Preparation of (6R, S)-5,6,7,84etrahydro-~-biopterine, 7,8-dihydro-~-biopterine, L-sepiaptcrine, deoxysepiapterine, (6R, S)-5,6-dihydrodeoxysepiapterine and 2'-deoxybiopterine
SummaryWe describe the preparation of (6 R, S)-5,6,7,8-tetrahydro-~-biopterine (11) by catalytic-(reported in our short communication [2]) and natriumdithionite reduction of L-biopterine (I) and a preparation of 7,8-dihydro-L-biopterine (IV) by treatment of L-biopterine (I) with natriumdithionite. Whereas oxidation of tetrahydrobiopterine I1 in the presence of oxygen leads to L-sepiapterine (III), dihydrobiopterine IV in acetic acid without oxygen gives deoxysepiapterine (VII), 5,6-dihydro-deoxysepiapterine (VI) and 2'-deoxybiopterine (V) by a disproportionution reaction.(6 R)oder (6 S)-5,6,7,8-Tetrahydro-~-biopterin (11) und 7,8-Dihydro-~-biopterin (IV) sind fur die biogenetische Hydroxylierung von Phenylalanin von zentraler Bedeutung [3]. Deshalb wurde vielfach versucht, diese Verbindungen herzustellen. Die Herstellung von I1 wurde von uns kiirzlich [2] beschrieben. Fur die Herstellung von IV wurde rnit wenig Erfolg die Reduktion von L-Biopterin (I) mit Zink/Natronlauge [4], sowie die partielle Riickoxydation von I1 mit Sauerstoff versucht [5]. Spater gelang es jedoch japanischen Autoren, ausgehend von natiirlichem I, durch Reduktion mit Natriumdithionit, kristallines IV zu erhalten [6]. Da synthetisches L-Biopterin (I) heute leicht zuganglich ist [2], konnen wir in der vorliegenden Mitteilung die Herstellung von (6 R, S)-5,6,7,8-Tetrahydro-~-biopterin (11) und 7,8-Dihydro-L-biopterin (IV) beschreiben. Dabei kann I1 aus I einerseits durch katalytische Reduktion, andererseits durch Reduktion mit Natriumdithionit erhalten werden (vgl. Schema und exper. Teil). I) Teil der geplanten Dissertation von B.
