The excision of hydrogen from an aliphatic carbon chain to produce an isolated olefin (desaturation) without over-oxidation is one of the most impressive and powerful biosynthetic transformations for which there are no simple and mild laboratory substitutes. The versatility of olefins and the range of reactions they undergo is unsurpassed in functional group space. Thus, the conversion of a relatively inert aliphatic system to its unsaturated counterpart can open new possibilities in retrosynthesis. In this article, the invention of a directing group to achieve such a transformation under mild, operationally simple, metal-free conditions is outlined. This “portable desaturase” (TzoCl) is a bench-stable, commercial entity (Aldrich, cat # L510092) that is facile to install on alcohol and amine functionalities to ultimately effect remote desaturation, while leaving behind a synthetically useful tosyl group.

Taxanes are a large family of terpenes comprising over 350 members, the most famous of which is Taxol (paclitaxel) — a billion-dollar anticancer drug. Here, we describe the first practical and scalable synthetic entry to these natural products via a concise preparation of (+)-taxa-4(5),11(12)-dien-2-one, which possesses a suitable functional handle to access more oxidised members of its family. This route enabled a gram-scale preparation of the ”parent” taxane, taxadiene, representing the largest quantity of this naturally occurring terpene ever isolated or prepared in pure form. The taxane family’s characteristic 6-8-6 tricyclic system containing a bridgehead alkene is forged via a vicinal difunctionalisation/Diels–Alder strategy. Asymmetry is introduced by means of an enantioselective conjugate addition that forms an all-carbon quaternary centre, from which all other stereocentres are fixed via substrate control. This study lays a critical foundation for a planned access to minimally oxidised taxane analogs and a scalable laboratory preparation of Taxol itself.

The first successful effort to replicate the beginning of the Taxol oxidase phase in the laboratory is reported, culminating in the total synthesis of taxuyunnanine D, itself a natural product. Through a combination of computational modeling, reagent screening, and oxidation sequence analysis, the first three of eight C–H oxidations (at the allylic sites corresponding to C-5, C-10, and C-13) required to reach Taxol from taxadiene were accomplished. This work lays a foundation for an eventual total synthesis of Taxol capable of delivering not only the natural product but also analogs inaccessible via bioengineering.

Asymmetric cyclopropane synthesis currently requires bespoke strategies, methods, substrates, and reagents, even when targeting similar compounds. This approach slows down discovery and limits available chemical space. Introduced herein is a practical and versatile diazocompound and its performance in the first unified asymmetric synthesis of functionalized cyclopropanes. The redox‐active leaving group in this reagent enhances the reactivity and selectivity of geminal carbene transfer. This effect allowed the asymmetric cyclopropanation of various olefins, including unfunctionalized aliphatic alkenes, that enables the three‐step total synthesis of (−)‐dictyopterene A. This unified synthetic approach delivers high enantioselectivities that are independent of the stereoelectronic properties of the functional groups transferred. Our results demonstrate that orthogonally differentiated diazocompounds are viable and advantageous equivalents of single‐carbon chirons.

Abstract. There has been a lengthy debate on whether the abundance of adult reef fishes depends on prerecruitment or postrecruitment processes; however, we still do not have the ability to predict the magnitude of local fish recruitment. Here we show that the success of the leopard grouper (Mycteroperca rosacea) recruitment in the Gulf of California, Mexico, is determined by the availability of nursery habitat, which in turn is strongly correlated to climate conditions. Observational and experimental studies showed that leopard grouper larvae recruit preferentially on shallow rocky bottoms with brown algal (Sargassum spp.) beds, and that abundance of recruits is determined by the availability of Sargassum. The biomass of Sargassum decreases linearly with an increase in the Multivariate El Nin˜o Southern Oscillation (ENSO) Index (MEI; an index positively correlated with water temperature and negatively correlated with nutrient availability). We analyzed the relationship between the interannual variation of MEI and the recruitment of the leopard grouper using field estimates of abundance of juvenile groupers. Our results show that there is a nonlinear relationship between recruitment and the oceanographic climate, in that the density of recruits decreases exponentially with increasing MEI. The predictability of leopard grouper recruitment has important implications for fisheries management, since it could allow adaptive management without expensive stock assessment programs.
