Uma das vertentes da bioquímica está relacionada com o estudo das estruturas tridimensionais das macromoléculas envolvidas na formação celular. Por não serem visíveis a olho nu, exige um alto grau de complexidade para sua compressão. O objetivo do trabalho caracterizou-se na produção de materiais didáticos feita com materiais reutilizáveis, relacionado os conceitos bioquímicos e estruturais das macromoléculas. Inicialmente foi aplicado um questionário aos estudantes do 1° ano do Ensino Médio de um colégio público estadual do Recife. Observou-se que as concepções prévias dos estudantes apresentavam conceitos generalistas e/ou equivocados. Estas concepções foram relevantes para intervenção, realizada de forma expositiva e dialogada, utilizando modelos didáticos, em seguida, o mesmo questionário foi respondido. Os modelos didáticos podem ser utilizados como ferramentas em auxílio ao professor no processo ensino-aprendizagem bem como para acompanhar o progresso dos estudantes.
Inserida nos cursos de saúde, biológicas e agrárias, a bioquímica aborda conceitos de biologia e química para estudar fenômenos nos organismos cuja compreensão prejudica-se pela quantidade de informações e necessidade de abstração do conteúdo. As ferramentas didáticas atuais são insuficientes para facilitar o entendimento, tornando-se necessário o desenvolvimento de atividades lúdicas para visualização das estruturas. O objetivo do trabalho foi elaborar modelos das macromoléculas presentes nos seres vivos como ferramenta didática para o ensino da bioquímica e sua contribuição para formação discente. O trabalho foi desenvolvido com alunos cursando bioquímica em um curso de graduação da Universidade Federal Rural de Pernambuco. Primeiramente, responderam um questionário para investigar conhecimentos prévios e divididos em grupos para confecção da macromolécula a ser apresentada ao final da disciplina. Após a culminância da atividade aplicou-se questionários sobre conteúdos bioquímicos e da prática realizada. Analisou-se as respostas qualitativamente. Inicialmente os discentes apresentaram desinteresse e poucos conhecimentos prévios. Adiante, perceberam correlação entre a disciplina e o meio, bem como a presença de conceitos anteriormente ausentes ou insipientes. A atividade foi satisfatória ao aprendizado, e aplicável na prática docente. O material confeccionado auxiliou no processo ensino-aprendizagem e ampliou metodologias de ensino existentes para abordagem do conteúdo da bioquímica.
Thrombosis is characterized by the pathological formation of fibrin clots within a blood vessel, leading to the obstruction of blood flow. Fibrinolytic enzymes from microorganisms have exhibited promising effects to dissolve clots in a more efficient and safe way. Then, the aim of this study was to evaluate the biomass and fibrinolytic enzyme production of Tetradesmus obliquus under autotrophic and mixotrophic conditions using different concentration of corn steep liquor (CSL). Different extraction and precipitating methods were tested, and the enzyme was purified by ion exchange chromatography. More advantageous culture condition was mixotrophic using 0.25% CSL, showing the highest values of biomass productivity (Px = 169.3 ± 44.36 mg∙L-1day-1) and specific growth rate (µmax = 0.17 ± 0.00 day-1), and significant fibrinolytic production (391.34 ± 40.03 U∙mg-1). Moreover, fibrinolytic activity was higher when extracted by homogenization and precipitated using acetone, which exhibited clear zone of fibrin degradation in the fibrinolytic plate assay. Additionally, the purified enzyme showed specific activity of 1176.90 ± 140.37 U∙mg-1 and molecular weight around 97 kDa. Finally, the enzyme has higher enzymatic activity than various fibrinolytic enzymes, and the obtained enzyme has potential to be developed as a therapeutic agent in thrombosis treatment. Additional studies are need to investigate the biochemical properties and biological profile of this enzyme.
A bioquímica é complexa, extensa e de caráter interdisciplinar. O ensino tradicional gera uma disciplina decorativa e desinteressante. O jogo lúdico caracteriza-se como ferramenta para auxiliar na construção do conhecimento e desenvolvimento cognitivo e social. O objetivo foi elaborar e avaliar um jogo lúdico sobre enzimas visando uma aprendizagem colaborativa e divertida. A atividade tem como público alvo discentes de nível superior. O jogo de tabuleiro é composto por materiais simples, de baixo custo e fácil acesso. Questionário constituído por cinco questões objetivas foi utilizado para avaliar o jogo. Observou-se entusiasmo, motivação e troca de conhecimentos entre discentes durante o jogo. Após análise dos questionários, observou-se que os discentes aprovaram o jogo como ferramenta complementar no estudo da estrutura química, organização tridimensional e função de enzimas, visto que a maioria dos estudantes entrevistados concordaram que o jogo melhorou a aprendizagem sobre enzimas, além de incentivar os estudos sobre o tema. Esses resultados caracterizam o jogo como uma importante estratégia didática de baixo custo facilitando o acesso e tornando viável seu uso em diferentes níveis de ensino.
