Background
Diagnosis of tuberculosis (TB) in the era of technological sophistication requires accuracy and speed to provide as much information as possible so that TB treatment can be carried out quickly and precisely. New studies have also begun to be carried out to diagnose TB, one of which is by examining genes, either by looking at polymorphisms, mutations, or expressions. Several previous studies have confirmed the association of MC3R and TB genes with polymorphisms; MC3R is a gene that participates in the regulation of the inflammatory process and is also found in macrophages; therefore, we tried to analyze gene expression in the active TB group, household contacts, and healthy controls for looked at the differences between the three groups and confirmed the correlation of MC3R with TB by seeing which group's gene expression increased the most expression of the three groups so that the results can be considered as a TB diagnostic biomarker in the future.
Methods
This study included 122 people, 49 patients with confirmed TB, 46 close relatives of patients, and 27 healthy controls. This study used a real-time PCR technique to analyze MC3R gene expression in the three groups, and all data were analyzed using Bio-Rad CFXTM software version 3.1 and one-way ANOVA using SPSS 21.0.
Results
The value of MC3R gene expression in the active TB group increased 3.6-fold in the healthy group (p = 0.143), and that of gene expression in the healthy control group increased 1.09-fold in the healthy group (p = 0.007).
Conclusion
There is a relationship between MC3R and TB based on the results of gene expression analysis that increased in the active TB group compared to the household contact group and healthy controls.
BACKGROUND: Melanocortin 3 Receptors (MC3R) levels plays a role in many biological systems, including energy homeostasis and regulation of fat metabolism. However, very few have researched the relationship between MC3R and tuberculosis (TB) and body mass index.
AIM: This study explores the differences in serum MC3R levels in active TB, household contacts, and control groups, as well as at different body mass index status. This study tries to find out the relationship between MC3R and other variables.
METHODS AND MATERIALS: Blood samples were taken from 53 active TB patients, 49 household contacts, and 30 healthy people as controls. The 132 samples were subjected to IGRA and ELISA examinations to determine differences in MC3R levels in all groups.
RESULTS: The highest mean of MC3R levels were found in the active TB group at 1.259.55 (p = 0.028) and had a positive correlation with a value of p = 0.008. In the sex group, men had the highest levels (p = 0.551). In the 30–49 year age group, the median value increased significantly in the three groups (p = 0.028), and there was a correlation between MC3R and the 17–29 year age group, although the correlation was negative (p = 0.021), in the 30–49 year age group with a positive correlation (p = 0.050). The mean MC3R value increased significantly in the overweight group in the three groups (p = 0.006) but did not significantly correlate.
CONCLUSION: The high level of MC3R in TB patients is related to its role as a defence against microbes that enter the body through the immune process to prevent further infection and inflammation. Meanwhile, high levels of MC3R in excess Body mass index were associated with the function of MC3R as an inhibitor of pro-opiomelanocortin (POMC) neurons to release α-MSH.
Penelitian ini dilakukan untuk mengetahui Pengaruh Pengelolaan Keuangan Daerah, Akuntabilitas, dan Transparansi Terhadap Kinerja Keuangan Pemerintah Daerah Kabupaten Sinjai. Penelitian ini menggunakan data primer yang diperoleh dari jawaban responden atas kuesioner yang diberikan. Populasinya dalam penelitian ini adalah seluruh OPD kabupaten Sinjai yang berjumlah 40 instansi, sementara itu teknik pemilihan sampel menggunakan metode purposive sampling dengan memilih unit populasi berupa dinas sehingga diperoleh sampel sebesar 66. Selain itu, penelitian ini menggunakan analisis regresi berganda serta menggunaka SPSS sebagai alat pengolahan data. Berdasarkan pengujin yang dilakukan menunjukkan bahwa Pengelolaan Keuangan Daerah tidak berpengaruh dan tidak signifikan terhadap Kinerja Keuangan Pemerintah Daerah, sedangkan Akuntabilitas berpengaruh positif dan signifikan terhadap Kinerja Keuangan Pemerintah Daerah, dan Transparansi berpengaruh positif dan signifikan terhadap kinerja keuangan pemerintah daerah Kabupaten Sinjai
