Although the changes in urinary glycosaminoglycans have been investigated in several endocrinopathies, no information was hitherto available on the content and composition of urinary glycosaminoglycans in hypothyroidisin. Urinary glycosaminoglycans were therefore investigated in patients with hypo thy roidism and in healthy subjects. The total daily excretion of urinary glycosaminoglycans was found to be significantly increased (by 41 %) in hypothyroidism. Two electrophoretic bands were always detected in both examined groups: a major band of chondroitin sulphate and a minor band of heparan sulphate. Heparan sulphate and chondroitin sulphate levels were respectively 114% and 42% higher in patients with hypothyroidism than in controls. The respective increases in chondroitin-4-sulphate and chondroitin-6-sulphate were 31% and 41%. The relative quantities of chondroitin-4-sulphate, dermatan sulphate, chondroitin-6-sulphate and non-sulphated chondroitin sulphate were unchanged in the two examined groups. The changes observed in the levels of the excreted glycosaminoglycans may reflect the altered metabolism of connective tissue in hypothyroidism.

We have successfully synthesized a lipid containing the pyranine dye as the hydrophilic head group. This lipid was incorporated into liposomes with 1-palmitoyl-sn-glycero-3-phosphocholine as the major component. The resultant liposomes displayed differential enhancements in fluorescence emission intensity in the presence of nanomolar concentrations of different glycosaminoglycans. Linear discriminant analysis of the fluorescence response data demonstrate that the liposomes are able to distinguish between different GAGs. In addition, we also demonstrate that the liposomes incorporating the pyranine lipid are able to distinguish between dilute serum from healthy individuals and serum containing elevated chondroitin sulfate (simulated serum from an Alzheimer’s disease patient).

The neutral sugars and hcxosamines in the seromucoid fraction of normal human serum were identified and determined simultaneously by gas-liquid chromatography of their aldononitrilc acctates. Besidcs the sialic acid, normal seromucoid contained fucose, arabinose, mannosc, glucose, galactose, glucosamine and galactosamine. The presence of arabinose and glucose, not previously reported in seromucoid, äs well äs the other known carbohydrate components determined by GLC were confirmed from their fragmentation patterns in the electron impact mass spectra and chemical ionization spcctra. Die Protein-gebundenen Kohlenhydrate von Seromucoid aus normalem Serum des Menschen Zusammenfassung: Die Neutralzucker und Hexosamine in der Seromucoidfraktion aus normalen Serum des Menschen wurden durch Gaschromatographie ihrer Aldononitrilacetate identifiziert und gleichzeitig bestimmt. Neben Neuraminsäure enthielt normales Seromucoid Fucose, Arabinose, Mannose, Glucose, Galaktose, Glucosamin und Galaktosamin. Das bisher nicht mitgeteilte Vorkommen von Arabinose und Glucose in Seromucoid als auch die anderen durch Gaschromatographie bestimmten Kohlenhydrat-Komponenten wurden durch ihre Fragmentierungsmuster im Elektronenstoß-und chemischen lonisations-Massenspektrum bestätigt.
