A study was conducted to determine the epidemiological status of bovine tuberculosis in the state of Rio Grande do Sul. The state was divided in seven regions, and in each of them, a pre-established number of farms was randomly sampled. In each farm, cows with age equal to or greater than 24 months were selected at random and submitted to the comparative cervical tuberculin test. The animals whose tests were inconclusive were retested with the same diagnostic procedure within a minimum interval of 60 days. In all, 9,895 animals from 1,067 farms were tested. An epidemiological questionnaire was applied in the farms in order to identify risk factors associated with bovine tuberculosis. The prevalence of infected herds in the state was 2.8% [1.8; 4.0] and that of infected animals was 0.7% [0.4; 1.0]. There was a trend towards a concentration of infected herds in the northern part of the state, with a predominance of dairy and mixed herds. The risk factors associated with the condition of infected herds were being a dairy herd (OR = 2.90 [1.40; 6.13]) and herds with 16 or more cows (OR = 2.61 [1.20; 5.49]). Thus, the best strategy to be adopted by the state is the implementation of surveillance systems to detect and remediate the infected herds, preferably incorporating elements of risk-based surveillance. In addition, the state must carry out a solid action of health education so that the producers test animals for bovine tuberculosis before introducing them in their herds. This study aimed to evaluate the effectiveness of a bovine brucellosis vaccination program in Rio Grande do Sul, with prevalence as the indicator, and to identify risk factors for the disease. The state was divided into seven regions. For each region, a predetermined number of properties were randomly sampled, in which a pre-established number of randomly selected females aged over 24 months were tested. The serodiagnosis protocol consisted of a screening test using buffered acidified antigen, followed by a confirmatory test using 2-mercaptoethanol. An epidemiological questionnaire was utilized to identify possible risk factors associated with bovine brucellosis. In the state of Rio Grande do Sul, the prevalence of infected herds was found to be 3.54% [2.49-4.88], and the prevalence of infected animals was 0.98% [0.57-1.57]. In assessments of specific regions, the infected herd prevalence ranged from 0.66% to 3.09%, and among the animals, from 0.06% to 2.03%. In herds comprising 15 or more cows, beef type and pasture sharing emerged as risk factors for bovine brucellosis in the state. The epidemiological status of bovine brucellosis in Rio Grande do Sul has remained unchanged since 2004, even though adequate vaccination coverage has been recorded since 2009. Thus, the state should continue its vaccination program, with emphasis on the quality of the process and on encouraging the use of nonantibody inducing vaccines. In addition, the state must make a greater effort to educate producers on the importance of testing for brucellosis in breeding animals before introducing them onto their properties, and on the importance of avoiding shared grazing among herds whose health conditions are unknown. preestabelecido de propriedades nas quais foi testado um número também preestabelecido de fêmeas com idade igual ou superior a 24 meses, aleatoriamente selecionadas. Nas propriedades foi aplicado um questionário epidemiológico sobre possíveis fatores de riscos associados à brucelose bovina. No estado do Rio Grande do Sul, a prevalência de focos foi de 3.54% [2.49 -4.88] e a de animais 0.98% [0.57 -1.57]. Nas regiões, as prevalências de focos variaram de 0.66% a 9.03% e a de animais de 0.06% a 2.03%. Rebanhos com 15 ou mais vacas, tipologia corte e compartilhamento de pastagens emergiram como fatores de risco para brucelose bovina no estado. A situação epidemiológica da brucelose bovina no Rio Grande do Sul manteve-se estável desde 2004, a despeito de boas coberturas vacinais terem sido registradas a partir de 2009. RESUMO.-[Reação em cadeia da polimerase para o diagnóstico de campilobacteriose genital bovina.] Campilobacteriose genital bovina é uma doença venérea comum em bovinos. A prevalência desta doença pode ser subestimada na maioria das vezes pela natureza microaeróbica do agente etiológico, Campylobacter fetus subspecies venerealis. O propósito do presente estudo foi avaliar a utilização da reação de polimerase em cadeia (PCR) no diagnóstico de campilobacteriose genital em amostras obtidas de aspirado prepucial de touros, muco cervical de vacas e conteú-do abomasal de fetos abortados, coletados em meio enriquecido. Cinco protocolos diferentes de extração de DNA foram testados: termo extração, lise com proteinase K, lise com guanidine isothiocyanate, lise com DNAzol e lise com hexadeciltrimetilamônio brometo (CTAB). A especificidade, sensibilidade e a aplicação da técnica da PCR foram também avaliadas com amostras clínicas e comparadas com bactérias isoladas por cultura padrão. DNA extraído pelo protocolo de CTAB demonstrou os melhores resultados na PCR, e foi capaz de detectar 63 unidades formadoras de colônias de C. fetus por ml de meio. Bovine genital campylobacteriosis is a common venereal disease of cattle; the prevalence of this disease can be underestimated mostly because of the nature of the etiological agent, the microaerobic Campylobacter fetus subspecies venerealis. The purpose of the current study was to evaluate the utilization of polymerase chain reaction (PCR) in the diagnosis of genital campylobacteriosis in samples obtained from bull prepuce aspirate, cow cervical mucus, and abomasum contents of aborted fetuses, collected into enrichment medium. Five different DNA extraction protocols were tested: thermal extraction, lysis with proteinase K, lysis with guanidine isothiocyanate, lysis with DNAzol, and lysis with hexadecyltrimethylammonium bromide (CTAB). The specificity, sensitivity, and technical application of the PCR assay were also evaluated with clinical samples and compared to bacterial isolation by standard culture. DNA extraction by the CTAB protocol provided better results in PCR, and it was able to detect 63 colony-forming units per ml of C. fetus.Out of 277 clinical samples tested, 68 (24%) were positive for Campylobacter fetus using PCR, while only 8 (2.8%) of the samples were positive by bacterial isolation in solid medium, proving the superiority of the PCR technique when compared to the standard isolation method, and providing evidence for its usefulness as a better screening test in cattle for the diagnosis of bovine genital campylobacteriosis. amostras foram positivas por isolamento bacteriológico, provando a superioridade da técnica de PCR quando comparada com métodos padrão de isolamento, e fornecendo evidências de sua utilização como um teste de melhor projeção para diagnóstico em campilobacteriose genital bovina.
RESUMO: Avaliaram-se os resultados de testes para diagnóstico da brucelose e tuberculose bovinas objetivando analisar a distribuição de positividade e características de performance do programa de controle. A análise utilizou como dados secundários todos os resultados de testes para diagnóstico realizados em bovinos no ano de 2008 no estado do Rio Grande do Sul (RS) por médicos veterinários habilitados para atuação no âmbito do Programa Nacional de Controle e Erradicação de Brucelose e Tuberculose Animal. Os dados foram estatisticamente ajustados para minimizar os efeitos de valores extremos e de dados faltantes. Os testes alcançaram 66,80% dos 497 municípios do RS e incluíram a participação de 165 médicos veterinários habilitados. 40,21% dos testes foram realizados nos meses de abril, setembro e outubro, sendo que em 73,90% dos estabelecimentos foram realizados testes para ambas as enfermidades. No caso da brucelose verificou-se que foram testados 35.289 animais, com uma frequência de positivos de 0,25%, enquanto a frequência de estabelecimentos positivos entre os 3.406 testados foi 0,94%. Fêmeas apresentaram frequência de positividade mais elevada (0,29%, de 26.724 testadas) do que machos (0,13%, de 5.235 testados). Animais entre 48-60 meses de idade apresentaram chance de positividade superior às demais faixas etárias (Razão de Chances (RC) =2,63; IC 95%=1,63-4,26). Animais de aptidão leiteira representaram 62,66% dos animais testados, e apresentaram maior chance de positividade do que animais de corte (RC=2,32; IC 95%=1,38-3,90). Adicionalmente, 73,74% dos estabelecimentos testados eram dedicados à produção de leite. Já no caso da tuberculose foram testados 62.149 animais distribuídos em 5.151 propriedades, com frequência de positivos de 0,87% e 3,13%, respectivamente. A chance de positividade aumentou com a idade, sendo que animais com idade acima de 48 meses de idades apresentaram valores superiores aqueles verificados em animais mais jovens (RC=2,07; IC 95%=1,73-2,48). 59,74% dos animais testados eram de aptidão leiteira, os quais apresentaram mais chance de serem positivos do que aqueles de corte (RC=5,03; IC 95%=4,09-6,94). Propriedades leiteiras representaram 78,50% da totalidade dos testes para tuberculose. A análise comparativa do presente trabalho com estudos precedentes de prevalência da brucelose sugere que as ações de controle em curso têm sido efetivas na redução da prevalência da enfermidade no RS. Por outro lado, as informações obtidas no âmbito da tuberculose podem ser indicadoras da condição desta enfermidade, especialmente pela inexistência de estudos de prevalência realizados há menos de 30 anos. Adicionalmente, conclui-se que a utilização de dados secundários, desde que devidamente ajustados, pode ser uma ferramenta eficaz na gestão de programas de saúde animal e em sistemas de monitoramento e vigilância.
Venereal campylobacteriosis is associated with infection of Campylobacter fetus subsp. fetus and Campylobacter fetus subsp. venerealis. The etiological agent is transmitted by natural bull breeding or artificial insemination using contaminated semen. The present study aimed to determine the in vitro susceptibility of C. fetus subsp. venerealis isolates to antimicrobial drugs generally used in clinical and semen treatment. Reference strains of C. fetus subsp. fetus and C. fetus subsp. venerealis and 21 C. fetus isolates were tested. The susceptibility test was performed by using the modified KirbyBauer diffusion disc method. C. fetus subsp. fetus reference strain was resistant to lincomycin and penicillin. C. fetus subsp. venerealis was susceptible to all antimicrobial tested, with exception to C.C. KrewerDnalidixic acid. C. fetus subsp. venerealis samples were sensible to amikacin, ampicillin, cefalotin, streptomycin, gentamycin, penicillin and tetracycline. Drug resistance was observed on 42.86% of lincomycin, 4,76 % of enrofloxacin, and 100% to nalidixic acid. In addition 4.76% of the isolates showed intermediate susceptibility to enrofloxacin, neomycin, polimixin B and 9.52% to lincomycin. The susceptibility of C. fetus isolates to antimicrobial drugs commonly used in clinical and semen treatment was demonstrated.INDEX TERMS: Campylobacteriosis, cattle, semen, antibiomicrobials.
This manuscript reports on the photoelectrochemical properties of anodized 0.1 and 0.15 at% Nb-doped NTs prepared using electrolytes containing different amount of water (2 to 12 wt%). It is shown that Nb-doped NTs perform better than undoped NTs, achieving a photocurrent density up to 1.35 mA/cm 2 at 1 V vs RHE when the water content is sufficiently high (12 wt%). The IPCE also confirms the superiority of Nb-doped NTs, again the larger values (55% IPCE at 300 nm) are obtained when using 12 wt% H 2 O in the electrolyte. EIS analysis and capacitance measurements highlight the peculiarity of the Nb-doped TiO 2 NTs which behave differently than undoped NTs. The electron transport resistance in Nb-doped NTs is similar (10 3 cm 2) to the one of undoped NTs. However, Nb-doped NTs exhibit larger electron mobility and require lower activation energy for electron transport (i.e. a less negative bias) than undoped NTs. Furthermore, Nb-doping, combined with an optimal water content of 12 wt%, is found to be an effective strategy to fully suppress the formation of deep intra band-gap energy states. The presence of Nb in the TiO 2 lattice dictates the fate and the energy of defects introduced during anodization and annealing. These defects are shallower than those observed in undoped NTs.
