Daun alpukat mengandung komponen bioaktif yang tinggi sehingga dapat dimanfaatkan sebagai sumber antioksidan alami. Tujuan dari penelitian ini adalah mendapatkan metode pengeringan yang tepat sehingga menghasilkan ekstrak daun alpukat dengan aktivitas antioksidan yang tinggi. Daun alpukat baik yang muda maupun yang tua dikeringkan dengan tiga metode pengeringan yaitu dijemur dibawah sinar matahari, dikeringanginkan dalam ruangan, dan dikeringkan dengan oven. Kualitas daun kering kemudian dianalisis aktivitas antioksidan, kadar air, total fenol, total flavonoid, dan total tanin. Hasil penelitian menunjukkan bahwa daun tua yang dikeringkan dengan menggunakan oven pada suhu 40˚C selama 24 jam menghasilkan aktivitas penghambatan radikal bebas tertinggi yaitu 19,83% dengan kadar air 7,54%, total fenol 6,42 mg/100 g ekstrak, total flavonoid 12,07 mg/100 g ekstrak, dan total tanin 2,48 mg/100 g ekstrak. Kesimpulannya, metode pengeringan dapat memberikan dampak terhadap kadar senyawa bioaktif ekstrak daun alpukat dan aktivitas antioksidannya.Effect of Drying Methods on the Antioxidant Activity of Avocado LeavesAbstractAvocado leaves contain high bioactive components that may be used as a source of natural antioxidants. The purpose of this study was to obtain proper drying method to produce avocado leaf extract with high antioxidant activity. Avocado leaves, both young and old, were dried with three drying methods, which were dried in the sun, room, and oven. Quality of leaves was then measured for antioxidant activity, moisture, total phenol, total flavonoid, total tannins. The results showed that old leaves dried using an oven at 40˚C for 24 hours resulted in the highest radical scavenging activity of 19.83% with 7.54% moisture content, 6.42 mg/100 g extract of total phenol, 12.07 mg/100 g extract of total flavonoid, and 2.48 mg/100 g extract of total tannins. In conclusion, the drying methods might produced specific antioxidant activities.

Seeds from different indigenous watermelons were analysed for their chemical composition such as phenolics, proteins, oils, minerals, ash and fibre content as well as antioxidant activity measured in methanol extract using 2.2-diphenyl-1-picrylhydrazyl (DPPH) assay. The morphological variations of watermelons, which revealed diversity in terms of fruit and seed colour were also assessed. Average percentages for lipid content, total proteins, crude fibre, ash and total carbohydrates as 34.4, 16.5, 23.1, 3.99, and 3.16%, respectively, were recorded for Capricorn district and 31.6, 14.9, 22.0, 4.58, and 5.26% were recorded for Sekhukhune landraces. The Capricorn landraces had higher content of flavonoids (0.222 mg/g) than Sekhukhune varieties (0.130 mg/g). But, landraces from Sekhukhune had higher antioxidant activity (46.5%) which corresponded to higher total phenolic content of 0.59 mg/g. Thus, suggest that watermelon seeds may serve as potential source of natural products for food, feed and pharmaceutical applications.

Statistical Modelling of Ultrasonic-Aided Extraction of <i>Elaeis guineensis</i> Leaves for Better-Quality Yield and Total Phenolic Content

The present study highlighted the statistical modeling of an ultrasonic-aided extraction (UAE) of Elaeis guineensis leaves extract for maximal extraction yield (EY) and total phenolic content (TPC). A Box-Behnken design investigated the effects of ethanol concentration (X1: 0−100%), extraction time (X2: 5−55 min), the solvent-to-solid ratio (X3: 15:1−35:1 mL/g) and sonification amplitude (X4: 20−100 %). Under optimized conditions, the highest EY of 14.38% was attained using 50% (v/v) ethanol:water ratio, 55 min, 35 mL/g solvent-to-solid ratio, 60% sonication amplitude, whereas maximum TPC was 209.70 mg gallic acid equivalent (GAE)/g [50% (v/v) ethanol:water ratio, 30 min, 25 mL/g solvent-to-solid ratio, 60% sonication amplitude]. Second-order polynomial models of EY and TPC showed the R2 value corresponding to 0.9303 and 0.9500, respectively, indicating their significance (p < 0.05) to predict the responses. HPLC chromatograms revealed gallic acid and catechin were present in the UAE extracts. UAE technique afforded better EY (14.38%) and TPC (209.70 mg GAE/g) than maceration (3.73%, 85.23 mg GAE/g) and Soxhlet (6.86%, 102.13 mg GAE/g) extractions, as based on scanning electron micrographs of untreated, UAE, macerated and Soxhlet treated samples. Cell walls of ultrasonic-treated E. guineensis leaves were visibly disrupted to facilitate the higher release of bioactive plant materials, thus justifying the higher EY and TPC. The application of ultrasound appeared to remarkably increase the extraction efficiency of E. guineensis leaves to extract as compared to the conventional methods.
