Lactuca indica is an edible wild vegetable, which is frequently availed as a folk remedy for its anti-inflammatory, antibacterial, and other treatments in Asia. This study was executed to evaluate the cytotoxicity, antioxidant and phenolic contents of the methanol extracts from different organs of Lactuca indica. The highest amount of phenolics found in the methanol extracts of leaf (35.09 ± 0.15 mg/g), followed by stem (15.44 ± 0.20 mg/g), root (13.50 ± 0.19 mg/g) and flower (12.50 ± 0.39 mg/g) while the highest flavonoid contents also observed in the methanol extracts of leaf (26.90 ± 0.22 mg/g). Methanol extracts of leaf showed a pronounced DPPH radical scavenging activity (90.37 ± 0.15 mg/mL) and the ABTS radical scavenging activity (99.84 ± 0.02 mg/mL) at 10 mg/mL and 20 mg/mL respectively. Using MTT assay, the methanol extracts of leaf showed the highest cytotoxicity (IC 50 113.84 μg/mL) against human breast adenocarcinoma cell (MCF-7). To this end, the results revealed that the phenolic contents were highly correlated with the DPPH and ABTS radical scavenging activity. The findings suggest that it could contribute to the antioxidant properties of different organs of the studied plant. In this regard, the cytotoxicity and antioxidant properties could be helpful for the evaluation of the bioactive properties of Lactuca indica.

Abstract. The present study assessed the chemical composition, antioxidant properties, and hepatoprotective effects of subacute pre-treatment with chamomile (Matricaria recutita L.) decoction extract (CDE) against ethanol (EtOH)-induced oxidative stress in rats. The colorimetric analysis demonstrated that the CDE is rich in total polyphenols, total flavonoids and condensed tannins, and exhibited an important in vitro antioxidant activity. The use of LC/MS technique allowed us to identify 10 phenolic compounds in CDE. We found that CDE pretreatment, in vivo, protected against EtOH-induced liver injury evident by plasma transaminases activity and preservation of the hepatic tissue structure. The CDE counteracted EtOH-induced liver lipoperoxidation, preserved thiol -SH groups and prevented the depletion of antioxidant enzyme activity of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). We also showed that acute alcohol administration increased tissue and plasma hydrogen peroxide (H 2 O 2 ), calcium and free iron levels. More importantly, CDE pre-treatment reversed all EtOH-induced disturbances in intracellular mediators. In conclusion, our data suggest that CDE exerted a potential hepatoprotective effect against EtOH-induced oxidative stress in rat, at least in part, by negatively regulating Fenton reaction components such as H 2 O 2 and free iron, which are known to lead to cytotoxicity mediated by intracellular calcium deregulation.

Abstract. Hypericum ascyron L. (Great St. Johnswort), which belongs to the Hypericaceae family, has been used for the treatment of hematemesis, metrorrhagia, rheumatism, swelling, stomach ache, abscesses, dysentery and irregular menstruation for >2,000 years in China. The aim of the present study was to clarify the anticancer activity compounds from H. ascyron L. and the underlying molecular mechanism. Anticancer activity of H. ascyron L. extract was evaluated using an MTT assay. To confirm the anticancer mechanism of activity compounds, Hoechst 33258, Annexin V-fluorescein isothiocyanate/propidium iodide, 2',7'-dichlorodihydrofluorescein diacetate, rhodamine 123 staining and caspase-3 activity analysis were performed. The results demonstrated that the anti-proliferative action of the mixture of kaempferol 3-O-β-(2"-acetyl) galactopyranoside (K) and quercetin (Q) (molar ratio, 1:1) was significantly increased compared with either of these two compounds separately, and the active fraction of the H. ascyron L. extract |(HALE). HALE, indicating that the anti-proliferative function of H. ascyron L. may be a synergic effect of K and Q. Furthermore, the inhibitory effect of KQ on the growth of HeLa cells was mediated by the induction of apoptosis. To the best of our knowledge, the present study is the first to identify that KQ exhibits significant anti-proliferation activity on HeLa cells via the apoptotic pathway, and is also the first to evaluate the anticancer potential of H. ascyron L. The results of the present study may provide a rational base for the use of H. ascyron L. in the clinic, and shed light on the development of novel anticancer drugs.
