TRPM2 Promotes Atherosclerotic Progression in a Mouse Model of Atherosclerosis

Abstract: Atherosclerosis is a chronic inflammatory arterial disease characterized by build-up of atheromatous plaque, which narrows the lumen of arteries. Hypercholesterolemia and excessive oxidative stress in arterial walls are among the main causative factors of atherosclerosis. Transient receptor potential channel M2 (TRPM2) is a Ca2+-permeable cation channel activated by oxidative stress. However, the role of TRPM2 in atherosclerosis in animal models is not well studied. In the present study, with the use of adeno-… Show more

“…Hematoxylin–eosin (HE) staining was adopted to analyze the ratio of the aortic arch injury area to the total vessel area. The ratio of plaque area to total vessel area was assessed with Oil red O staining 23,24 . Three sections per mouse in each group were selected for analysis.…”

“…Cardiac and liver lipid accumulation was measured qualitatively using Oil Red O staining as described previously. 28 Briefly, serial 8 μm sections were washed with deionized water, immersed in 60% isopropyl alcohol, and incubated with Oil Red O staining solution. The slides were immersed in 60% isopropanol again, washed with deionized water, and then counterstained with hematoxylin.…”

“…TRPM2 interacts with extra synaptic NMDA receptors (NMDAR) and enhances NMDAR activity in ischemic stroke [1164]. Activation of TRPM2 in macrophages promotes atherosclerosis [1165,1147]. Moreover, silica nanoparticles induce lung inflammation in mice via ROS/PARP/TRPM2 signaling-mediated lysosome impairment and autophagy dysfunction [1028].…”
