“Total Synthesis” Supramolecular Style: Design and Hydrogen-Bond-Directed Assembly of Ternary Supermolecules

Abstract: Getting the right balance between intermolecular interactions is crucial for the synthesis of supermolecules in a preconceived manner. The three‐component supermolecule (see picture) in the ternary cocrystal of 3,5‐dinitrobenzoic acid, isonicotinamide, and 4‐(dimethylamino)benzoic acid (1:1:1) assembles through a “primary” (between the stronger acid and pyridine) and a “secondary” hydrogen‐bonding interaction (between the weaker acid and amide).

“…[12][13][14][15][16] Benzoic acid (BA) and isonicotinamide (isoNCT) have both been widely explored as co-formers for pharmaceutical co-crystal synthesis. [17][18][19][20][21] They are known to form both 1:1 (BA:isoNCT) 22 and 2:1 (BA 2 :isoNCT) 18 co-crystals. Possible synthetic routes to these include solution methods, 18,22 melt crystallisation, 21,23 template synthesis using BA crystals as a substrate, 21 and thermal inkjet printing.…”

“…Since the odds are stacked firmly in favor of a homomeric product, how do we go about developing reliable and versatile synthetic methods for the directed assembly of co-crystals? This presentation will attempt to answer the question by outlining several modular hydrogen-bond driven strategies for the design and synthesis of binary and ternary supermolecules and co-crystals [1][2][3].…”

“…It is trivial with the existing search software [1] to find all CSD entries containing a particular solvent, say water, and a survey of the 50 most common solvents in the CSD has been published [2]. It might be interesting, however, to know if any of these solvates also exist in an unsolvated form; given a crystal structure of di-isopropylammonium chloride hydrate in the CSD, for example, one could wonder if a crystal structure of di-isopropylammonium chloride itself is present in the database.…”
