Das grossc Interesse, welches Acetyl-cholin als ,,Vagusstoff" besnsprucht, regte zu wrschiedenartigen chemischen "ariationen seiner Molekel an1). Diesc lassen sich in drei Gruppen einteilen: die Verintlerungen der Estergruppe A, tlcr Kohlenstoffkctte R untl der quartarm Animoniumgruppe C.c Rei tlcr letzten Art r o n Umw-andlungen kanicn Welch untl R o e p h 2 ) zu tlem interessanten Ergehnis, dztss man die Trimethyl-ammonium-Gruppe (lurch eine Trimethyl-phosphonium-oder Trimethyl-arsonium-Gruppe ersetzen kann, ohne die Wirkung des Acetyl-cholins als ,,Vagusstoff" qualitativ zu andern. Quantitativ wird die Wirkung durch die Vertauschung des Stickstoffs mit tien Elementen tlcrselben Gruppe des periodischen Systems ahgeschwacht, und zwar bei dem ,,Phospho"-cholin-chlorid im Verhaltnis r o n etwa 100 : 8 untl bei dcm ,,Ammo"-cholin-chloritl im Verhaltnis von etwa 100 : 1. Das spezifische Ferment tlw Acetyl-cholin-Spaltung im Organismus, die Cholin-Esterase, wirkt auf beide Verbindungen qualitativ unti quantitatir fast gleich wie auf Acetyl-cholin-chloritl, die schwachere pharmakologische Wirkung ist also nicht auf eine grossere Spaltungsgeschffintligkeit zuruckzufuhren. Wir haben im Rahmen eirier Reihe ron Arbeiten, die unternommen uurden, um den Stickstoff in verschietlenen biogenen Aminen durch Schwefcl zu ersetzen, such das bishcr unbekannte Schwefelanalogon tles Acetyl-cholin-chlorids, das D i m e t h y 1 -( @a c c t o x yat h y 1 )s u l f o n i u mc h 1 o r i tl CH,COO -CH,CH,* S(CH,),CI dargestellt (wir bezeichnen es in tlieser Arbeit abgekurzt als ASCh).Die Darstellung der empfintllichen Yerbintlung gelang durch schonende Acetylicrung des Dimethyl-( @-oxyathy1)-sulfonium-chlo-rids3) ntit dcetylchlorid.
