Based on simulation studies, it was shown that the type and size of experimental populations can exert an influence on the accuracy of genetic maps. A hypothetical genome map (one chromosome with nine equidistant molecular markers) was generated for the following population types: F 2 with dominant and co-dominant markers, backcrossing, recombinant inbred lines (RIL) and double-haploid. The population sizes were 50, 100, 150, 200, 500 and 1000 individuals and 100 simulations were made for each population. The inaccuracies of the populations with the lowest number of individuals were shown by inversions in the order of the markers and the establishment of more than one linkage group in up to 38% of the simulations, depending on the population type. Stress and variance values of the distances between adjacent markers were significantly reduced with the increased size of the population. More accurate maps were obtained for the co-dominant F 2 and RIL whereas the maps for the dominant F 2 population were less accurate. The higher the number of individuals, the more precise was the map. In all populations, a total of 200 individuals were considered as being sufficient for the construction of reasonably accurate genetic maps. Although this paper deals with plant populations this approach is equally applicable to other organisms.
O objetivo deste trabalho foi avaliar a produtividade e obter as estimativas de parâmetros genéticos e não genéticos de 40 materiais genéticos do programa de melhoramento genético de café Conilon do Incaper, no Estado do Espírito Santo. Foram analisados dados de dois experimentos, nos municípios de Marilândia e Sooretama, ES, nas safras de 1996, 1997, 1998, 1999, 2000, 2001 e 2002, em que se avaliaram 16 características. Realizou-se, inicialmente, a análise de variância individual, por local em cada ano, com base na média de parcelas, em blocos ao acaso. Posteriormente foi feita a análise de variância conjunta. Os genótipos apresentaram grande variabilidade genética para a maioria das características avaliadas. Os elevados coeficientes de determinação genotípico e coeficientes de variação genéticos, associados às altas produtividades e à variabilidade genética indicam a possibilidade de obtenção de êxitos em programas de melhoramento genético para diferentes características avaliadas nos dois municípios.
Molecular DNA markers are used for detection of polymorphisms in individuals. As they are independent of developmental stage of the plant and environmental influences, they can be useful tools in taxonomy. The alleles of simple sequence repeat (SSR) markers (or microsatellites) are traditionally used to identify taxonomic units. This application demands the laborious and costly delimitation of exclusive alleles in order to avoid homoplasy. Here, we propose a method for identification of species based on the amplification profile of groups of SSR markers obtained by a transferability study. The approach considers that the SSR are conserved among related species. In this context, using Psidium as a model, 141 SSR markers developed for Psidium guajava were transferred to 13 indigenous species of Psidium from the Atlantic Rainforest. Transferability of the markers was high and 28 SSR were conserved in all species. Four SSR groups were defined and they can help in the identification of all 13 Psidium species studied. A group of 31 SSR was genotyped, with one to six alleles each. The H0 varied from 0.0 to 0.46, and PIC from 0.0 to 0.74. Cluster analysis revealed shared alleles among species. The high percentage of SSR transferability found in Psidium evidences the narrow phylogenetic relationship existing among these species since transferability occurs by the preservation of the microsatellites and anchoring regions. The proposed method was useful for distinguishing the species of Psidium, being useful in taxonomic studies.
Knowledge of the genetic variability of a population is essential to guide its preservation and maintenance in addition to increasing the efficiency of genetic breeding programs. On this basis, this study was conducted to evaluate the genetic diversity of Coffea canephora genotypes using multivariate statistical procedures applied to a set of morpho-agronomic variables. The materials employed in this study constitute a crop located in Vila Valério - ES, Brazil, where the genotypes are arranged in a randomized-blocks experimental design with four replicates. Significant differences were detected by the F test at the 1% or 5% probability levels among the genotypes for all evaluated traits, demonstrating heterogeneity of genetic constitution in the studied population, which is favorable to breeding, as it indicates the possibility to identify superior and divergent individuals. Based on the generalized Mahalanobis distance, the most divergent combinations were obtained between genotypes 23 and 10 (256.43) and 23 and 17 (250.09). The clusters formed by Tocher's optimization and the UPGMA hierarchical method agreed, both similarly grouping the genotypes into three clusters. Of the analyzed traits, mature fruit weight (19.08%), yield (15.50%), plant diameter (12.42%), and orthotropic-shoot internode length (10.94%) were the most efficient to explain the dissimilarity among the genotypes.
Resumo -O objetivo deste trabalho foi avaliar uma metodologia de análise de adaptabilidade e estabilidade fenotípica de genótipos de café baseada em regressão não paramétrica. A técnica utilizada difere das demais, pois reduz a infl uência na estimação do parâmetro de adaptabilidade de algum ponto extremo, ocasionado pela presença de genótipos com respostas demasiadamente diferenciadas a determinado ambiente. Foram utilizados dados provenientes de um experimento sobre produtividade média de grãos de 40 genótipos de café (Coffea canephora), com delineamento em blocos ao acaso, com seis repetições. Os genótipos foram avaliados em cinco anos (1996, 1998, 1999, 2000 e 2001), em dois locais (Sooretama e Marilândia, ES) no total de dez ambientes. Adaptability and stability based on nonparametric regression in coffee genotypesAbstract -The objective of this work was to evaluate a methodology of phenotypic adaptability and stability analyses of coffee genotypes based on nonparametric regression. The technique used differs from other techniques because it reduces the infl uence of extreme points resulting from the presence of genotypes whose answers to a certain environment are too different on the estimation of the adaptability parameter. Data from an experiment studying the average yield of 40 coffee (Coffea canephora) genotypes in a randomized block design with six replicates were used to evaluate the method. The genotypes were evaluated along fi ve years (1996, 1998, 1999, 2000 and 2001) in two locations (Sooretama and Marilândia, ES, Brazil), in a total of ten environments. The methodology proposed proved adequate and effi cient, since it eliminates the disproportionate effects induced by the presence of extreme points and avoids misleading recommendations of genotypes in terms of adaptability.
