Using correlated light and electron microscopic preembedding enkephalin immunocytochemistry combined with post-embedding GABA immunogold staining, we found morphological evidence of a direct connection between the enkephalinergic and GABAergic systems in the rat hippocampus. Enkephalin-immunoreactive boutons were found to be presynaptic to GABA-immunoreactive postsynaptic profiles, establishing type 2 symmetrical synapses on GABA-positive cell bodies and dendritic shafts in strata radiatum and lacunosum moleculare of the CA1 region. Thirty-six percent of all studied postsynaptic targets (n = 40) were non-pyramidal, including all somatic (n = 7) and 47% of the dendritic (n = 13) postsynaptic targets. The remaining 64% consisted of pyramidal dendritic shafts and spines. These results support previous physiological experiments suggesting that the opioidergic system takes part in disinhibitory processes in the hippocampal formation.
Zusammenfassung
Vergleichende Prüfungen auf Toxinbildung von Bordetella‐bronchiseptica‐ und Pasteurella‐multocida‐Stämmen aus 2 Ra‐freien und 12 Ra‐positiven Beständen mit unter‐schiedlicher Ra‐Morbiditat ergaben folgende Hinweise:
— Bordetellen‐Stämme mit starker bis mittelgradiger Toxinbildung wurden von allen 12 Ra‐positiven Schweinen isoliert. In den 2 Ra‐freien Beständen wurden ein atoxischer und ein schwach toxinbildender Stamm festgestellt.
— Der Nachweis eines hitzelabilen, dermonekrotischen Toxins von Pasteurellen, das offensichtlich eine gleiche Wirkung auf die Nasenconchen wie das Bordetellen‐Exotoxin besitzt, gelang vorwiegend bei Carter D‐, jedoch auch bei A‐ und nicht typisierbaren Pasteurella‐multocida‐Stämmen. Zum In‐vitro‐Nachweis eignet sich die Prüfung von bakterienfreien Ultraschall‐Extrakten im Meerschweinchenhauttest und im intraperitonealen Mäuseltalitätstest. Außerdem wurde von uns erstmals zum Toxinnachweis von Pasteurellen der Lienotoxtest mit guten, reproduzierbaren Ergebnissen eingesetzt.
— Nur in 50 % der Ra‐positiven Schweine wurden toxische Pasteurellen‐Stämme nachgewiesen, ebenfalls wurden 2 toxische D‐Stämme aus gesunden, sekundären SPF‐Schweinen isoliert. Somit bestand zwischen Ra‐Status und der Toxinbildung der Pasteurel‐len‐Isolate kein unmittelbarer Zusammenhang.
— Es wird geschlußfolgert, daß beide Erregerarten in der Pathogenese der Ra eine Rolle spielen und sich beide Erreger in ihrer Wirkung durch gleiche Pathogenesemechanismen unterstützen, wobei die Faktoren für das Wirksamwerden von toxischen Pasteurellen‐Stämmen noch nicht restlos geklärt sind.
Summary
The significance of Bordetelfa bronchiseptica and Pasteurella multocida in swine atrophic rhinitis
Comparative studies on toxin production of Bordetella bronchiseptica and Pasteurella multocida strains from two AR‐free and twelve AR‐positive herds with varying AR‐morbidity gave the following results:
— Bordetella strains with marked or medium toxin production were isolated from all 12 AR‐positive herds. In the two AR‐free herds an atoxic and a weak toxin‐producing strain were isolated.
— The authors demonstrated a heat labile, dermonecrotic Pasteurella toxin which apparently had the same effect as Bordetella exotoxin and belonged mainly to Cardet D‐, but also to A and to untypable Pasteurella strains. For in vitro demonstration the most suitable technique was demonstration of bacteria‐free ultrasonic extracts in the mouse skin test and the intraperitoneal mouse lethality test. In addition, the authors showed for the first time toxin demonstration of Pasteurella in the Lienotox test which gave well reproducible results.
— Only in 50 % of AR‐positive pigs were toxic Pasteurella strains detected; furthermore, two toxic D‐strains were isolated from healthy secondary SPF pigs. There thus exists no direct relationship between AR‐strains and toxin production of Pasteurella isolates.
