γ-Tubulin controls neuronal microtubule polarity independently of Golgi outposts

Abstract: Microtubule orientation controls polarized trafficking in neurons. In this work, γ-tubulin is identified as a key regulator of both axonal and dendritic microtubule polarity. In addition, the idea that γ-tubulin works in dendrites by residing at Golgi outposts is tested.

“…7; Movies 1 and 2). γ-tubulin37C was selected as a control to target with RNAi as it is a γ-tubulin expressed maternally that is not present in somatic tissues including neurons (Wiese, 2008); γ-tubulin23C is the somatic replacement that nucleates microtubules in neurons (Nguyen et al, 2014).…”

“…52 However, no increase in g-tubulin has been observed at either somatic or dendritic Golgi, 50 and removing Golgi outposts from dendrites using activated kinesin constructs did not disrupt g-tubulin localization. 55 Thus the microtubule nucleation machinery associated with Golgi outposts may not function through the same g-tubulin recruitment mechanism as the PCM. A recent model proposes that Golgi drive microtubule nucleation by localizing the accessory factors that increase the nucleation activity of g-TuRC, while g-TuRC itself remains unenriched in the adjacent cytoplasm.…”
