Highly porous hosting materials with conducting (favorable to electron transfer) and magnetic (favorable to product separation) bicontinuous networks should possess great potentials for immobilization of various enzymes in the field of biocatalytic engineering, but the synthesis of such materials is still a great challenge. Herein, bifunctional graphene/γ-Fe2 O3 hybrid aerogels with quite low density (30-65 mg cm(-3) ), large specific surface area (270-414 m(2) g(-1) ), high electrical conductivity (0.5-5 × 10(-2) S m(-1) ), and superior saturation magnetization (23-54 emu g(-1) ) are fabricated. Single networks of either graphene aerogels or γ-Fe2 O3 aerogels are obtained by etching of the hybrid aerogels with acid solution or calcining of the hybrid aerogels in air, indicative of the double networks of the as-synthesized graphene/γ-Fe2 O3 hybrid aerogels for the first time. The resulting bifunctional aerogels are used to immobilize β-glucuronidase for biocatalytic transformation of glycyrrhizin into glycyrrhetinic acid monoglucuronide or glycyrrhetinic acid, with high biocatalytic activity and definite repeatability.

Due to its weak polarity as it lacks glycosyl, glycyrrhetinic acid is easily transported and absorbed in the blood, so its physiological activity is much stronger than that of glycyrrhizic acid.…”

Saponins are a class of glycosides whose aglycones can be either triterpenes or helical spirostanes. It is commonly recognized that these active ingredients are widely found in various kinds of advanced plants. Rare saponins, a special type of the saponins class, are able to enhance bidirectional immune regulation and memory, and have anti-lipid oxidation, anticancer, and antifatigue capabilities, but they are infrequent in nature. Moreover, the in vivo absorption rate of saponins is exceedingly low, which restricts their functions. Under such circumstances, the biotransformation of these ingredients from normal saponins—which are not be easily adsorbed by human bodies—is preferred nowadays. This process has multiple advantages, including strong specificity, mild conditions, and fewer byproducts. In this paper, the biotransformation of natural saponins—such as ginsenoside, gypenoside, glycyrrhizin, saikosaponin, dioscin, timosaponin, astragaloside and ardipusilloside—through microorganisms (Aspergillus sp., lactic acid bacteria, bacilli, and intestinal microbes) will be reviewed and prospected.

The purification of glycosylated recombinant b-glucuronidase from Pichia pastoris GS115 (PGUS-P) was investigated by a novel two-step process: ammonium sulfate precipitation and molecular sieve chromatography. The highest purification fold obtained was 66.79. The catalytic properties of glycosylated PGUS-P in hydrophobic ionic liquids (ILs)/buffer biphasic system were investigated. A 2.2-fold enhancement in the catalytic efficiency was observed using 50% (v/v) 1-butyl-3-methylimidazolium hexafluorophosphate, in comparison with the acetate buffer medium. When compared with the glycosylated PGUS-P, the deglycosylated enzyme at T 55 and T 65 exhibited low activity and low thermal stability in both ILs and acetate buffer. It was also observed that ILs had effect on the pH profile of deglycosylated PGUS-P -the optimum pH was extended from 5.0 (acetate buffer) to 5.0-7.0 (ILs). Therefore, this study indicates that the glycosylation of PGUS-P plays an important role in both catalytic activity and stability in hydrophobic ionic liquids (ILs)/buffer biphasic system.
