4RESUMO -A germinação de sementes é dependente de fatores abióticos, sendo a temperatura um dos principais, cuja influência, em condições extremas, causa danos às sementes. Este trabalho teve por objetivo investigar o efeito das diferentes temperaturas durante a germinação de Dalbergia nigra e suas implicações na fisiologia das sementes. Avaliaram-se o percentual de germinação, o índice de velocidade de germinação (IVG) e a integridade de membranas celulares pelo teste de condutividade elétrica de sementes em diferentes tempos de exposição às temperaturas de 5, 15, 25 (controle), 35 e 45 °C. A temperatura de 25 ºC correspondeu à temperatura ideal de germinação. Em temperaturas de 5 e 45 ºC, a germinação foi nula. Houve redução da germinação de sementes de D. nigra com o aumento do tempo de exposição das sementes às temperaturas de 5, 15, 35 e 45 °C. Diferentemente das demais temperaturas, a semipermeabilidade da membrana não é recuperada nas temperaturas de 5 e 45 ºC. A condutividade elétrica é uma técnica eficiente para avaliar a qualidade fisiológica das sementes em diferentes temperaturas.Palavras-chave: Fisiologia; Jacarandá-da-bahia; Membranas celulares. -Seed dormancy is a phenomenon that affects the distribution of plant species in time. However, it may slow germination and consequently hinder seedling production in nurseries. Many seeds of forest species of the Fabaceae family, such as Libidibia ferrea (Brazilian ironwood), have physical dormancy caused by impermeability of the seed coat to water. Therefore, the aim of this study was to evaluate the physiological, physical, and morpho-anatomical characteristics of L. ferrea seeds before and after application of different treatments for overcoming dormancy. We evaluated the imbibition curves, germination percentage, germination speed index (GSI), the force required to puncture the micropylar region, and morphoanatomical changes through images obtained from a scanning electron microscope (SEM) before and after application of treatments for overcoming dormancy. L. ferrea seeds immersed in sulfuric acid show a triphasic pattern of imbibition. The required force to puncture the micropylar region of L. ferrea seeds is less for both the treatment with boiling water and for treatments with sulfuric acid. In addition, the required force to puncture the micropylar region decreases during imbibition of the seeds after application of sulfuric acid. The time of application of sulfuric acid influences the thickness of the material removed from the macrosclereid layer of the seed coat.Index terms: Libidibia ferrea ((Mart. ex Tul.) L.P. Alterações fisiológicas, físicas e morfo-anatômicas em sementes deLibidibia ferrea após superação de dormência RESUMO -A dormência de sementes é um fenômeno que influencia a distribuição das espécies de vegetais no tempo. Porém, pode retardar a germinação, e consequentemente, dificultar a produção de mudas em viveiros. Muitas sementes de espécies florestais da família Fabaceae, como Libidibia ferrea (pau-ferro), apresentam dormência física causada pela impermeabilidade do tegumento à água. Portanto, o objetivo neste trabalho foi avaliar as características fisiológicas, físicas e morfo-anatômicas das sementes de L. ferrea antes e após aplicação de diferentes tratamentos para superação de dormência. Avaliaram-se as curvas de embebição, a porcentagem de germinação, o índice de velocidade de germinação, a força requerida para ruptura da região micropilar e as alterações morfo-anatômicas por meio de imagens obtidas de microscópio eletrônico de varredura antes e após a aplicação de tratamentos para superação de dormência. Sementes de L. ferrea imersas em ácido sulfúrico apresentam padrão trifásico de embebição. A força de ruptura da região micropilar de sementes de L. ferrea é menor tanto para o tratamento com água fervente quanto para os tratamentos com ácido sulfúrico. Além disso, a força necessária para ruptura da região micropilar diminui durante a embebição das sementes após aplicação de ácido sulfúrico. O tempo de aplicação de ácido sulfúrico influencia na espessura removida da camada de macroesclereídios do tegumento das sementes.Termos par...
-Germination is a complex process that involves molecules properties that make up the cell walls, hydrolytic enzymes that break the bonds between the polymers and action of reactive oxygen substance. Melanoxylon brauna is a forest species of high economic value. In order to evaluate the physiological and biochemical changes that occur in the embryonic axis during germination, fresh matter, length, activities of the enzymes pectin methylesterase, polygalacturonase, superoxide dismutase, catalase, peroxidase and hydrogen peroxide levels were quantified in the embryonic axis. Furthermore, in the micropyle area the composition of carbohydrates and micropyle physical resistance were evaluated with and without drying. During soaking, if there are increases in fresh matter and length of the embryonic axis, there is the same trend of polygalacturonase and pectin methylesterase enzymes. The hydrogen peroxide content was reduced during the soaking, as well as the puncture force of the micropylar area. It is concluded that the seed coat and the cotyledons are responsible for 90% of the water soaked by the seeds. The events in the micropyle and embryonic axis occur independently in the first 16 hours. The weakening of the micropyle features an elastic step and a plastic one. Enzymes pectin methylesterase and polygalacturonase act in cellular expansion of the embryonic axis.Index terms: puncture force, hydrogen peroxide, cell wall, germination.Eventos micropilar e embrionário na hidratação de sementes de Melanoxylon brauna Schott RESUMO -A germinação é um processo complexo que envolve propriedades das moléculas que compõem a parede celular, enzimas hidrolíticas que quebram ligações entre polímeros da pectina e da hemicelulose e ação das substâncias reativas de oxigênio. Melanoxylon brauna é uma espécie florestal de alto valor econômico. Com o objetivo de avaliar alterações fisiológicas e bioquímicas no eixo embrionário durante a germinação, foram quantificados a massa fresca, o comprimento, as atividades das enzimas pectina metilesterase, poligalacturonase, superóxido dismutase, catalase, peroxidase e os teores de peróxido de hidrogênio. Foram avaliados na região micropilar a composição dos carboidratos e a resistência da micrópila seca e sem secagem. Durante a embebição, ocorrem aumentos na massa fresca e comprimento dos eixos embrionários, mesma tendência das enzimas poligalacturonase e pectina metilesterase, que participam da expansão celular. O conteúdo de peróxido de hidrogênio reduziu durante a embebição, assim como a força de ruptura da região micropilar. Conclui-se que o tegumento e os cotilédones são responsáveis por 90% da água embebida pelas sementes. Os eventos na micrópila e eixo ocorrem independentes nas primeiras 16 horas. ALTERAÇÕES FISIOLÓGICAS E ATIVIDADE ENZIMÁTICA EM SEMENTES ARMAZENADAS DE Melanoxylon brauna Schott.RESuMO: A temperatura e a umidade relativa estabelecem as condições fundamentais na conservação de sementes, quer sejam recalcitrantes, quer sejam ortodoxas. A conservação de sementes de braúna sob umidades relativas de 55, 75 e 93% e temperatura de 20ºC foi realizada com os objetivos de avaliar as respostas da germinação e do vigor, relacionandoas à permeabilidade da membrana, atividade de enzimas, teor e peroxidação de lipídios. A qualidade das sementes decresce com o aumento do teor de água do ambiente, com decréscimo também das atividades das enzimas superóxido dismutase e catalase e aumento da atividade da enzima glicose-6-fosfato desidrogenase. A permeabilidade das membranas é ampliada com o aumento da deterioração das sementes. Não há relação entre as alterações no teor e peroxidação de lipídios e a deterioração das sementes. PHYSIOLOGICAL AND ENZYME ACTIVITY CHANGES IN STORED SEEDS OF Melanoxylon brauna Schott.AbSTRACT: Temperature and relative humidity set the basic conditions for the preservation of recalcitrant and orthodox seeds. The seed conservation of braúna under relative humidities of 55, 75 and 93% at 20°C was performed with the aim of evaluating germination and vigor responses, relating them to the membrane permeability, enzyme activity, content and lipid peroxidation. Increasing environment water content decreases seed quality and also decreases the activity of the enzymes superoxide dismutase and catalase, and increases activity of glucose-6-phosphate dehydrogenase. The membrane permeability increases with increasing deterioration of the seeds. There is no relationship between the changes in lipids content and peroxidation and seed deterioration.
-Seed germination is dependent on abiotic factors, temperature being one of the main ones, whose influence causes seed damage under extreme conditions. The aim of the present study was to investigate the effect of different temperatures during germination of D. nigra seeds and their physiological and biochemical implications. We assessed germination percentage and production of superoxide anion (O 2 -) and hydrogen peroxide (H 2 O 2 ) in seeds subjected to temperatures of 5, 15, 25, 35 and 45 ºC for different periods of time. Hydration is promoted at 45 ºC and inhibited at 5ºC, without germination in either, whereas it is minimal at 15 °C and at a maximum level at 25 °C. Superoxide production increases at higher temperatures (25 and 35 ºC) after 72 hours of hydration, coinciding with the beginning of radicle protrusion. Production of hydrogen peroxide decreases at all temperatures, except for 5 ºC, with values near each other at temperatures of 15, 25, and 35 ºC, where there was radicle protrusion. <p>O manejo do solo pode alterar as propriedades físicas do solo, sendo de grande importância para garantir o estabelecimento de plântulas e produtividade. A inovação de implementos e máquinas agrícolas e excesso de tratos culturais durante o manejo são os principais fatores que interferem nestas características. Assim, objetivou-se avaliar o efeito da compactação de solos com diferentes classes texturais (argilosa e muito argilosa) no crescimento inicial de ervilha. O experimento foi montado em casa de vegetação com sementes de ervilha Itapuã 600. Cada tratamento contou com 4 repetições de 50 sementes em diferentes graus de compactação dos solos, com aplicações três cargas sobre os solos que resultaram em: argiloso: controle: 0,21, T1: 0,26, T2: 0,28 e T3: 0,33 MPa, textura muito argilosa: controle: 0,40, T1: 0,42, T2: 0,46 e T3: 0,53 MPa. Foram avaliados a porcentagem de emergência, IVE, VE, comprimento de parte aérea e raiz, massa seca de parte aérea, raiz e massa seca total. Para as duas classes texturais a compactação teve influência sobre a massa seca da parte aérea no solo com textura argilosa e para IVE no solo com textura muito argilosa. Porém, ambos não tiveram relação direta com o nível de compactação.</p><p align="center"><strong><em>Initial growth of</em></strong><strong><em> pea under different compaction levels of two types of soils</em></strong></p><p><strong>Abstract: </strong>Soil management can alter the physical properties of the soil, being of great importance to ensure adequate levels of crop productivity. The innovation of agricultural implements and machinery and excess cultural practices during handling are the main factors that affect these characteristics. The objective of this study was to evaluate the effect of compaction of soils with different texture classes (clay and heavy clay) on the initial growth of pea. The experiment was conducted in a greenhouse at the Department of Fitotecnia at UFV. Itapuã 600 pea seeds were used. Each treatment consisted of 4 replicates of 50 seeds for each compression level (T1, T2, T3 and without compression) for the two soil textural classes. Were evaluated: emergency, IVE, VE, length of shoot and root dry weight of shoot, root mass and total dry mass. For both textural classes compaction is not observed influence on the parameters analyzed, except for dry weight of shoots in clayey soil, IVE in clayey soil. However, both were not directly related to the level of compression.</p><p><strong> </strong></p>
