The regeneration of tissues affected by periodontal disease is a complex process; it encompasses the formation of bone, cementum and periodontal ligament. We developed a semi-rigid PLGA (polylactide-co-glycolide acid)/CaP (calcium phosphate) bilayered biomaterial construct to promote periodontal regeneration, which has a continuous outer barrier membrane and an inner topographically complex component. Our experimental model compared periodontal prophylaxis alone with prophylaxis and biomaterial implantation in the treatment of class II furcation defects in dogs. Clinical evaluation, micro-computed tomography, histology and backscattered electron imaging were used for data analysis. Healing occurred uneventfully and bone volumetric values, trabecular number and trabecular thickness were all significantly greater in the treated group; while trabecular separation was significantly greater in the control group. New cementum, bone, and periodontal ligament with Sharpey fibre insertions were only seen in the treated group. Although periodontal regeneration has been reported elsewhere, the advantages of employing our bilayered PLGA + CaP construct are twofold: 1)it did not collapse into the defect; and, 2) its inner side was able to retain the blood clot throughout the buccal defect. The result was greater periodontal regeneration than has previously been reported with traditional flexible membranes.
Com o objetivo de estudar a hidroxiapatita sintética (HAP-91) como substituto ósseo, foram utilizados oito cães adultos, sem raça definida, clinicamente sadios. Após protocolo anestésico e cirúrgico habituais, foi provocado um defeito ósseo na diáfise proximal das tíbias esquerda e direita, sendo implantada a HAP-91 apenas na tíbia direita. Os animais, dois de cada vez, foram sacrificados aos 8, 30, 60 e 120 dias após a cirurgia, quando foram obtidas amostras do local da lesão, que foram fixadas, lavadas, polimerizadas, cortadas e submetidas a dupla coloração em solução aquosa de acetato de uranila a 1% e em solução de citrato de chumbo. Essas secções foram avaliadas e fotografadas ao microscópio eletrônico de transmissão. Os componentes tissulares na tíbia tratada e na controle foram similares. A absorção da HAP-91 caracterizou-se pela presença de células multinucleadas na interface entre HAP-91 e osso, morfologicamente consideradas como osteoclastos. Ainda, encontraram-se grânulos de HAP-91 no interior de células morfologicamente caracterizadas como macrófagos. A absorção celular de grânulos de HAP-91, concomitante com a formação adjacente de osso novo, sugere que a osteointegração da HAP-91 seja análoga ao processo normal de reabsorção-aposição óssea.
RESUMOAvaliaram-se os compósitos de hidroxiapatita sintética e carbono (HAC) e hidroxiapatita sintética, carbono e fosfato biácido de sódio (HACF), ambos na forma sólida, como substituto ósseo em 36 coelhos adultos. Após protocolos anestésico e cirúrgico habituais, retirou-se um fragmento de aproximadamente 2cm de comprimento no terço médio da ulna esquerda. Os animais do grupo 1 receberam, em cada falha provocada, blocos de HAC e HACF, e os do grupo 2 somente HAC. Os do grupo 3 não receberam tratamento. Os animais foram submetidos a avaliações clínicas e radiológicas nos dias 8, 15, 30, 60, 120 e 180 de pós-operatório. Nas mesmas datas, dois animais de cada grupo foram sacrificados e deles colheuse material para histologia. Observou-se evolução normal da regeneração óssea em todos os animais, sendo mais precoce nos dos grupos que receberam os compósitos. BackgroundPeriodontal disease is a highly prevalent illness that affects many dogs, reaching up to 85 % prevalence in individuals over the age of 4 years. Currently the drug of choice for combating the formation of dental plaque in these animals, the etiologic agent of the disease, is chlorhexidine, which has several side effects reported. Thus, surveys are conducted throughout the world in order to identify potential substitutes for antimicrobial therapy and prevention of periodontal disease. The objective of the work was to evaluate the antimicrobial activity of β-caryophyllene against bacteria from dog’s dental plaque in vitro and in vivo. The minimum inhibitory concentration was evaluated by agar microdilution assay, the induction or inhibition of bacterial adherence by sub-inhibitory concentrations in 96-well plates, and reduction of dental plaque formation in mongrel dogs subjected to topical solution with β-caryophyllene for 15 days.ResultsResults showed minimum inhibitory concentrations above 100 mg/mL for 25 % of the isolates, 100 mg/mL for 3 %, 50 mg/mL for 25 %, 25 mg/mL for 12 %, 12.5 mg/mL for 19 % and 6.25 mg/mL for 16 %. Bacterial adherences of three Enterococcus sp., one Streptococcus sp., one Haemophilus sp., one Aerococcus sp., one Bacillus sp. and one Lactococcus sp. isolates were inhibited by subinhibitory concentration. One Lactococcus sp., one Bacillus sp. and one Streptococcus sp. were stimulated to adhere by concentrations of 0.19, 1.56 and 0.78 mg/mL, respectively. In vivo assay showed reduction in dental plaque formation by β-caryophyllene, with final plaque coverage of 23.3 ± 2.6 % of the total area of the teeth, with significant difference compared with chlorhexidine group (37.5 ± 3.7 % - p < 0.05) and negative control group (65.5 ± 2.5 % - p < 0.001).ConclusionsThe results showed that β-caryophyllene has antimicrobial activity against the proliferation of dog’s dental plaque-forming bacteria representing a suitable alternative to the use of chlorhexidine in prophylaxis and treatment of periodontal disease of dogs.
Optically transparent membranes from bacterial cellulose (BC)/polycaprolactone (PCL) have been prepared by impregnation of PCL acetone solution into dried BC membranes. UV-Vis measurements showed an increase on transparency in BC/PCL membrane when compared with pristine BC. The good transparency of the BC/PCL can be related to the presence of BC nanofibers associated with deposit of PCL nano-sized spherulites which are smaller than the wavelength of visible light and practically free of light scattering. XRD results show that cellulose type I structure is preserved inside the BC/PCL membrane, while the mechanical properties suggested indicated that PCL acts as a plasticizer for the BC membrane. The novel BC/PCL membrane could be used for preparation of fully biocompatible flexible display and biodegradable food packaging.
RESUMO Prevalência da doença periodontal em cães e ciência dos proprietários a respeito da doençaA doença periodontal (DP) é amplamente conhecida pelos médicos veterinários por sua alta prevalência e graves conseqüências aos cães. Objetivou-se neste estudo avaliar sua ocorrência em cães da micro-região de Viçosa atendidos no Hospital Veterinário da Universidade Federal de Viçosa (HVT), assim como o grau de conhecimento dos proprietários em relação à doença. Para tanto, todos os cães atendidos no HVT no período de 10 de março de 2009 a 30 de novembro de 2009, em dias alternados, tiveram suas cavidades orais examinadas. Informações, abrangendo dados de anamnese como idade, tipo de alimentação, queixa principal e anuência do proprietário, halitose, presença de cálculo dentário, inflamação e recessão gengival e perda dentária foram colhidas. Observou-se uma prevalência de 88,67% para a DP em cães encaminhados ao HVT, sendo que 2,67% foram encaminhados devido a essa doença. De todos os proprietários que participarem do estudo, 43,83% conheciam a doença periodontal e, destes 17,46% utilizavam alguma medida de prevenção ou tratamento. Portanto, a doença periodontal é de grande prevalência e os proprietários não estão cientes da doença. Com isso, torna-se necessário estabelecer um programa de esclarecimento dos proprietários de cães da região atendida pelo HVT-UFV quanto à doença periodontal.Palavras-chave: caninos, epidemiologia, periodontia.
Digestive tract obstruction in pets attended in a Veterinary Hospital during 2010This study aimed to examine the cases of simple, partial and complete obstructions of the digestive tract of small animals treated at the Veterinary Hospital of the Federal University of Viçosa during the year of 2010, reporting the evolution period, clinical and laboratorial changes, and the outcome of patients, as well as determining breed, sexual and age predilections. Predisposition of small breeds and ages ranging from two to twelve months and after five years was observed, with breed predilection for Teckels The largest number of foreign bodies was found in the stomach and intestines, with greater severity and higher mortality rate when compared with esophageal foreign bodies. The longest clinical course contributed to higher mortality rates. Clinical symptoms of the animals were quite diverse, but apathy and anorexia occurred in most cases. Regurgitation and vomiting were frequent when the foreign body was esophageal or gastrointestinal, respectively. The laboratory tests were nonspecific. O objetivo deste estudo é relatar os tipos de obstrução do trato digestório, após ingestão de corpos estranhos, em animais de companhia, atendidos no Hospital Veterinário da Universidade Federal de Viçosa (HVT-UFV), durante o ano de 2010; a predisposição racial e por faixa etária; a localização da obstrução; os sintomas relacionados; os achados laboratoriais; o tratamento proposto e a evolução clínica dos pacientes. Foi observada predisposição para animais de raça pequena e de faixas etárias de até um ano e após cinco anos, com predileção pela raça Teckel. O maior número de corpos estranhos foi encontrado no estômago e intestinos, com maior gravidade e elevada taxa de mortalidade, em relação aos encontrados no esôfago. O maior período de evolução do quadro clínico, desde a observação inicial, contribuiu para maiores taxas de mortalidade. A sintomatologia clínica dos animais foi variada, mas quadros de apatia e de anorexia ocorreram na maioria dos casos. Regurgitação e vômito foram frequentes, quando a localização do corpo estranho foi esofágica ou gastrointestinal, respectivamente. In order to test the performance of bacterial cellulose/polycaprolactone composite (BC/PCL) and pure bacterial cellulose (BC) as tissue substitutes in rabbits' cornea, a superficial ulcer containing 5mm in diameter and 0.2mm deep was made in the right cornea of 36 rabbits, then a interlayer pocket was created from the basis of this ulcer. Twelve rabbits received BC/PCL membrane and 12 were treated with BC membranes, both membranes with 8mm in diameter. The remaining rabbits received no membrane constituting the control group. The animals were clinically followed up for 45 days. Three animals of each group were euthanized at three, seven, 21, and 45 days after implantation for histological examination of the cornea along with the implant. Clinical observation revealed signs of moderate inflammatory process, decreasing from day 20th in the implanted groups. Histology showed absence of epithelium on the membranes, fibroplasia close to the implants, lymph inflammatory infiltrate with giant cells, collagen disorganization, with a predominance of immature collagen fibers in both groups with implants. Although inflammatory response is acceptable, the membranes used does not satisfactorily played the role of tissue substitute for the cornea during the study period.
