1 Endothelin-1 causes ET A receptor-mediated enhancement of capsaicin-induced nociception in mice. We have assessed if this hyperalgesic eect of endothelin-1 is also accompanied by other proin¯ammatory eects, namely nociception and oedema, and characterized the endothelin ET receptors involved. 2 Intraplantar (i.pl.) hind-paw injection of endothelin-1 (0.3 ± 30 pmol) induced graded nociceptive responses (accumulated licking time: vehicle, 20.5+3.3 s; endothelin-1 at 30 pmol, 78.1+9.8 s), largely con®ned to the ®rst 15 min. Endothelin-1 (1 ± 10 pmol) potentiated ipsilateral capsaicininduced (0.1 mg, i.pl.; at 30 min) nociception (vehicle, 40.2+2.6 s; endothelin-1 at 10 pmol, 98.4+5.8 s, but 30 pmol was inactive), and caused oedema (increase in paw weight 5 min after capsaicin: vehicle, 46.3+2.3 mg; endothelin-1 at 30 pmol, 100.3+6.1 mg). 3 Selective ET B receptor agonists sarafotoxin S6c (up to 30 pmol) and IRL 1620 (up to 100 pmol) were inactive, whereas endothelin-3 (up to 30 pmol) induced only modest oedema. 4 ET A receptor antagonists BQ-123 (1 nmol, i.pl.) or A-127722-5 (6 mmol kg 71 , i.v.) prevented all eects of endothelin-1 (10 pmol), but the ET B receptor antagonist BQ-788 (1 or 10 nmol, i.pl.) was ineective. 5 BQ-788 (10 nmol, i.pl.) unveiled hyperalgesic eects of 30 pmol endothelin-1 and endothelin-3. Sarafotoxin S6c (30 pmol, i.pl.) did not modify endothelin-1-induced (10 pmol) nociception or oedema, but abolished hyperalgesia. 6 Thus, endothelin-1 triggers ET A receptor-mediated nociception, hyperalgesia and oedema in the mouse hind-paw. Simultaneous activation of ET B receptors by endothelin-1 or selective agonists can limit the hyperalgesic, but not the nociceptive or oedematogenic, eects of the peptide.
1 The contribution of endogenous endothelins to nociceptive responses elicited by ovalbumin (OVA) in the hind-paw of mice sensitised to this antigen (50 mg OVA þ 5 mg Al(OH) 3 , s.c., 14 days beforehand) was investigated. 1 mg). Treatment with BQ-123 (3 nmol, i.pl.) reduced only the hyperalgesic effect of compound 48/80, whereas BQ-788 (3 nmol) was ineffective. 6 Thus, immune-mediated Type I hypersensitivity reactions elicit mast cell-and endothelindependent nociception in the mouse hind-paw, which are mediated locally by both ET A and ET B receptors. The nocifensive response to antigen is amenable to blockade by systemic treatment with dual ET A /ET B or selective ET A receptor antagonists, but is sharply potentiated by systemic selective ET B receptor antagonist treatment. The apparently distinct roles played by ET B receptors in this phenomenon at local and other sites remain to be characterised.
OBJETIVO: Conhecer a automedicação em crianças moradoras de uma cidade da região Sul do Brasil. MÉTODOS: Estudo transversal com amostra selecionada por sorteio, inicialmente identificando as equipes de Estratégia Saúde da Família e, posteriormente, as residências que seriam visitadas. Foi adotado, para a entrevista com o responsável pelas crianças, um questionário para avaliar o perfil da criança, da família e da prática da automedicação. Para apresentação dos dados, adotou-se a estatística descritiva e o teste do qui-quadrado (p<0,05). RESULTADOS: Entrevistaram-se os responsáveis pelas crianças em 83 domicílios. Foram coletadas informações de 121 crianças (seis meses a 14 anos). Dos entrevistados, 75% afirmaram já ter praticado a automedicação, sendo as mães responsáveis por 95% desses casos. Quanto às situações que motivaram a automedicação, praticidade (88%), febre (58%) e dor (12%) foram as mais relatadas. O paracetamol (45%) e a dipirona (15%) foram os fármacos mais utilizados. A análise estatística evidenciou associação entre a reutilização de antigas prescrições e a idade da criança inferior a sete anos, bem como entre a utilização de medicamentos sem prescrição de profissional habilitado e morar em domicílios com mais de quatro pessoas. CONCLUSÕES: A automedicação é uma prática frequente na população investigada, sendo geralmente mais comum em crianças de até sete anos e realizada principalmente pelas mães; esse fato sugere a necessidade de promover educação em saúde que vise à promoção do uso racional de medicamentos.
Este trabalho avaliou o conhecimento e aceitação das Práticas Não-Convencionais em Saúde (PNCS) por estudantes do curso de Medicina da Universidade do Sul de Santa Catarina (Unisul). A pesquisa realizada foi do tipo descritiva de campo, aplicada na forma direta extensiva por meio de questionários. As perguntas avaliaram o conhecimento e o interesse a respeito de PNCS. Os entrevistados (n = 197) afirmaram conhecer a maioria das PNCS apresentadas no estudo, destacando-se ioga (96,6%), homeopatia (92,9%), chás caseiros (91,9%), acupuntura e orações (88,8%), além de benzedeiras (83,2%), com porcentagem superior a 80%. Ainda que estas práticas não façam parte do currículo atual deste curso, mais de 50% dos alunos afirmaram ter interesse em aprender sobre 10 das 13 PNCS apresentadas neste estudo. Para PNCS como ioga, acupuntura, fitoterapia e orações, mais de 50% dos alunos afirmaram que indicariam ou apoiariam o uso delas por seus pacientes. Concluiu-se que há interesse dos acadêmicos em práticas não-convencionais, bem como evidências da necessidade de inclusão de disciplinas curriculares que abordem as PNCS nos cursos de graduação em Medicina.
The present study investigates the influence of endothelin (ET) related peptides (0.3-30 pmol/paw) on both phases of nociception and on edema induced by intraplantar injection of formalin (0.5% in 20 microL) in the mouse hind paw. The first phase of nociception (0-5 min after injection) was significantly potentiated by simultaneous injection of either ET-1 (10 or 30 pmol/paw) or ET-3 (10 pmol/paw), but not of the selective ET3 receptor agonist sarafotoxin S6c (SRTX-c; up to 30 pmol/paw). All three peptides potentiated the second phase (10-30 min after injection) of formalin-induced nociception (at 3-30, 1-30, and 10-30 pmol/paw for ET-1, ET-3, and SRTX-c, respectively), whereas only ET-1 (10 or 30 pmol/paw) effectively enhanced edema caused by formalin (30 min after injection). Histamine also potentiated all three responses triggered by formalin, but was 30- to 100-fold less potent than ET-1. Treatment with the mixed ETA/ETB receptor antagonist bosentan (10 mg/kg i.p., 1 h beforehand) did not influence nociceptive and edematogenic responses to formalin or their potentiation by histamine (3 nmol/paw), but did inhibit the potentiations induced by ET-1 (10 pmol/paw). Thus, ET-1 potentiates formalin-induced nociception and edema in the mouse. These actions are possibly mediated via ETB and ETA receptors, respectively, but their true identity and the mechanisms involved still remain to be fully elucidated.
