Summary The Chinese jujube (Ziziphus jujuba Mill.), a member of the Rhamnaceae family, is an important perennial fruit tree crop of substantial economic, ecological and nutritional value, and is also used as a traditional herbal medicine. Here, we report the resequencing of 493 jujube accessions, including 202 wild and 291 cultivated accessions at >16× depth. Our population genomic analyses revealed that the Shanxi–Shaanxi area of China was jujube's primary domestication centre and that jujube was then disseminated into East China before finally extending into South China. Divergence events analysis indicated that Ziziphus acidojujuba and Ziziphus jujuba diverged around 2.7 Mya, suggesting the interesting possibility that a long pre‐domestication period may have occurred prior to human intervention. Using the large genetic polymorphism data set, we identified a 15‐bp tandem insertion in the promoter of the jujube ortholog of the POLLEN DEFECTIVE IN GUIDANCE 1 (POD1) gene, which was strongly associated with seed‐setting rate. Integrating genome‐wide association study (GWAS), transcriptome data, expression analysis and transgenic validation in tomato, we identified a DA3/UBIQUITIN‐SPECIFIC PROTEASE 14 (UBP14) ortholog, which negatively regulate fruit weight in jujube. We also identified candidate genes, which have likely influenced the selection of fruit sweetness and crispness texture traits among fresh and dry jujubes. Our study not only illuminates the genetic basis of jujube evolution and domestication and provides a deep and rich genomic resource to facilitate both crop improvement and hypothesis‐driven basic research, but also identifies multiple agriculturally important genes for this unique perennial tree fruit species.

Ultrasound Assisted Extraction of Phenolic Compounds from a Jujube By-Product with Valuable Bioactivities

Jujube plant is a potential source of polyphenols with biological propreties. The purpose of this study was to investigate the application of ultrasound technique for extracting phenolic compounds (TPC) from seeds of Zizyphus lotus under optimization conditions based on response surface methodology. A maximum TPC, total flavonoids content (TFC), and total condensed tannins content (TTC) of 2383.10 ± 0.87 mg GAE/100g, 486.50 ± 0.38 mg QE/100g and 15,787.10 ± 0.10 mg CE/100g, respectively obtained under ethanol concentration 50.16%, sonication temperature 29.01 °C, sonication time 15.94 min and solvent-to-solid ratio 34.10:1 mL/g. The optimized extract was then evaluated for its antioxidant, antiacethylcholinesterase, antihypercholesterolemia, and antiproliferative activities. The results showed that ultrasound method is a green and safe method that can be used to effectively extract TPC from jujube seeds. The biological activity of Zizyphus extract exhibited a very good antioxidant against DPPH (EC50 = 0.39 µg/mL) and FRAP (1670.42 ± 6.5 mg/100 g). Additionally, it possesses acetylcholinesterase (AChE) inhibitory effect (IC50 = 0.93 ± 0.01 mg/mL) and HMGR inhibition (45.41%) using 100 µg/mL. The extract significantly inhibits cell proliferation on the MCF-7 and HepG2 tumor cell lines with an IC50 values of <0.05 and 3 ± 0.55 mg/mL, respectively. Therefore, the ultrasound method can be considered a method for obtaining a significant anticancer activity with respect to the lines and therefore makes it possible to recover a maximum of phenolic compounds in less time with an AChE and HMGR inhibitory activity. Thus, it can be suggested that Zls extract is a promising fruit for the development of supplementary dietary due to its potential behaviour as nutraceutical.

One of the most important attributes of a genome is genome size, which can to a large extent reflect the evolutionary history and diversity of a plant species. However, studies on genome size diversity within a species are still very limited. This study aims to clarify the variation in genome sizes of Chinese jujube and sour jujube, and to characterize if there exists an association between genome sizes and geographical variation. We measured the genome sizes of 301 cultivars of Chinese jujube and 81 genotypes of sour jujube by flow cytometry. Ten fruit traits, including weight, vertical diameter, horizontal diameter, size, total acids, total sugar, monosaccharide, disaccharide, soluble solids, and ascorbic acid were measured in 243 cultivars of Chinese jujube. The estimated genome sizes of Chinese jujube cultivars ranged from 300.77 Mb to 640.94 Mb, with an average of 408.54 Mb, with the highest number of cultivars (20.93%) falling in the range of 334.787 to 368.804 Mb. The genome size is somewhat different with geographical distribution. The results showed weakly significant positive correlation (p < 0.05) between genome size and fruit size, vertical diameter, horizontal diameter, and weight in the Chinese jujube. The estimated sour jujube genome sizes ranged from 346.93 Mb to 489.44 Mb, with the highest number of genotypes (24.69%) falling in the range of 418.185 to 432.436 Mb. The average genome size of sour jujube genotypes is 423.55 Mb, 15 Mb larger than that of Chinese jujube. There exists a high level of variation in genome sizes within both Chinese jujube cultivars and sour jujube genotypes. Genome contraction may have been occurred during the domestication of Chinese jujube. This study is the first large-scale investigation of genome size variation in both Chinese jujube and sour jujube, which has provided useful resources and data for the characterization of genome evolution within a species and during domestication in plants.
