Treatment of cutaneous leishmaniasis (CL) is difficult due to the scarce number of drugs able to eliminate completely the intracellular form of the parasite. In the present study, the aim was to evaluate: i) phytochemical properties of extracts from Libidibia ferrea; ii) antileishmanial activity of extracts from L. ferrea against promastigotes and amastigotes of Leishmania (Leishmania) amazonensis and Leishmania (Viannia) guyanensis; iii) the effects of topical treatment using hydrogel containing active extract of L. ferrea on golden hamsters infected with L. (L.) amazonensis. Extracts from leaves, branches and fruits of L. ferrea were obtained with hexane and methanol and were tested by in vitro assays in promastigotes and murine macrophages J774 experimentally infected with amastigotes of Leishmania. Groups of hamsters with CL received topical treatment with a formulation of extract (10%) hydrogels, 50 mg.day ) e baixa citotoxicidade em células de macrófagos J774. Os resultados destacaram que as lesões cutâneas de animais que receberam tratamento com hidrogel + extrato (GelFrMeOH) apresentaram redução significativa (42,78%), menos úlceração e redução da carga parasitária detectada por RT-qPCR, microscopia e análise em cultura e alterações histopatológicas, como menor infiltrado inflamatório após 40 dias de tratamento. A análise química demonstrou que FrMeOH contém altos níveis de compostos fenólicos. Os resultados apontam para uma possível terapia alternativa para CL utilizando fitoterápicos. ResumoA Leishmaniose é considerada uma doença negligenciada. Nos últimos anos estudos tem demonstrado que várias plantas possuem atividade contra esses patógenos, servindo de base para a formulação de fitoterápicos ou como alvo para a obtenção de princípios ativos contra a doença. Esta pesquisa teve como objetivo avaliar a atividade biológica in vitro antileishmania dos extratos hexânicos e metanólicos das flores e folhas de Tanacetum vulgare contra Leishmania amazonensis e Leishmania guyanensis. Foi realizada a extração das flores e folhas do material vegetal a partir dos solventes hexânico e metanólico. The results showed that the extract metanolic of the flower showed activity against L. amazonensis (IC50 = 31.5 μg mL -1 ) and L. guyanensis (IC50 < 100 μg mL -1 ) in 72 h of incubation. Therefore, it is verified that the methanolic extract of the T. vulgare flower performed better in relation to antileishmania activity, allowing the possibility of future studies to determine its therapeutic efficacy.
RESUMOAs Doenças Tropicais Negligenciadas (DTNs) são um grupo de doenças que afetam principalmente a população de países tropicais e em desenvolvimento de baixa a média renda. Nos últimos anos, no cenário internacional, tem havido debates sobre as formas de prevenir, combater e erradicar as doenças tropicais negligenciadas, bem como pesquisas para desenvolver novos fármacos que ajudem no tratamento dessas doenças, onde através da literatura obtivemos inúmeros estudos científicos mostrando moléculas bioativas provenientes de plantas com atividades biológicas, dentre elas Piper aduncum, já que seu composto majoritário, o dilapiol, apresenta uma estrutura química natural que pode ser modificada através de semissíntese, uma alternativa de obtenção de novas moléculas ativas, esses análogos do dilapiol, testados sobre doenças negligenciadas, principalmente contra leishmaniose cutânea, inibindo as formas promastigotas e amastigotas de Leishmania de diversas espécies.
Las Enfermedades Tropicales Desatendidas (NTD, por sus siglas en inglés) son un grupo de enfermedades que afectan principalmente a la población de países tropicales y en desarrollo de ingresos bajos a medios. En los últimos años, en el panorama internacional, ha habido debates sobre las formas de prevenir, combatir y erradicar las enfermedades tropicales desatendidas, así como investigaciones para desarrollar nuevos fármacos que ayuden en el tratamiento de estas enfermedades, donde a través de la literatura hemos obtenido numerosos estudios científicos que muestran moléculas bioactivas de plantas con actividades biológicas, entre ellas Piper aduncum, ya que su compuesto mayoritario, el dilapiol, tiene una estructura química natural que puede ser modificada mediante semisíntesis, una alternativa para la obtención de nuevas moléculas activas, estos análogos del dilapiol, probados en enfermedades desatendidas, principalmente contra la leishmaniasis cutánea, inhibiendo las formas promastigotes y amastigotes de Leishmania de varias especies.
A Leishmaniose tegumentar é uma doença tropical negligenciada de grande relevância. O seu arsenal terapêutico é antigo e limitado, pois não atende a necessidade dos enfermos, com aplicações invasivas, inúmeros efeitos adversos e tratamento prolongado. Desse modo, existe o estimulo pela busca de novas substâncias candidatas para uma farmacoterapia mais eficiente. O estudo teve como objetivo a seleção de substâncias bioativas de origem orgânica e inorgânica processadas em laboratório de pesquisa referência no Estado do Amazonas, que apresentassem atividade potencial no tratamento da Leishmaniose cutânea (LC). As substâncias foram testadas in vitro para avaliação da atividade antiLeishmania. A partir dos ensaios de triagem foram selecionados os fármacos mais promissores para os estudos pré-clínicos e elaboração de protótipo, desenvolvendo formulações candidatas a serem produzidas e recebidas no mercado, como drogas para o tratamento alternativo ou em combinação com outras drogas já utilizadas comercialmente no tratamento da LC. Os fármacos e formulações mais promissoras foram então testados por meio de experimentação animal para confirmação de sua atividade in vivo. Essa etapa serviu para avaliar sua ação, toxicidade, dose, efeito e eficácia. Os resultados foram promissores, com 56 substâncias avaliadas, duas substâncias de origem orgânica e uma inorgânica nanoparticulada indicadas como candidatas ao tratamento, duas patentes e três depósitos de patentes. A próxima etapa a partir desses resultados, deverá ser a realização dos estudos clínicos, para eleger a formulação para o tratamento alternativo da Leishmaniose cutânea.
