Visando selecionar clones de palma forrageira resistentes à cochonilha-do-carmim, realizou-se um experimento em casa de vegetação no período de setembro de 2001 a janeiro de 2002. A partir de infestação artificial com a cochonilha, foram testados 20 clones de palma forrageira, em delineamento inteiramente casualizado, com quatro repetições. Foram observadas a fixação de colônias sobre os cladódios e a porcentagem de cladódios infestados, por meio de uma escala de notas variando de 0 a 5. Os clones que apresentaram maior resistência ao ataque da praga foram Miúda e Orelha de Elefante.
RESUMOPropõe-se, neste trabalho, estabelecer protocolos de micropropagação para a cv. Miúda (Nopalea cochenillifera-Salm Dyck), considerando-se ser esta espécie resistente à praga cochonilha do carmim. Cladódios jovens, entre 5 e 8cm, foram esterilizados em álcool etílico a 70% e em solução de hipoclorito de sódio a 2%. Explantes de 5 mm 3 foram cultivados em meio de cultura Murashige e Skoog (MS) completo, em um tratamento com três níveis de AIA (0,0; 0,1 e 0,25mg.L -1 ); após seis semanas, os brotos foram transferidos para um meio de crescimento MS básico, suplementado com 3 ou 5% de sacarose, 1,0 mg L -1 de BAP e 0,1 mg L -1 AIA; metade do total de brotos foi cortada longitudinalmente antes da transferência e a outra metade foi transferida intacta; após oito semanas, os brotos foram repicados para meio MS básico sem reguladores de crescimento e, posteriormente, aclimatizados. Não se constatou diferenças em relação a utilização do AIA, na primeira fase. Foi verificada diferença significativa tanto para o corte longitudinal como para o nível de sacarose. O corte proporcionou uma multiplicação 70% maior e o nível de sacarose de 3%, uma multiplicação 50% maior para os explantes não cortados.Palavras-chave: biotecnologia, cultura de tecidos, melhoramento de plantas Micropropagation of pricly-pear cv. Miúda (Nopalea cochenillifera-Salm Dyck), considering that this variety presents resistance to the plague carmine cochineal. In this experiment, young prickly-pear cladodes (measuring between 5 to 8 cm) were sterilized in 70% alcohol and in solution of sodium hypochlorite (2%). Explants measuring 5 mm 3 were cutivated in complete Murashige and Sckoog.(MS) culture medium, in a treatment with three levels of AIA (0; 0.1; 0.25 mg L -1 ). Six weeks later, the shoots were changed to a basic culture medium supplemented with 3 or 5% of sucrose, with 1.0 mg L -1 BAP and 0.5 mg L -1 AIA. Half of them were longitudinally sectioned before the transference, while the other remained intact. Eight weeks later, the shoots were put in a MS medium without growth regulators and posteriorly acclimatized. In "phase one" no differences among the three levels of AIA were observed. Significant difference was verified in the longitudinal cut and in the sucrose level. The cut increased multiplication by 70%, while the best sucrose level (3%) showed a 50% increase in multiplication compared to shoots without cut.
