This study aimed to investigate the in vitro antioxidant activity of the novel anticonvulsant levetiracetam and benzodiazepine clonazepam. To do this, the mice brain homogenates were incubated with levetiracetam (50, 100 or 200 g/ml) or clonazepam (50, 100 or 200 g/ml), and then, submitted to heating at 37°C for 1 h. Ascorbic acid (vitamin C, 200 g/ml) was used as reference antioxidant drug. The markers of oxidative stress, such as lipid peroxidation, nitrite-nitrate content, catalase activity, and reduced glutathione (GSH) levels, were measured in brain homogenates. The group submitted to the heating-induced oxidative stress showed an increase in lipid peroxidation, nitrite-nitrate content, and catalase activity. Previous incubation with levetiracetam and clonazepam, mainly at lower doses (50 and 100 g/ml), and similarly to vitamin C, prevented these pro-oxidative changes, reducing the lipid peroxidation, nitrite-nitrate contents and catalase activity, and increasing GSH levels. These findings demonstrate antioxidant properties of levetiracetam and clonazepam, and help to elucidate the role of protection against oxidative stress in the neuroprotective mechanism of antiepileptic drugs.
Introdução: A epilepsia é uma desordem cerebral proveniente de descargas elétricas anormais e recorrentes, que levam a convulsões. O abrasamento tem sido utilizado como modelo para estudar a epilepsia clínica, já que os animais submetidos a este modelo tem aumento progressivo da atividade convulsiva, através de aplicações repetidas de doses subconvulsivas de um agente químico, como o pentilenotetrazol (PTZ). O estresse oxidativo pode ser um fator causador da progressão das convulsões. O tratamento da epilepsia ainda é um desafio, devido à refratariedade em 20 a 30% dos pacientes. A liraglutida (LIRA) é um agonista do receptor de GLP-1 aprovado para o tratamento do Diabetes Mellitus tipo 2. GLP-1 é uma incretina e também um neuropeptídeo produzido por células neuronais. Existem evidências do efeito promissor dos agonistas de GLP-1 no tratamento dos transtornos neurológicos, portanto, decidimos investigar o efeito deste na epilepsia e o possível sinergismo com o anticonvulsivante levetiracetam (LEV). Objetivos: Avaliar o efeito da LIRA e da associação desta ao LEV no modelo de abrasamento induzido por PTZ através da determinação do estresse oxidativo no hipocampo (HC) dos animais. Métodos: Os camundongos swiss (25-34g) receberam PTZ (35mg/kg, i.p. n = 8, cada) em dias alternados, por 21 dias e foram pré-tratados com LEV 50 mg/kg e/ou LIRA (nas doses 75 μg/kg e 150μg/kg) ou salina, por via i.p. Após o último dia de administração, os animais foram sacrificados e o HC de cada animal foi dissecado e, então, foram realizados os testes para a determinação do estresse oxidativo (nitrito, MDA e GSH). Os dados foram analisados por ANOVA seguida por Tukey, com significância de p <0,05. O projeto foi aprovado pela Comissão de Ética em Pesquisa Animal (CEPA) da UFC sob o número 97/15.
