a b s t r a c tThe marine bacterium Acinetobacter sp. strain HC8-3S, capable of degrading petroleum hydrocarbons, was previously shown to be applicable for bioremediation. Here, we evaluated the performance of the strain during industrial-scale fermentation, long-term storage, and biodegradation processes. Fermentation parameters were optimized for industrial-scale culturing using lower salinity and fewer inorganic salts in the culture medium. The shelf-life of the fermentation broth was evaluated in different storage conditions. Biodegradation efficiency of the strain was evaluated using gas chromatography. Results indicated that the optimum conditions for one-ton scale fermentation were 30 C, pH 7.4e7.8, and rotation speed 90 rpm; the cell density reached 3.6 ± 1.9 Â 10 10 CFU ml À1 after 12 h. The low temperature can preserved fermentation broth for longer time. When the storage temperature was down to 4 C from room temperature, half-life of strain HC8-3S extended from 9 days to 34 days. The biodegradation rate of the saturated hydrocarbon fraction of crude oil was 94% after treatment with the strain for 5 days. The results indicate that Acinetobacter sp. strain HC8-3S can be enriched efficiently on a large scale, making it a potentially useful industrial strain. This crude oil degradative capability of the strain HC8-3S provides possible application for the clean-up of crude oil-contaminated environment.

Background Soil contamination with petroleum has long been a major environmental problem. Many methods have been developed for its remediation, including bioremediation, one of the most applied procedures. The biodegradation rate of bioremediation depends on the soil environment and can be enhanced by microorganism immobilization. Here, a modified biofuel ash was proposed as a new highly efficient and inexpensive immobilization carrier and tested experimentally in soil systems. Results The degradation rate of petroleum hydrocarbon in soil by inoculated microorganisms immobilized on humic acid‐modified biofuel ash reached up to 47.8% during 60 days, which was twice as high as that by free microorganisms. In treatments with immobilized microbes, n‐alkanes with C28 and C31–34 were not detected and the degradation rates of C17–19 were 70.6% (unsterilized soil) and 54.2% (sterilized soil). The change in enzymatic activity reflected the basic respiration rate: it gradually increased in the pre‐remediation stage, then decreased, and finally stabilized. Microorganism immobilization carriers had no significant contribution to microbial diversity in unsterilized soil, but they increased in sterilized soil. Conclusions Humic acid‐modified biofuel ash inoculated with microorganisms improves the degradability of petroleum and biodiversity in soil without microbes and, through the application of biofuel ash, reuses power plant waste. © 2019 Society of Chemical Industry

BACKGROUND: Cleaning up the environment by the elimination of harmful contaminants from textile effluents (TE) is a critical and challenging crisis that requires long-term appropriate solutions. In this work, a fungal consortium-GR containing two fungi Geotrichum candidum F4 and Rhodotorula glutinis F7, in the ratio 1:1, is demonstrated to treat the organic contaminants in TEs. RESULTS:The immobilized consortium-GR in coconut fibre is 24% more efficient at treating TE than free consortium-GR, showing a maximum decolorization of 88.4% in 6 h immersion, at optimal conditions of pH 5.5, temperature 35°C, inoculum size 5% and fibre quantity 2.5 g L −1 . Biodegradation was confirmed by using UV-visible spectrophotometry, high-performance liquid chromatography and Fourier transform infrared spectroscopy, and the degradation pathway of methyl orange was confirmed by gas chromatography-mass spectroscopy study. The novel immobilized consortium-GR in the bioreactor was assessed by the reduction of toxic parameters in 6 h: colour by 70%, total dissolved solids by 62%, total suspended solids by 81%, biological oxygen demand by 51%, chemical oxygen demand by 58%, chloride by 78%, ammonia by 64%, sulfate by 55%, lead by 12.5% and chromium by 75%. During this process of bioconversion, maximum laccase activity of 3.25 U L −1 was observed in 6 h by immobilized consortium-GR. Phytotoxicity and genotoxicity studies revealed the detoxification of TE. CONCLUSION: Through the continuous reuse of treated water in subsequent dyeings, the proposed TE treatment process represented a 92.86% reduction in water use and a total cost reduction of 22.47%.This study is a step towards upcoming onsite, field and short-term applications of consortium-GR for the management of TE.
