Reproducible and high-frequency transgenic plant regeneration from callus and embryo axes of four different genotypes of chickpea (Cicer arietinum) was achieved after Agrobacterium-mediated transformation. Three different strains of Agrobacterium (EHA105, AGL1 and LBA4404) harboring the binary vector pCAMBIA1301 containing β-glucuronidase (GUS) and hygromycin phosphotransferase (hpt) genes under the control of a CaMV35S promoter were used. The highest number of transgenic plants was obtained from cotyledonary node-derived calli of genotype Pusa-256. A highly efficient rooting was achieved on Murashige and Skoog medium supplemented with indole-3-butyric acid. The stable integration of the gene was confirmed by molecular analyses of the transformed plants. Inheritance of GUS and hpt gene was followed through two generations and they showed the expected 3:1 inheritance.Additional key words: Cicer arietinum, cotyledon-and cotyledonary node-derived calli, β-glucuronidase, hygromycin phosphotransferase.
An efficient callus induction and plant regeneration system has been developed using three local soybean (Glycine max L.) varieties. All the varieties showed good callusing (78.30 -88.80%) from shoot tip (ST) in MS + 3.0 mg/l 2,4-D + 1.0 mg/l BAP. Best callus induction and embryo formation were recorded in T5 for BS-6 as 88.80 and 81.20%, respectively from ST. In case of cotyledonary node (CN) the maximum callus induction (82.40%) and embryo formation (74.20%) were recorded also in T5 for BS-6. Highest frequency (79.40%) of plant regeneration was recorded where ST were used and cultured in MS supplemented with 2.5 mg/l BAP + 1.0 mg/l NAA as well as 76.30% from CN in BS-6. The length of shoot was observed 5.32 and 4.62 cm, respectively from ST and CN for BS-6 with the same medium composition. It was observed that half strength of MS + 2.0 mg/l IBA showed best rooting (9.04). Among the genotypes BS-6 proved to be best explants than ST that exhibited better performance on callus induction and green plant regeneration for all parameters.
A rapid and an efficient protocol for germination of immature seeds of Vanda tessellata, PLBs development and plant regeneration system have been developed. The results indicate that immature capsules performed better in terms of germination (100.0%) and development of PLBs (90.0%). Around 45 - 60 days old PLBs were transferred to half strength of MS supplemented with different concentrations of auxins and cytokinins. The highest weight of PLBs (5.53 g), the best plant regeneration (16.50) and shoot length (3.25 cm) were recorded when the medium was supplemented with BAP (1.0 mg/l) and NAA (1.5 mg/l). The highest number (5.0) and length (3.22 cm) of roots, the maximum number of plantlets (3.75) were obtained in the medium supplemented with 1.5 mg/l BAP and 1.0 mg/l IAA. The maximum numbers of leaves (3.33) were recorded in the medium fortified with 1.5 mg/l BAP + 1.00 mg/l IAA; the highest length of leaves (2.59 cm) was observed in the medium combining with of BAP and IAA (2.0 mg/l). Well shooted - and rooted plants were acclimated in pots that contained two types of compost.
