Wooden Cultural Heritage (WCH) represents a significant portion of the world’s historical and artistic heritage, consisting of immovable and movable artefacts. Despite the expertise developed since ancient times to enhance its durability, wooden artefacts are inevitably prone to degradation. Fungi play a pivotal role in the deterioration of WCH in terrestrial ecosystems, accelerating its decay and leading to alterations in color and strength. Reviewing the literature of the last 25 years, we aimed to provide a comprehensive overview of fungal diversity affecting WCH, the biochemical processes involved in wood decay, and the diagnostic tools available for fungal identification and damage evaluation. Climatic conditions influence the occurrence of fungal species in threatened WCH, characterized by a prevalence of wood-rot fungi (e.g., Serpula lacrymans, Coniophora puteana) in architectural heritage in temperate and continental climates and Ascomycota in indoor and harsh environments. More efforts are needed to address the knowledge fragmentation concerning biodiversity, the biology of the fungi involved, and succession in the degradative process, which is frequently centered solely on the main actors. Multidisciplinary collaboration among engineers, restorers, and life sciences scientists is vital for tackling the challenges posed by climate change with increased awareness. Traditional microbiology and culture collections are fundamental in laying solid foundations for a more comprehensive interpretation of big data.

This paper investigates the potential of clove (Eugenia caryophyllata) essential oil (C-EO) as a wood protection system for use in cultural heritage conservation. The preventive and curative antifungal efficacy of C-EO was compared to a boron containing wood preservative (Diffusit S) in two laboratory tests on European beech (Fagus sylvatica) and Scots pine sapwood (Pinus sylvestris) mini-blocks. Non-sterile, air-infected wood samples were employed in the first test, while wood samples with active attack, as result of inoculation with three fungal strains isolated from cultural heritage sites, in the second test. Untreated controls and C-EO-treated wood blocks were incubated up to 150 days to monitor and quantify fungal growth. The experimental results showed antifungal efficacy of C-EO to be higher than that of the boron wood preservative at the concentrations used. Concentrations of C-EO 5 % -10 % in ethyl alcohol prevented mould growth for at least 150 days, while a concentration of 10 % delayed or suppressed active attack by decay fungi. This effect, determined up to 90 days of incubation, was dependant on the type of fungus.

Nitrites and nitrates act as curing agents in meat products. Traditionally, the determination of these additives is performed via colorimetric spectrophotometry, an extremely labor intensive method. Therefore, the objective was to develop and validate a methodology for sodium nitrite and nitrate determination in sausage through ion chromatography, a technique of high sensitivity and precision. Validation parameters of the method analized were linearity, accuracy, precision and limits of detection and quantification. The linear working range of the method was 0.1-2000 mg kg-1 and 0.5-10000 mg kg-1 , the mean recovery was 105.5% and 100.8%, detection limit 13 and 8,1 mg kg-1 , quantification limit 32 and 23 mg kg-1 and variation coefficient 4.8 and 2.6 % for nitrite and nitrate, respectively. The "r" correlation coefficients were above 0.995. The developed method proved to be efficient for the analysis of nitrite and nitrate in sausage. Tradicionalmente, a determinação destes aditivos é realizada via colorimétrica por espectrofotometria, método extremamente trabalhoso. Por isso, objetivou-se desenvolver e validar uma metodologia para determinação de nitrito e nitrato de sódio em salsicha através da cromatografia de íons, uma técnica de alta sensibilidade e precisão. Os parâmetros de validação do método desenvolvido foram linearidade, exatidão, precisão e limites de detecção e de quantificação. A faixa linear de trabalho do método foi de 0,10-2000 mg kg-1 e de 0,50-10000 mg kg-1 , recuperação média foi de 105,5% e 100,8%, limite de detecção 13 e 8,1 mg kg-1 , limite de quantificação 32 e 23 mg kg-1 e coeficiente de variação 4,8 e 2,6% para o nitrito e nitrato, respectivamente. Os coeficientes de correlação "r" foram acima de 0,995. O método desenvolvido mostrou-se eficiente para análise de nitrito e nitrato em salsicha.
