Significant advances in the synthesis of polypeptides by N-carboxyanhydride (NCA) polymerisation over the last decade have enabled the design of advanced polypeptide architectures such as star-shaped polypeptides. These materials combine the functionality offered by amino acids with the flexibility of creating stable nanoparticles with adjustable cargo space for therapeutic delivery. This review highlights recent advances in the synthesis of star polypeptides by NCA polymerisation followed by a critical review of the applications of this class of polymer in the delivery of therapeutic agents. This includes examples of traditional small-molecule drugs as well as the emerging class of biologics such as genetic therapeutics (gene delivery).
Bionanomaterials have received increasing attention over the last decade, owing to their potential to advance medical science by providing novel solutions for disease diagnostics and treatment. [1] However, bionanomaterial engineering imposes considerable challenges, as significant design criteria and properties, which are dictated by the specific application, must be met. MRI-trackable magnetic nanoparticles (MNPs) and their assemblies, which are capable of targeting specific cell types, such as cancer cells, for image-guided treatment are increasingly being investigated. [2] The typical design criteria for MNPs are biomedical in nature, including cellular selectivity (biorecognition), extended blood circulation times and biocompatibility, there are also practical considerations such as processability and scalability. Additional MRI-specific criteria include colloidal stability and high image contrast. The efficacy of an MNP suspension for generating contrast by enhancing, or suppressing, a local MRI signal is quantified by the spin-lattice (r 1 ) and spin-spin (r 2 ) relaxivities, respectively. These are the water relaxation enhancement per millimolar concentration of iron. For T 1weighting applications (localized signal enhancement), r 1 must be high, and r 2 and the r 2 /r 1 ratio must be low. Particle aggregation increases the r 2 value, so T 1 -weighting can only be achieved by ensuring full MNP dispersion and long-term colloidal stability.Monosaccharides offer colloidal stability through charge and steric interactions along with the possibility of cellular targeting, and so have been used to stabilize MNPs. Recently, MNP clusters have been prepared by encapsulation of alkylstabilized MNPs using poly(maleic anhydride-alt-1-octadecene) followed by EDC coupling of aminated sugars. [3] Detailed analysis of the effect of coating on cellular uptake showed that high grafting density is critical for avoiding nonspecific cellular interaction. However, the encapsulation approach resulted in extensive aggregation, as is usually the case, thus limiting the possibility of use in a T 1 -weighting application. Aggregation was exploited in a recent report where clickable monosaccharides were used to stabilize MNP clusters using complex peptide linkers, [3] which allowed differentiation of cancer cells by quantitative profiling of carbohydrate binding through changes in T 2 (local signal suppression). In a prominent recent example, a range of phosphonate-functionalized sugars were used to fully disperse MNPs, and the effect of MNP size on the T 1 -weighting and hyperthermic properties in vitro were investigated. [4] However, only one sugar per linker was possible, thus limiting the total sugar density.Hence, there is a need for platform technologies that maximize the surface density of functional monosaccharides while allowing full MNP dispersion. Multifunctional polymers have the potential to address the first issue; there have been two recent reports of a "grafting-onto" approach for MNP modification with glycopolymers....
Το βασικό ερευνητικό ερώτημα της διατριβής είναι το εξής: πώς και γιατί ο άνθρωπος άρχισε να εκφράζεται μουσικά. Η διερεύνησή του είναι διεπιστημονική καθώς συλλέγονται δεδομένα από διάφορα επιστημονικά πεδία. Κοινό σημείο αναφοράς των σύγχρονων προσεγγίσεων είναι η εξέλιξη και οι μηχανισμοί της προσαρμογής. Στο Α΄ μέρος που τιτλοφορείται ''Μουσική και Προσαρμογή'' εξετάζονται οι κυριότερες θεωρίες που εκλαμβάνουν τη μουσική έκφραση ως αποτέλεσμα των διαδικασιών της εξέλιξης ενώ παράλληλα επισημαίνονται οι αδυναμίες τους. Ο προσανατολισμός αυτός όμως έρχεται σε ευθεία αντίθεση με τη συνήθη προβεβλημένη άποψη ότι η μουσική και η τέχνη γενικά δεν έχουν καμία πρακτική χρησιμότητα ή επιβιωτική αξία. Σχετικά με τις προτεινόμενες θεωρίες υπογραμμίζεται η επικοινωνιακή τους διάσταση και αναδεικνύεται ο υπόρρητος χαρακτήρας των επικοινωνιακών τους μηνυμάτων. Αντιθέτως στο Β΄ μέρος ''Μουσική και Γλώσσα'', λαμβάνοντας υπόψη τις ομοιότητες ανάμεσα στα δύο συστήματα, της μουσικής και της γλώσσας, προβάλλεται σε διάφορες εκδοχές μέσα στο ίδιο παραδειγματικό πλαίσιο η υπόθεση μιας προδρομικής μορφής πρωτογλώσσας με μουσικά χαρακτηριστικά, κατά τη χρήση της οποίας το επικοινωνιακό περιεχόμενο είναι ρητό και καλύπτει πολύ περισσότερες επικοινωνιακές ανάγκες. Ένα πρόσφορο πεδίο έρευνας όπου έχουμε τη δυνατότητα να αναζητήσουμε απηχήσεις των θεωριών που αναλύονται στα δύο πρώτα μέρη της διατριβής είναι ο αρχαίος ελληνικός πολιτισμός, ένας κατεξοχήν μουσικός πολιτισμός. Πολλά συμπεράσματα βγάζουμε αναλύοντας τη μουσική φύση της αρχαίας ελληνικής γλώσσας, όπως και τη γλώσσα του μύθου. Το κομβικό σημείο όμως κατά το οποίο η μουσική ως σύστημα αυτονομείται από το σύστημα της γλώσσας εντοπίζεται στην περίοδο όπου εκπληρώνονται οι γνωσιακές προϋποθέσεις για την εμφάνιση της μουσικής. Επιπλέον, υποστηρίζεται ότι η γέννηση του αυτόνομου συστήματος της μουσικής συντελείται κάτω από ιδιαίτερες συνειδησιακές καταστάσεις, γνωστές στη διεθνή βιβλιογραφία ως διαφοροποιημένες καταστάσεις συνείδησης (altered states of consciousness) και κυρίως κατά τη διάρκεια των τελετουργιών που συνηθίζεται να αποκαλούνται σαμανιστικές υπό την ευρεία έννοια. Ο αρχαίος ελληνικός πολιτισμός μας δίνει πληθώρα παραδειγμάτων όπου ανιχνεύεται η στενή σχέση των διαφοροποιημένων καταστάσεων συνείδησης και του σαμανισμού με τη μουσική τα οποία εξετάζονται διεξοδικά στο Γ΄ μέρος. Υπό αυτό το πρίσμα μπορούμε να προσεγγίσουμε και να ερμηνεύσουμε κι άλλα επίμαχα ζητήματα όπως τη σχέση μεταξύ ορφικής και διονυσιακής τελετουργίας, ακόμη και τη διαμόρφωση των πνευματικών προϋποθέσεων για τη γέννηση της φιλοσοφίας στην Ελλάδα. Επιγραμματικά, η διατριβή στηρίζεται στην υπόθεση ότι στις απαρχές η μουσική υπηρετεί αποκλειστικά επικοινωνιακούς σκοπούς, αρχικά σε προγλωσσικό και έπειτα σε πρωτογλωσσικό στάδιο. Ως πλήρως αυτόνομη δραστηριότητα αναπτύσσεται σε τελικό στάδιο κατά το οποίο αποκτά νέο πνευματικό περιεχόμενο. Tα πρώτα ωστόσο χαρακτηριστικά της εξακολουθούν να κατέχουν σημαντικό ρόλο στη λειτουργία της.
