Xanthogranulomatous Pyelonephritis is Associated with Higher Tissue Expression of Monocyte Chemotactic Protein-1

Abstract: MCP-1 would seem to play a key role in the pathogenesis of XGP by mediating the recruitment of circulating monocytes or by cells resident in the interstitial space.

“…MCP-1, a key chemokine, initiates chemotaxis due to which migration and accumulation of macrophages occurs at the site. [ 35 ] Other inflammatory cells including lymphocytes and plasma cells also accompany the macrophages. It has been documented in orbital xanthogranulomatous disease that TIMP signaling pathway is activated in macrophages by stimulating the phosphorylation of JAK2 and STAT3.…”

“…Indeed, cholesterol is a known activator of the danger sensing structure, NLRP3 inflammasome, in macrophages, which orchestrates production of certain inflammatory cytokines [24]. In xanthogranulomatous pyelonephritis, increased monocyte chemotactic protein-1 expression has been suggested to play an important role in its pathogenesis [25]. A monocyte/macrophage typically transforms into foamy macrophage under the stimulus of IL-6, TNF-alpha and M-CSF [26].…”

“…Данные литературы о роли ЕGF при заболеваниях почек противоречивы. Большинство из них позволяют сделать вывод о значительной роли EGF, синтезируемого тубулярными элементами, в патогенезе тубулоинтерстициального повреждения [41][42][43][44]. Активация EGFR с повышением экспрессии EGF способствует развитию и прогрессированию заболеваний почек в различных экспериментальных моделях на животных.…”
