Обоснование. Известно, что механизм цитотоксического действия противоопухолевого средства-каталитической системы «терафтал + аскорбиновая кислота» (ТФ+АК)-связан с образованием в клетке активных форм кислорода (ROS) и индукцией в ней окислительного стресса. Система «гемоксигеназа-1/ ферритин» (HО-1/Ft) защищает клетки от окислительного стресса. Цель. Исследовать значение системы HО-1/Ft в защите лейкозных клеток от токсического воздействия противоопухолевого средства ТФ+АК. Материалы и методы. В работе использовались лейкозные клетки человека линий K562 и U937. Базальную и препарат-индуцированную экспрессию HО-1/ Ft на уровне мРНК и белка изучали методами ОТ-ПЦР в реальном времени и вестерн-блоттинга, концентрацию ROS в клетках-методом проточной цитометрии, цитотоксичность препаратов-методом МТТ. Результаты. По нашим данным, в миеломонобластах линии U937 присутствует конститутивно-активная НО-1, а в эритробластах линии K562 экспрессия этого белка заблокирована на уровне мРНК. Гемин, агонист НО-1, индуцирует в клетках U937 коэкспрессию HО-1 и Ft на уровне мРНК и белка. Активация системы HО-1/Ft гемином в клетках U937 не влияет на чувствительность их к ТФ+АК и увеличивает чувствительность, например, к цитарабину в 2 раза. Установлено, что система ТФ+АК вызывает up-регуляцию генов HО-1/Ft, экспрессия которых увеличивается в 4 и 1,5 раза соответственно по сравнению с базальным уровнем. Предынкубация миеломонобластов U937 с хелатором железа дефероксамином приводит к увеличению их устойчивости к ТФ+АК в 2 раза. В то же время в присутствии железосодержащего аналога ТФ цитотоксичность этой системы увеличивается в 2 раза. Заключение. В лейкозных клетках линии U937 с конститутивно-активной системой НО-1/Ft гем-зависимый путь ее активации не играет существенной роли в за
