The genetic connectivity of soft coral is influenced by current and distance between islands. The complexity of islands and geographical region in Indonesia might influence the distribution of soft corals. The information of genetic connectivity can be used to design marine protected areas and to avoid destruction and possible extinction. The objective of the present study was to analyze genetic connectivity of one species of soft coral, Sarcophyton trocheliophorum, in three populations spanning Java, Nusa Tenggara, and Sulawesi's waters, and to describe its implication for marine protected area. The mitochondrial protein-coding gene (750 bp of ND2) was used to analyze genetic population structure and genetic connectivity. Genetic connectivity was found in all populations with Fst value of 0.227 to 0.558, indicating populations had the close genetic relationship. The local and Indonesian currents were expected to distribute the larva to islands as a stepping stone, they moved slowly to spread them self far away. Tanakeke island (Sulawesi population) might be a center connectivity of S. trocheliophorum populations. This island connected with islands in west and east Indonesia, therefore that area need to protect.
ABSTRAKBahan bakar fosil merupakan bahan bakar yang berasal dari pelapukan sisa mahluk hidup. Bahan bakar fosil bersifat tidak terbaharukan, maka pencarian bahan bakar alternatif yang terbarukan perlu dilakukan salah satunya bioetanol. Tetraselmis chuii dapat dijadikan bahan baku pembuatan bioetanol. Tujuan dari penelitian ini adalah untuk mengetahui volume bioetanol yang dihasilkan dari Fermentasi Tetraselmis chuii. Kultur Tetraselmis chuii selama 6 hari. Hidrolisis dilakukan dengan menambahkan H 2 SO 4 0,2 M pada suhu 121 o C dengan tekanan 1 atm selama 30 menit, Fermentasi dengan Saccharomyces cereviseae selama 5 hari. Penelitian ini menghasilkan gula reduksi Tetraselmis chuii dengan kadar gula 4% dan hasil fermentasi Tetraselmis chuii menghasilkan 12 ml etanol dengan konsentrasi 1%. ABSTRACT THE ROLE AND UTILIZATION OF MICROALGAE Tetraselmis chuii AS BIOETHANOL.Fossil fuels are the fuel produced from the weathering of living things. Fossil fuels are non-renewable, therefore the research of renewable energy is needed. Bioethanol could be good solution. Tetraselmis chuii can be used as the raw material for bioethanol. The purpose of this study was to determine the volume of ethanol through fermentation of Tetraselmis chuii. Tetraselmis chuii was cultured for 6 days. The hydrolysis used 0.2 M H 2 SO 4 at 121 0 C and pressure of 1 atm for 30 minutes, and the fermentation used Sacscharomyces cereviseae for 5 days. The results showed that Tetraselmis chuii produced 4% sugar and The fermentation produced 1% bioethanol with volume was 12 ml.
Penelitian dilaksanakan di Pulau Baai Kota Bengkulu pada bulan Oktober – November 2017. Dari penelitian kondisi ekosistem mangrove di Pulau Baai kota Bengkulu, didapatkan 5 spesies dari 4 Family mangrove terdiri dari Family Rhizoporaceae (Rhizophora mucronata dan Bruguiera cylindrica), Family Avicenniaceae (Avicennia marina), Family Lythraceae (Sonneratia alba)dan Family Combretaceae (Lumnitzera littoreae). Indeks Nilai Penting (INP) mangrove menunjukkan peran penting mangrove untuk Pulau Baai kota Bengkulutergolong sedang untuk tingkat pohon berkisar antara 105,42 – 150,11. Nilai indeks keanekaragaman (H’) untuk tingkat pohon, anakan, semai untuk stasiun 1 termasuk dalam kategori rendah dalam seluruh kategori, sedangkan stasiun 2 termsuk dalam kategori rendah dalam seluruh kategori. Untuk stasiun 3 termasuk dalam kategori sedang untuk pohon, sedang untuk anakan dan rendah untuk semai. Nilai indeks dominansi (D) untuk tingkat pohon, anakan, dan semai untuk stasiun 1 berturut-turut dalam kategori tinggi untuk seluruh kategori, sedangkan untuk stasiun 2 yaitu tinggi untuk seluruh kategori dan untuk stasiun 3 yaitu sedang untuk pohon, sedang untuk anakan, dan tinggi untuk semai. Hal ini menunjukan bahwa terdapat jenis yang mendominasi jenis lainnya pada ekosistem mangrove di Pulau Baai kota Bengkulu atau komunitas berada pada kondisi kurang stabil. Hasil pengamatan parameter kualitas air seperti suhu, salinitas dan pH menunjukkan bahwa kondisi lingkungan yang ada di Pulau Baai kota Bengkulu tergolong baik untuk pertumbuhan ekosistem mangrove.
Circulation of synthetic antibacterial chemicals which is used continuously can cause resistance to bacteria. Alternative synthetic antibiotics research then comes up as an urgent need. Recently, the discovery of new antibacterial agents is looking for Actinomycetes. Actinomycetes are one of the active marine bacteria which produce secondary metabolites that could be used as antibacterial. The objective of this study was to evaluate the antibacterial activities of Actinomycetes symbiont with Halimeda macroloba, Gelidiella acerosa, and Turbinaria ornata in Pangandaran beach, Indonesia. A total of 41 Actinomycetes were isolated from 3 species of seaweed. Rep PCR amplification method was used to characterize and identify the relationship between samples. Primer BOX A1R (5’-CTACGGCAAGGCGACGCTGACGCTGACG-3’), 27F (5'-AGTTTGATCMTGGCTCAG-3') and 1492R (5'-TACGGYTACCTTGTTACGACTT-3') was used in amplification 16s DNA. Six isolates from the representative cluster dendogram were selected for secondary metabolites extraction using ethyl acetate solvent, meanwhile agar diffusion method was used as Antibacterial activity assay using E. coli, Proteus sp., and Enterobacter sp. as the bacterial test. The results showed that the activity of secondary metabolite extracts of Actinomycetes symbiont with Halimeda macroloba presented the highest inhibition zone on 3 bacterial tests. In the future, it could be a potential antibacterial agent against animal pathogenic bacteria.
Kapang adalah mikroorganisme yang dapat diisolasi dari beberapa sumber seperti sedimen, air, serasah, rumput laut dan masih banyak lagi. Kapang dapat menghasilkan enzim yang memiliki banyak fungsi dan keuntungan. Tujuan dari penelitian ini adalah untuk mengisolasi kapang dari sedimen, serasah, dan air yang berasal dari sekitar lingkungan Caulerpa sp. Dan mengidentifikasi potensi enzim agarase yang dihasilkan. Sebanyak 41 isolat berhasil diisolasi. 5 isolat memiliki aktivitas enzim yang potensial (A10, A11, A13, SUC 7 dan SEC 8). Isolat A13 adalah isolat terbaik karena memiliki aktivitas agarase tertinggi dan waktu pertumbuhan yang lebih cepat daripada yang lain.
