efeito do cultivo de plantas de copo-de-leite (Zantedeschia aethiopica) em sistema hidropônico, no crescimento vegetativo, no teor de nutrientes na parte aérea e nas trocas gasosas. Utilizou-se o delineamento experimental inteiramente casualizado, em arranjo fatorial triplo (2x2x2), com oito tratamentos, três repetições e oito plantas por parcela. Os tratamentos consistiram de duas dimensões de espuma fenólica (2,5x2,5x2,5 e 5,0x5,0x3,8 cm, expressas em altura x comprimento x largura), duas soluções nutritivas e dois perfis hidropônicos (100 e 150 mm). As plantas de copo-de-leite foram cultivadas em sistema hidropônico fluxo laminar de nutriente (NFT) e apresentaram teores de nutrientes na parte aérea, na seguinte sequência decrescente: K>N>Ca>P>S>Mg para macronutrientes e Fe>Mn>Zn>B>Cu para micronutrientes. Plantas de copo-de-leite cultivadas em sistema hidropônico NFT, em espuma fenólica de 5,0x5,0x3,8 cm e em perfil hidropônico de 150 mm, apresentam maior acúmulo de massa de matéria seca na parte aérea e no rizoma, bem como maiores taxas fotossintéticas, independentemente da solução nutritiva.Termos para indexação: Zantedeschia aethiopica, floricultura, fotossíntese, nutrição mineral. Nutrient content and gas exchange in calla lily plants grown in a hydroponic systemAbstract -The objective of this work was to evaluate the effect of growing calla lily plants (Zantedeschia aethiopica) in a hydroponic system on vegetative growth, nutrient content in shoots, and gas exchange. A completely randomized design in a triple factorial arrangement (2x2x2) was used, with eight treatments, three replicates, and eight plants per plot. The treatments consisted of two dimensions of phenolic foam (2.5x2.5x2.5 and 5.0x5.0x3.8 cm, expressed as height x length x width), two nutrient solutions, and two hydroponic profiles (100 and 150 mm). The calla lily plants were grown in a nutrient film technique (NFT) hydroponic system and presented nutrient content in the shoots in the following decreasing order: K>N>Ca>P>S>Mg for macronutrients and Fe>Mn>Zn>B>Cu for micronutrients. Calla lily plants grown in a NFT hydroponic system, in the phenolic foam of 5.0x5.0x3.8-cm dimensions and in the hydroponic profile of 150 mm, show greater dry matter accumulation in shoots and bulbs, as well higher photosynthetic rates, regardless of nutrient solution.
