The baru (Dipteryx alata Vogel), is fruit tree displaying economic, environmental, industrial and food importance. The objective of this study was to adapt to the tetrazolium test methodology to baru seeds and evaluate the physiological quality of seeds stored for 12 months at 10 and 20 ºC, using X-ray images as a toll for detection damage throughout the storage period while also carrying out germination, emergence and electrical conductivity tests. Baru seeds remained viable for 12 months of storage, albeit with a small decrease in vigor after nine months. The applied tetrazolium test methodology was efficient in determining internal seed damage. The pre-soak time for the tetrazolium test and solution concentration was 24 hours and 0.05%. No changes in seed morphology and radiographic images were observed during the storage periods, regardless of the assessed temperature. The X-ray test enabled a precise visualization of internal baru seed structures.

A avaliação de sementes e plântulas em vias experimentais é uma importante fonte de conhecimentos úteis para a produção agrícola sustentável. Neste sentido este artigo tem como base a reunião e compilação de informação sobre os principais métodos de avaliação de experimentos com sementes, em relação à germinação e ao vigor. Para tal, foram enumerados pontos como a qualidade de sementes, protocolos experimentais e suas respectivas exemplificações. Assim, pode ser percebido que a utilização de outros testes e variáveis podem ser ligadas a experimentação com sementes, permitindo um uso melhor dos planos experimentais a fim de contribuir com uma melhor caracterização a este processo científico. A pesquisa com sementes pode ser melhor otimizada para que seus resultados possam ser entendidos e aplicados no ambiente agrícola com maior ênfase.

Protocolos de testes de germinação e tetrazólio padronizados são importantes para a análise de sementes, inclusive para a espécie Aspidosperma pyrifolium, esseencia florestal de valor econômico e ainda pouco estudada segundo a literatura, sendo, portanto, este o objetivo do presente trabalho. Assim, foram testados protocolos de Germinação (efeito de substratos e temperaturas) e Tetrazólio (efeitos de tempos de embebição e de imersão em concentrações da solução de tetrazólio). Para o teste de Germinação, os substratos rolo de papel e vermiculita e as temperaturas de 25, 30 e 25-30ºC apresentaram os melhores valores (médias entre 65-90% de germinação). No teste de tetrazólio o tratamento com 10h de embebição e 3h de imersão na solução com concentração de 0,075% (média de 90% de viabilidade) foi o melhor. Foi constatada correlação positiva (Pearson) entre a utilização do substrato papel a 25ºC e o referido tratamento de Tetrazólio. Ambos protocolos descritos [Germinação (substrato papel a 25ºC por oito dias) e Tetrazolio (10h de embebição e 3h de imersão na solução com concentração de 0,075%)] podem testados em sementes de outras populações desta espécie para a verificação da efetividade deste teste.
