3 (5)‐Amino‐pyrazol liefert mit 1,3‐Dicarbonylverbindungen R1COCH2COR2 5, 7‐disubstituierte Pyrazolo[1,5‐a]pyrimidine. Wenn R1 ≠ R2, erhält man definierte Bicyclen über substituierte 5‐Vinylamino‐1‐p‐toluolsulfonyl‐pyrazole, z.B. 15, die aus 5‐Amino‐1‐p‐toluolsulfonyl‐pyrazol, R1COCH2COR2 und katalytischen Mengen Aminsalzen gewonnen werden. Bei der Methylierung von 7‐Hydroxy‐5‐methyl‐pyrazolo [1, 5‐a]pyrimidin 11 werden 4, 5‐Dimethyl‐(20a) und 1, 5‐Dimethyl‐pyrazolo [1, 5‐a]pyrimidin‐ (7) (21a) isoliert. Ein O‐Alkyl‐Derivat von 11 wird aus 7‐Chlor‐5‐methyl‐pyrazolo [1,5‐a]pyrimidin und C3H7ONa gewonnen. Die Strukturen 15, 20a und 21a werden durch Diskussion von τ‐ und J‐Werten (1H‐NMR‐Spektren) für die Pyrazol‐Protonen festgelegt.
Methyl-, Cyclohexyl-und @-Phenyl-Bthylhydrazin werden im Gegensatz zu Phenylhydrazin neutral, sauer und alkalisch am substituierten N-Atom cyan-Bthyliert. Struktur und Cyclisierung der erhaltenen Produkte werden untersucht. Versuche zur Cyanathylierung am unsubstituierten N-Atom verliefen negativ. Obwohl iiber die Cyanathylierung von Aminen umfangreiche Literatur existiert, ist iiber die Addition N-monosubstituierter Hydrazine an Acrylnitril bisher wenig bekannt. Sie kann grundsiitzlich mit einer Anlagerung des unsubstituierten (A) oder des substituierten N-Atoms (B) an das positivierte GAtom des Acrylnitrils beginnen. Beide Moglichkeiten wurden beim Phenylhydrazin realisiert, das in verd. Essigsiiure 1) oder besser in siedendem Athanol2) 3a liefert, in Gegenwart von 50-proz. waDr. HaN-[C H&-NHa H~N-[CH~]S-NH-R + NH3 + R-NHz H z N F F R 2 / 3 t 1) S. Pietru, Boll. sci.
Bicyclische Systeme aus Acetessigester und 5-Amino-l-methyl-, 5-Amino-1-benzyl-sowie 3(5)-Amino-pyrazol Acetessigester liefert mit 5-Amino-1 -methyl-(1 b) bzw. 5-Amino-1 -benzyl-pyrazol (1 c) die Pyrazolo[3.4-b]pyridine 6 h bzw. 6c. Die Substituentenverteilung in 6 wird durch Isolierung tind Cyclisierung der primiir entstehenden p-Amino-crotonsaureester 5 ermittelt, NMR-und IR-Spektren werden diskutiert. Die Struktur von 1 c, das uber sein 4-Athoxycarbonyl-Derivat (20b) gewonnen wird, folgt aus dem Vcrgleich der Kopplungskonstante J3,4 von l c mit denen strukturell festgelegter Verbindungen. Aus der Grone der Kopplungskonstante der CH-Protonen kann geschlossen werden, daB 3(5)-Amino-pyrazol vorwiegend in der Form 1 a existiert. Aus 1 a und Acetessigester entsteht 7-Hydroxy-5-methyl-pyrazolo[ 1.5-ulpyrimidin (12). Die Cyclisierung des aus Acetessigester und 1 a bzw. 5-Amino-1-p-toluolsulfonyl-pyrazol hergestellten 13-Amino-crotonsaureesters 5 a bzw. 10 ergibt ebenfalls 12 und kein 4-Hydroxy-6-methyl-1 H-pyrazolo[3.4-b]pyridin (6a). 6 a wird durch Debenzylierung von 6 c synthetisiert. 3, 4 und 7 sind Purin-Antagonisten. Im Pyrazolo[3.4-b]pyridin-System ist im Vergleich Zuni Purin ein N-Atom des Pyrimidinringes durch CH ersetzt und ein N-Atom des Imidazolringes um eine Stelle verschoben. Wir synthetisierten deshalb eine Reihe von Pyrazolo[3.4-b]pyridinen des Typs 3, 4, 6 und 7, von denen sich einige als beachtliche Purin-Antagonisten erwiesen. Man kann das Pyrazolo[3.4-b]pyridin-System ausgehend von einem substituierten Pyridinderivat auf bauen, z. B. aus 2-Chlor-nicotinsaurez) oder deren Ester31, aus 3-Cyan-pyridin-sulfonsaure-(2)4) bzw. aus 2-Chlor-3-cyan-pyridin5) und Hydrazin, wobei man Derivate mit einer OH-bzw. NHz-Gruppe in 3-Stellung erhalt.
