A comprehensive treatment of light propagation through intact leaves based on the theories of radiative transfer and absorption statistics was used to calculate the theoretical absorption spectra of the chlorophyll-containing particles under conditions of multiple scattering and pigment spatial distribution equivalent to those in a leaf. These spectra were compared with the experimental in vivo spectra of leaves and in vitro spectra of chlorophyll-protein complexes extracted form these leaves. We conclude that the main discrepancies between the in vivo and in vitro spectra are apparently due to the optical artifacts specific for light propagation in leaves-multiple scattering and distributional error. Alterations of the pigment properties upon extraction significantly contribute to these discrepancies. The method has an estimated accuracy of about 10% and can be applied to derive the intrinsic optical properties of the photosynthetic mechanism in a leaf, as well as for the systematic study of their changes in the course of light adaptation.

UDP-glucose: coniferyl alcohol glucosyltransferase was detected in hypocotyls of spruce (Picea abies (L.) Karst.) seedlings. Enzyme activity rose after germination to maximal activity on about the 10th day and then rapidly declined. An antiserum against the glucosyltransferase isolated from cambial sap of spruce (Schmid and Grisebach, 1982, Eur. J. Biochem. 123, 363-370) was employed for localization of the enzyme in cross sections of hypocotyls from 10-d-old spruce seedlings, using the immunofluorescent technique. The results show that the transferase is located predominantly in the epidermal and subepidermal layers and in the vascular bundles. Intracellularly, the enzyme is located in the parietal cytoplasmic layer. The results corroborate the assumption that coniferin (coniferyl alcohol β-D-glucoside) participates in lignification of spruce.

Phytomorphology -if concerned with development -often concentrates on correlative changes of form and neglects the aspects of age, time and clock, although the plant's spatial and temporal organisation are intimately interconnected. Common age as measured in physical time by a physical process is compared to biological age as measured by a biological clock based on a biological process. A typical example for a biological clock on the organ level is, for example, a shoot.
