Use of clomipramine as chemotherapy of the chronic phase of Chagas disease

Abstract: Chagas infection is a major endemic disease affecting Latin American countries. The persistence of Trypanosoma cruzi generates a chronic inflammatory reactivity that induces an immune response directed to the host's tissues. The effectiveness of the treatment in the chronic phase is still unsatisfactory due, amongst other reasons, to the collateral effects of the drugs used. We investigated the effect of clomipramine, a tricyclic antidepressant that, when used as a treatment of T. cruzi-chronically infected mi… Show more

“…Particularly, BZ produces free radicals and/or electrophilic metabolites and induces oxidative stress in the parasite (17). On the other hand, CMP inhibits trypanothione reductase (22,40,43), an essential component of the antioxidant defenses of T. cruzi. As a consequence, it is plausible to think that these mechanisms would work together against the parasite.…”

“…In particular, clomipramine (CMP), a tricyclic antidepressant, inhibits trypanothione reductase activity and has evidenced antitrypanocidal activity when used alone (22,49) or in combination with BZ (23). Furthermore, considering the possible existence of resistant parasite strains and more than one strain in the same patient, combined treatment with two drugs bearing different mechanisms of action might be a strategy to improve the pharmacotherapy of Chagas disease (14,23,24).…”

“…As with phenothiazines, these compounds have been previously reported to inhibit growth of T. cruzi [24], including a study showing activity of clomipramine in the mouse model of chronic T. cruzi infection [25][27]. The tricyclic antidepressants, similar to phenothiazines in structure, have also been shown to inhibit trypanothione reductase [28].…”
