To explore the mechanism of You-Gui-Wan (YGW) in improving endometrial receptivity in mouse model of controlled ovarian hyperstimulation (COH). Methods: Sixty female mice with two consecutive oestrous cycles were randomly divided into COH model group (n = 50) and normal control group (n = 10). The COH model was created by using GnRH-a + PMSG + HCG and further randomly divided into 5 subgroups of simple model, YGW high-dose, YGW medium-dose, YGW low-dose and aspirin control, 10 in each subgroup. YGW or aspirin or saline was administrated to the mice by gavage once a day for 11 days. Afterwards, blood and uterine samples were collected for examination of serum levels of estradiol (E2) and progesterone (P4), endometrial thickness and number of blood vessels as well as expressions of angiogenic factors of VEGF, Ang-1, Ang-2 and Tie-2 at transcription and translation levels by using H& E staining, ELISA, real-time qRT-PCR, and immunohistochemistry, respectively. Wei X, et al 2 Genetics and Molecular Research 16 (4): gmr16039833 Results: COH per se increased the serum concentrations of E2 and P4，but decreased the number of endometrial blood vessels and the protein expressions of the angiogenic factors when compared to the normal control group (P< 0.05 or 0.01). YGW increased the mRNA expressions of all angiogenic factors and restored the effect of COH on number of endometrial blood vessels (P< 0.01), and the protein expressions of the angiogenic factors when compared to the COH simple model group (P< 0.05 or 0.01). In comparison, aspirin could only restore the number of endometrial blood vessels and the protein expressions of Ang-2 and Tie-2 (P< 0.05 or 0.01). Conclusions: YGW can restore the side effect of COH on number of endometrial blood vessels and the expressions of angiogenic factors, thus improve endometrial receptivity in COH mice.

Purpose. This study was aimed to investigate the reproductive toxicity of Zishen Yutai Pill (ZYP) on fertility and early embryonic development in rats. Methods. SD rats were randomly divided into 5 groups: vehicle control group (distilled water, i.g.), positive control group (80 mg/kg of cyclophosphamide, i.p.), and three ZYP-treated groups (3, 6, and 12 g/kg/d, i.e., 12x, 24x, and 48x clinical doses, i.g.). The high dose was set as the maximum gavage dosage. Results. Cyclophosphamide showed diverse hazards, such as decreased weight of male reproductive organs and sperm density (P < 0.05). However, there were no obvious effects of ZYP on physical signs, animal behavior, and survival rate, as well as on weight and food intake during the premating and gestation periods. Importantly, there were no significant adverse effects of ZYP on indexes of copulation, fecundity and fertility indexes, weights and coefficients of male reproductive organs, epididymal sperm number and motility, estrous cycle, preimplantation loss rate, and implantation rate. Besides, the numbers of live and resorbed fetuses per litter were not significantly altered. Conclusions. ZYP had no reproductive toxicities on fertility and early embryonic development in rats at 48x equivalent clinical doses.

Objective: During the preparation of Zishen Yutai Pills, 5-Hydroxymethylfurfural may be used. To establish a HPLC method for determination of 5-HMF in Zishen Yutai pill and to provide the potential quality control standard of it. Methods: RP-HPLC method was applied with the chromatographic condition as follows: the chromatographic column was Phenomenex Gemini C18 column (250×4.6mm, 5µm), methanol-water (10:90) as the mobile phrase, the flow rate was 0.8mL/min, the detection wavelength was 284nm, the column temperature was 30°C, and the injection volume was 10µL. Results: The calibration curve was linear in the range of 2.16~43.27µg. mL -1 for 5-HMF (r 2 =0.9993). The RSD of the method repeatability was 0.97% (n=6) and the average recovery was 99.4% (RSD=0.58%). What's more, the specificity, precision and stability results were also good. Conclusion: The method for HPLC is not only sensitive and accurate. It is also easy to operate which can be used as the determination method for 5-HMF and has a guiding role for the quality control of the Zishen Yutai pill.
