Tujuan dari penelitian ini yaitu untuk mengetahui pengaruh lama proses pencarian menggunakan (thawing) air dingin pada semen beku sapi Peranakan Ongole terhadap kualitas semen meliputi motilitas, viabilitas, abnormalitas dan total spermatozoa motil. Metode: Metode yang digunakan yaitu ekperimental laboratorium dengan menggunakan lima perlakuan dan sepuluh kali ulangan pada sampel semen beku sapi Peranakan Ongole. Data dianalisis menggunakan varian RAK dan dilanjutkan dengan DMRT, apabila terdapat perbedaan nyata. Dilakukan uji Chi Square pada motilitas individu dan total spermatozoa motil, untuk membandingkan dengan Standar Nasional Indonesia (SNI) kualitas semen beku. Penelitian ini dilakukan dengan lima perlakuan yaitu P0 dengan lama waktu thawing 30 detik dengan suhu 37 o C, sedangkan P1, P2, P3 dan P4 masing-masing menggunakan lama waktu thawing secara berurutan yaitu 15, 30, 45 dan 60 detik dengan suhu yang sama yaitu 28 o C. Hasil: Hasil thawing terbaik adalah P2 menghasilkan rataan motilitas individu 49,9±6,0 %, viabilitas 65,1±7,2%, abnormalitas 4,0±1,8% dan total spermatozoa motil 19,3±4,0 juta/straw. Hasil thawing terbaik adalah P2 menghasilkan rataan motilitas individu 49,9±6,0 %, viabilitas 65,1±7,2%, abnormalitas 4,0±1,8% dan total spermatozoa motil 19,3±4,0 juta/straw. Kesimpulan: Terdapat pengaruh lama dan suhu thawing pada kualitas semen beku sapi Peranakan Ongole terhadap kualitas semen.
Objective: The objective of this study was to determine the effect of adding various doses of green tea extract to the semen of Kacang goats during the sexing process on motility, viability, membrane integrity, malondialdehide (MDA), and deoxyribonucleic acid (DNA) fragmentation. Materials and Methods: It started with the containment of the semen of the Kacang goat, followed by macroscopic and microscopic examinations. If the semen was considered viable, a diluter that had been added with various doses of green tea extract would be added to the semen. After that, sexing was carried out using the percoll gradient density medium. Next, the sexed semen was cryopreserved in liquid nitrogen. Then, an examination of motility, viability, membrane integrity, MDA, and DNA fragmentation was conducted. Result: There was a significant difference between the control and treatment (p ≤ 0.05). The highest result was obtained in the treatment of adding 0.05 mg of green tea extract/100 ml of Andromed®. Conclusion: The addition of green tea extract can improve the quality of the sexed semen of the Kacang goat after it has been cryopreserved.
