-The objective of this paper was to evaluate the effect of nitric oxide (NO) as a protecting agent of sesame seeds submitted to different concentrations of cadmium. The treatments were: water (control), water increased by sodium nitroprusside (SNP) and other treatments regarding the concentrations of cadmium increased by SNP. The following determinations were done: germination, first count of germination, germination speed index, length of hypocotyl and radicle and dry matter of hypocotyl and radicle, besides quantification of enzyme activities, superoxide dismutase, catalase, ascorbate peroxidase and total peroxidases. The statistical design was entirely randomized with five replicates. The data was submitted to a variance analysis and the averages obtained for the treatments were compared by the Tukey test at 5% significance. The averages obtained in the treatments with and without SNP were compared by the F test at 5% probability. The NO due to the application of SNP was beneficial, providing an increase in germination, vigor and growth of seedlings. There was a progressive increase of the antioxidant enzymes activity in the period of 0 to 24 hours, showing an organization of the antioxidant system in the sesame seeds throughout germination time.Index terms: SNP, vigor, germination, antioxidant system. Ação protetora do óxido nítrico em sementes de gergelim (Sesamum indicum L.) submetidas ao estresse por cádmio RESUMO -Objetivou-se avaliar o efeito do óxido nítrico (ON) como agente protetor em sementes de gergelim submetidas a diferentes concentrações de cádmio. Os tratamentos foram: água (controle), água acrescida de nitroprussiato de sódio (SNP) e os demais tratamentos referentes às concentrações de cádmio e cádmio acrescido de SNP. Foram feitas as seguintes determinações: germinação, primeira contagem de germinação, índice de velocidade de germinação, comprimento de hipocótilo e radícula e massa seca de hipocótilo e radícula, além da quantificação da atividade das enzimas, superóxido dismutase, catalase, ascorbato peroxidase e peroxidases totais. O delineamento estatístico utilizado foi o inteiramente casualizado com cinco repetições. Os dados foram submetidos à análise de variância e as médias obtidas para os tratamentos foram comparadas pelo teste de Tukey a 5% de significância. As médias obtidas nos tratamentos com e sem SNP foram comparadas pelo teste F a 5% de probabilidade. O ON devido à aplicação de SNP foi benéfico, proporcionando aumento na germinação, vigor e crescimento de plântulas. Houve aumento progressivo da atividade das enzimas antioxidativas no período de 0 a 24 horas, demonstrando organização do sistema antioxidante nas sementes de gergelim com o decorrer do tempo de germinação.Termos para indexação: SNP, vigor, germinação, sistema antioxidativo.
-Seed dormancy is a phenomenon that affects the distribution of plant species in time. However, it may slow germination and consequently hinder seedling production in nurseries. Many seeds of forest species of the Fabaceae family, such as Libidibia ferrea (Brazilian ironwood), have physical dormancy caused by impermeability of the seed coat to water. Therefore, the aim of this study was to evaluate the physiological, physical, and morpho-anatomical characteristics of L. ferrea seeds before and after application of different treatments for overcoming dormancy. We evaluated the imbibition curves, germination percentage, germination speed index (GSI), the force required to puncture the micropylar region, and morphoanatomical changes through images obtained from a scanning electron microscope (SEM) before and after application of treatments for overcoming dormancy. L. ferrea seeds immersed in sulfuric acid show a triphasic pattern of imbibition. The required force to puncture the micropylar region of L. ferrea seeds is less for both the treatment with boiling water and for treatments with sulfuric acid. In addition, the required force to puncture the micropylar region decreases during imbibition of the seeds after application of sulfuric acid. The time of application of sulfuric acid influences the thickness of the material removed from the macrosclereid layer of the seed coat.Index terms: Libidibia ferrea ((Mart. ex Tul.) L.P. Porém, pode retardar a germinação, e consequentemente, dificultar a produção de mudas em viveiros. Muitas sementes de espécies florestais da família Fabaceae, como Libidibia ferrea (pau-ferro), apresentam dormência física causada pela impermeabilidade do tegumento à água. Portanto, o objetivo neste trabalho foi avaliar as características fisiológicas, físicas e morfo-anatômicas das sementes de L. ferrea antes e após aplicação de diferentes tratamentos para superação de dormência. Avaliaram-se as curvas de embebição, a porcentagem de germinação, o índice de velocidade de germinação, a força requerida para ruptura da região micropilar e as alterações morfo-anatômicas por meio de imagens obtidas de microscópio eletrônico de varredura antes e após a aplicação de tratamentos para superação de dormência. Sementes de L. ferrea imersas em ácido sulfúrico apresentam padrão trifásico de embebição. A força de ruptura da região micropilar de sementes de L. ferrea é menor tanto para o tratamento com água fervente quanto para os tratamentos com ácido sulfúrico. Além disso, a força necessária para ruptura da região micropilar diminui durante a embebição das sementes após aplicação de ácido sulfúrico. -Seed germination is dependent on abiotic factors, temperature being one of the main ones, whose influence causes seed damage under extreme conditions. The aim of the present study was to investigate the effect of different temperatures during germination of D. nigra seeds and their physiological and biochemical implications. We assessed germination percentage and production of superoxide anion (O 2 -) and hydrogen peroxide (H 2 O 2 ) in seeds subjected to temperatures of 5, 15, 25, 35 and 45 ºC for different periods of time. Hydration is promoted at 45 ºC and inhibited at 5ºC, without germination in either, whereas it is minimal at 15 °C and at a maximum level at 25 °C. Superoxide production increases at higher temperatures (25 and 35 ºC) after 72 hours of hydration, coinciding with the beginning of radicle protrusion. Production of hydrogen peroxide decreases at all temperatures, except for 5 ºC, with values near each other at temperatures of 15, 25, and 35 ºC, where there was radicle protrusion. RESUMO -Sementes de Platymiscium pubescens foram colocadas para embeber em água, sendo retiradas amostras para as caracterizações bioquímica e cinética da enzima alfa-galactosidase do eixo embrionário e dos cotilédones. A atividade específica no eixo embrionário aumenta de zero até o tempo de 96 horas de embebição, estabilizando em seguida. A atividade da enzima dos cotilédones mostrou pequeno incremento durante esse mesmo tempo. A alfa-galactosidase do eixo embrionário apresentou atividade máxima no intervalo de pH de 4,5 a 6,0. Por outro lado, para a enzima proveniente dos cotilédones, a maior atividade foi detectada na faixa de 4,0 a 6,0. A temperatura de 55ºC foi a que mais estimulou as atividades da alfa-galactosidase do eixo embrionário e dos cotilédones. As enzimas do eixo embrionário e dos cotilédones mostraram-se termotolerantes, não se alcançando a meia vida na temperatura de 40ºC, no período de 1.500 minutos. A atividade da alfa-galactosidase do eixo embrionário foi inibida por melibiose, CuSO 4 e SDS, enquanto a dos cotilédones foi por todos os efetores, exceto com SDS, CuSO 4 e galactose que tiveram efeito neutro sobre a atividade da alfa-galactosidase dos cotilédones. CARACTERIZATION OF ALFA-GALACTOSIDASE IN EMBRYONIC AXIS AND COTYLEDONS OF Platymiscium pubescens SEEDSABSTRACT: Platymiscium pubescens seeds were placed to soak in water and sampled for biochemistry and kinetic characterizations of embryonic axis and the alfa-galactosidase cotyledon enzyme. The specific activity in the embryonic axis increased from zero to 96 hours of imbibition, stabilizing soon after. The activity of the cotyledon enzyme showed a small increase in the same period. The alfa-galactosidase of the embryonic axis presented its maximum activity in the interval of pH 4.5 to 6.0. On the other hand, for the cotyledon enzyme, the highest activity was detected in the interval of 4.0 to 6.0. The temperature of 55ºC was best to stimulate embryonic axis and cotyledon alfa-galactosidase acticvity. Enzymes of the embryonic axis and the cotyledons were heat tolerant, not reaching the half life at 40ºC, in 1.500 minutes. The embryonic axis alfa-galactosidase activity was inhibited by melibiose, CuSO 4 and SDS, while that of cotyledons was for all the effectors, except for SDS, CuSO 4 and galactose which had neutral effect. The values of K M for the embryo and for the cotyledon alfa-galactosidases were 3.37 and 0.26 mM, respectively.
