Acupuncture appears to be equivalent to drug therapy in these patients. It is a safe, effective and durable treatment for vasomotor symptoms secondary to long-term antiestrogen hormone use in patients with breast cancer.
Resveratrol (trans-3,4V,5-trihydroxystilbene) is a naturally occurring polyphenolic compound highly enriched in grapes, peanuts, red wine, and a variety of food sources. Resveratrol has antiinflammatory and antioxidant properties, and also has potent anticancer properties. Human glioma U251 cells were used to understand the molecular mechanisms by which resveratrol acts as an anticancer agent, since glioma is a particularly difficult cancer to treat and eradicate. Our data show that resveratrol induces dose-and time-dependent death of U251 cells, as measured by lactate dehydrogenase release and internucleosomal DNA fragmentation assays. Resveratrol induces activation of caspase-3 and increases the cleavage of the downstream caspase substrate, poly(ADP-ribose) polymerase. Resveratrol-induced DNA fragmentation can be completely blocked by either a general caspase inhibitor (Z-VAD-FMK) or a selective caspase-3 inhibitor (Z-DEVD-FMK), but not by a selective caspase-1 inhibitor. Resveratrol induces cytochrome c release from mitochondria to the cytoplasm and activation of caspase-9. Resveratrol also increases expression of proapoptotic Bax and its translocation to the mitochondria. Resveratrol inhibits U251 proliferation, as measured by MTS assay [3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt], and induces G 0 /G 1 growth arrest, as determined by flow cytometry. The cyclin-dependent kinase inhibitor, olomoucine, prevents cell cycle progression and resveratrol-induced apoptosis. These results suggest that multiple signaling pathways may underlie the apoptotic death of U251 glioma induced by resveratrol, which warrants further exploration as an anticancer agent in human glioma. The RTs studied are not useful for screening for NG at the point of care. In case of CT a recommendation about their use in routine care should be supported by a cost-effectiveness analysis. In screening populations at high risk of sexually transmitted infections or pregnant women, the RTs for syphilis should be used.
Problems with immunosuppression and graft survival limit clinical applications of neurotransplantation protocols for neurodegenerative disease. Sertoli cells, testes-derived cells with immunosuppressive and trophic properties, may serve as an alternative cell source for transplantation. Sertoli cells were transplanted into the striatum of rats following two injections of 3-nitropropionic acid (3-NP) to determine whether they could ameliorate abnormalities in a model of early stage Huntington's disease. 3-NP-induced locomotor hyperactivity was significantly reduced in rats receiving Sertoli transplants compared to controls, with some behaviors returning to baseline. Sertoli cells survived in the striatum without systemic immunosuppression and some formed tubule-like structures. These results show that Sertoli transplants are able to ameliorate locomotor abnormalities in a 3-NP model of early HD. Thus, Sertoli cells should be further evaluated as a possible treatment strategy for the early stages of Huntington's disease.
Alzheimer's disease dementia (AD dementia) is one of the most common neurodegenerative diseases worldwide, with a growing incidence during the last decades. Clinical diagnosis of cognitive impairment and presence of AD biomarkers have become important issues for early and adequate treatment. We performed a systematic literature search and quality appraisal of AD dementia guidelines, published between 2005 and 2011, which contained diagnostic recommendations on AD dementia. We also analyzed diagnostic recommendations related to the use of brief cognitive tests, neuropsychological evaluation, and AD biomarkers. Of the 537 retrieved references, 15 met the selection criteria. We found that Appraisal of Guidelines Research and Evaluation (AGREE)-II domains such as applicability and editorial independence had the lowest scores. The wide variability on assessment of quality of evidence and strength of recommendations were the main concerns identified regarding diagnostic testing. Although the appropriate methodology for clinical practice guideline development is well known, the quality of diagnostic AD dementia guidelines can be significantly improved.
Ozone exposure induces increased production of free radicals which may result in oxidative stress. The objectives of this study were to determine the antioxidant effects of vitamin E on memory deficits and lipid peroxidation due to oxidative stress caused by acute ozone exposure. Rats were exposed to 0.7 p.p.m. ozone for 4 h and 50 mg/kg vitamin E was administered either before or after exposure. Experiment 1 evaluated alterations in short-term and long-term memory in a passive avoidance task. Experiment 2 quantified lipid peroxidation levels of the striatum, hippocampus and frontal cortex. Vitamin E administered before or after ozone exposure blocked memory deterioration and increases in lipid peroxidation levels associated with oxidative stress.
Introducción. En los últimos decenios, la síntesis de compuestos químicos ha producido un alto número de sustancias utilizadas para proteger los cultivos y las cosechas de las plagas. La mayoría de pesticidas han sido usados en grandes cantidades para fines agrícolas y la exposición tóxica a estos compuestos es un problema de gran envergadura para la toxicología, pues tiene impacto en la salud pública por su importante morbilidad y discapacidad. Así, las intoxicaciones agudas y crónicas pueden ser comunes entre trabajadores agrícolas y población general. Estos compuestos desencadenan mecanismos moleculares relacionados con la peroxidación lipídica, donde las especies reactivas de oxigeno pueden incrementar procesos oxidativos intracelulares.Objetivo. Realizar una revisión de la literatura científica relacionada con enfermedades por estrés oxidativo producido por pesticidas.Materiales y métodos. Revisión de artículos que utilizó las palabras clave Estrés oxidativo; Pesticidas; Peroxidación de lípido; Toxicidad. Se eligieron los artículos relacionados con las enfermedades por estrés oxidativo producido por pesticidas.Resultados. Se encontró que algunas enfermedades hematológicas, neurológicas, metabólicas y genotóxicas pueden estar relacionadas con la peroxidación lipídica que producen los pesticidas.Conclusión. Se presenta una revisión de la literatura científica sobre el estrés oxidativo, la peroxidación lipídica inducida por pesticidas y las diferentes enfermedades que pueden afectar a la población en general.
