Validation of a new high‐throughput method to determine urinary S‐phenylmercapturic acid using low‐temperature partitioning extraction and ultra high performance liquid chromatography–mass spectrometry

Abstract: A new highly sensitive and environmentally friendly analytical method, using low-temperature partition extraction and ultra-high-performance liquid chromatography with tandem mass spectrometry, without the use of a labeled analyte, was developed and validated to determine and quantify urinary S-phenylmercapturic acid in urine samples. The World Health Organization, in its guidelines for air quality in Europe, recognizes that benzene is carcinogenic to humans and there is no safe level of exposure. Urinary S-ph… Show more

“…The determination of urinary S-PMA was performed with an Acquity ultra performance liquid chromatography (UPLC) system coupled with a triple quadrupole mass spectrometer (UPLC–MS–MS, Waters, Milford, MA, USA) using a recently published modified method [ 35 ]. For chromatographic separation, we used a UPLC HSS C18 column (2.1 × 150 mm, 1.7 μm).…”

“…The herein‐adopted LTPE‐based S ‐PMA extraction method was modified from the method previously described by Gomes et al. [22]. First, 600 μL of fortified urine pool ( S ‐PMA 25.0 μg L −1 ) was added to a plastic tube and acidified with hydrochloric acid at 25% (pH 1) for extraction.…”

“…S-PMA has been analyzed using several chromatography techniques, such as HPLC/UV, HPLC-MS/MS, and GC/MS. Furthermore, the derivatization step and different extraction techniques, such as liquid-liquid extraction (LLE), solid-phase extraction (SPE), solid-phase microextraction, and low-temperature partiture extraction (LTPE), are used for the analysis of this compound [9,[15][16][17][18][19][20][21][22]. Using internal standards is a routine practice adopted to increase the accuracy of the method developed [23].…”

“…The method used is called extraction with low-temperature liquid-liquid partitioning and is described by Gomes et al [37] (with modifications). Briefly, the method consisted of extracting the urinary S -PMA from 600 µL urine (whose pH had been corrected to 1 with HCl 25%).…”
