To the purposeful search for the DHFR-inhibitors among substituted pteridine-2,4,7triones and 7-aryl-(hetaryl-)furo[3,2-g]pteridine-2,4(1H,3H)-diones for further biological research. Methods. In vitro methods, molecular docking, SAR-analysis, statistical methods. Results. The DHFR-inhibitory activity of substituted 1-methylpteridine-2,4,7-triones (2, 3, 4) and 7-aryl-(hetaryl-)furo[3,2-g]pteridine-2,4(1H,3H)-diones (5, 6) was studied. It was estab- 6) inhibited DHFR by 14.59-52.11 %, and were less active comparing to methotrexate. It was found that the introduction of aryl moiety with electron-accepting group, naphthyl substituent or electron-accepting heterocycle (furan, thiophene and benzofuran) caused an increase in the DHFR-inhibitory activity. Additionally, it was shown, that annulation of the furan cycle to the pteridine system was reasonable in the scope of new DHFR-inhibitors synthesis. Thereby it may be concluded that the calculated values of affinity are not reliable predictors for the DHFR-inhibiting activity of studied compound. However, the molecular docking study may be used for evaluation of the interactions between the studied inhibitor and active center of DHFR. Conclusions. The conducted primary in vitro screening revealed low or moderate DHFR-inhibiting activity of the synthesized compounds. The visualization of molecular docking showed that despite the structural similarity to methotrexate, the obtained compounds form different ligand-enzyme interactions. The calculated values of affinity cannot be used as predictors of DHFR-inhibiting activity because of the absence of correlation between the abovementioned indicators. The obtained compounds may be of interest for further studies aimed at the search for anti-inflammatory, anti-viral, hypoglycemic, hypotensive, anti-ische mic agents due to the expected low-toxicity associated with the slight DHFR-inhibiting activity.
The present article is devoted to searching for biologically active agents among novel thio-containing pteridines. Synthetic protocols based on the condensation of 5,6diamino-2-thioxo-2,3-dihydropyrimidin-4(1H)-ones with dicarbonyl compounds were elaborated and used for the synthesis of target products. The directions for further modification of the obtained thio-containing pteridines were substantiated and realized. The spectral properties of the obtained compounds were studied and described. The results of the in silico study revealed that the predicted affinity of the obtained compounds to the dihydrofolate reductase (DHFR) active site is comparable with the affinity of methotrexate, despite the differences in the nature of the ligand-enzyme interactions. The in vitro study of DHFR-inhibiting activity revealed that the most active compounds 3.9 and 4.2 have lg IC 50 values of −5.889 and −5.233, respectively, significantly inferior to methotrexate (lg IC 50 = −7.605).Additionally, the synthesized compounds were studied for their antiradical activity as a possible mechanism of pharmacological effects. Among the obtained pteridines, compounds 5.1 (lg EC 50 = −4.82) and 5.3 (lg EC 50 = −4.92) have antiradical activity higher than the reference compound ascorbic acid (lg EC 50 = −4.81). The conducted structure-activity relationship analysis provided valuable data for the further search for biologically active agents among thio-containing pteridines and related compounds.
Search for new antibacterial agents with dihydrofolate reductase-inhibitory activity among N-(R-carbamothiol)cycloalkylcarboxamides using in silico and in vitro methodology, SAR analysis to optimize the synthesis of new potential antinicrobials. Methods. Molecular docking, in vitro DHFR inhibition assay, antimicrobial evaluation, SAR analysis, statistical methods. Results. According to the results of molecular docking to the active center of dihydrofolate reductase (DHFR), namely affinity, the main types of interactions and arrangement in the active center of the enzyme, several N-(R-carbamothioyl)cycloalkylcarboxamides were selected for their inhibitory effect. Based on in vitro screening, few promising compounds with high ability to inhibit DHFR were identified. It was found, that diacylsemicarbazides are more effective inhibitors of DHFR compared to acylthioureas. The studies on antibacterial activity have revealed several promising compounds, namely N-(2-R-hydrazine-1-carbonothioyl)cycloalkanecarboxamides, as highly active antimicrobial agents against E. сoli and St. aureus (MIC 3.125-25.0 μg/ml) with high DHFR-inhibitory effect, the activity of which competes with the comparison drug "Nitrofurazone". This justifies the continuation of systematic research in this direction. Conclusions. A well-founded search among N-(R-carbamothiol)cycloalkylcarboxamides for new antibacterial agents with dihydrofolate reductase-inhibitory activity, using in silico and in vitro methodology, established relationship between the chemical structure and activity aimed at further design of new potential drug agents.

Домінуючими амінокислотами в усіх зразках були аланін, аргінін, аспарагінова кислота, валін, гліцин та лізин.Ключові слова: портулак городній, портулак великоквітковий, амінокислоти.Вступ. Відомо, що амінокислоти відіграють важ-ливу роль у синтезі ферментів, гормонів, медіаторів, складових сполучної та м'язової тканини, в проце-сах кровотворення, входять до складу коферментів, жовчних кислот й антибіотиків. Амінокислоти вико-ристовують в медицині для лікування захворювань серцево-судинної, нервової, травної системи, для зміцнення імунної системи, нормального функціону-вання ендокринних залоз, з метою профілактики ате-росклерозу, пригнічення розвитку злоякісних новоут-ворень [1,2,3,5]. Крім того, амінокислоти є однією з основних груп біологічно активних речовин більшості рослин та є необхідними для синтезу ферментів, ві-тамінів, алкалоїдів, пігментів, флавоноїдів, поліфе-нольних та стероїдних сполук [6].Метою дослідження було вивчення якісного складу та кількісного вмісту амінокислот в листі, траві, коре-нях портулаку городнього Portulaca oleracea L. та пор-тулаку великоквіткового Portulaca grandi ora Hook.Методи дослідження. Об'єктами дослідження були листя, трава, корені портулаку городнього та портулаку великоквіткового, заготовлені в 2015-2016 рр. в смт. Томаківка Дніпропетровської області.Визначення якісного складу та кількісного вмісту амі-нокислот у досліджуваній сировині проводили методом іонообмінної хроматографії за допомогою автоматично-го амінокислотного аналізатора ААА Т-339М (Чехія).Близько 100 мг (точна наважка) здрібненої сиро-вини поміщали в пробірку для гідролізу, додавали рівну кількість води дистильованої та концентрованої кислоти хлористоводневої, ретельно перемішували, закривали герметично та залишали в термостаті протягом доби при температурі 120 °С. Після чого пробірку розкривали, відганяли надлишок кислоти хлористоводневої та нейтралізували над натрієм гід-роксидом в ексикаторі. Отриману пробу розчиняли в буферному розчині, фільтрували, встановлювали значення рН 2,2 та вводили в кількості 50 мкл до амі-нокислотного аналізатора. Калібрування аналізатора здійснювали шляхом пропускання стандартної сумі-ші амінокислот через катіоніт.Результати й обговорення. Результати визна-чення якісного складу та кількісного вмісту амінокис-лот у листі, траві, коренях портулаку городнього та портулаку великоквіткового представлені в таблиці 1.За даними дослідження, які наведені в таблиці 1, було ідентифіковано в листі, траві, коренях портула-ку городнього та портулаку великоквіткового 17 амі-нокислот, з яких 9 є незамінними.Найбільший вміст суми амінокислот було визначе-но в листі портулаку городнього (24,35 мг/100 мг), а найменший вміст суми амінокислот -у коренях пор-тулаку великоквіткового (11,97 мг/100 мг).Встановлено, що в листі та траві обох досліджува-них рослин домінуючими компонентами амінокислот-ного складу були г...
Nowadays the diabetes mellitus has epidemic character. That’s why searching and creating of new medicines for effective therapy of this pathology are actual. Phytotherapy is one of the components in the complex treatment of diabetes mellitus type 2. It combines a wide range of pharmacological actions, fewer side effects and toxic reactions on the human body than synthetic drugs. Plants of genus of Portulaca L. can be perspective sources for searching new medicinal plants. Purslane (Portulaca oleracea L.) is used in folk medicine as hypoglycemic agent. The aim of the research was to study the hypoglycemic activity of the infusion of Portulaca oleracea herb. Simulation of insulin resistance was performed with intramuscular injections of glucocorticoids (Dexamethasone, 0.125 mg/kg). Portulaca oleracea infusion administered orally at the dose 10 ml/kg. Gliclazide freshly suspension was used as a reference drug and it was administered orally by 50 mg/kg. Characteristic of glucose homeostasis was carried out using a short insulin test, oral glucose tolerance test. Also assessed the effect of Portulaca oleracea infusion and reference drug on adrenaline hyperglycemia. Glucose level at 30 minutes after administration of insulin in the group that received Portulaca oleracea infusion reduced on 53.9%. There was a decrease on 37.5% in the group of diabetic control, and in the intact group – on 60.2%. The result of oral test of glucose tolerance showed that the area under the glycemic curve in groups that received Portulaca oleracea infusion (314.9 mmol/L·min) and reference drug Gliclazide (376.0 mmol/L·min) did not significantly differ from the similar area of the intact group (372.7 mmol/L·min). The administration of the epinephrine hydrochloride solution caused a sharp and significant increase in the glucose level in the diabetic control group. 90 minutes after the epinephrine injection the glucose level in animals which received Portulaca oleracea infusion increased in 3.15 times, and in the reference-group – in 1.97 times, which significantly differs from the diabetic control group. The obtained results indicate that the infusion of Portulaca oleracea herb reduce the formation of insulin resistance and glucose tolerance in the conditions of experimental dexamethasone diabetes mellitus in rats.
