TNF-alpha expression, evaluation of collagen, and TUNEL of Matricaria recutita L. extract and triamcinolone on oral ulcer in diabetic rats

Abstract: Diabetes mellitus (DM) is a disease associated with delayed wound healing of oral ulcers by increased expression of proinflammatory cytokines and cellular apoptosis. Objective to evaluate the influence of Tumor Necrosis Factor alpha (TNF-α) and apoptosis in rats with DM treated with chamomile extract or triamcinolone.Material and Methods Wistar male rats (210.0±4.2 g) were divided into five groups: negative control group (NCG) without diabetes; positive control group (PCG) with DM (alloxan, 45 mg/kg); and grou… Show more

“…However, the triamcinolone was superior to M. recutita on the 6 th day [ 33 ]. This finding supports the results found in the study that showed that the extract provided pain relief in 82% of patients with stomatitis after 5, 10, and 15 minutes; the authors concluded that the analgesic effect improved the patients' quality of life [ 21 ].…”

“…The ulcers were measured using a digital pachymeter, and all measurements were performed by the same examiner. The largest (D) and smallest (d) diameters of the lesion were recorded and the area of the ulcer was calculated as previously described in the literature (Brizeno et al, ; Lim et al, ; Oliveira et al, ), using the following formula: A = π × D/2 × d/2.…”

“…However, it is known that, when in excess, it can delay healing (Brizeno et al, ; Oliveira et al, ) and result in a greater production of free radicals that may have toxic effects on tissues (Kasuya & Tokura, ). Thus, the modulation of the release of proinflammatory cytokines is a challenging goal when the aim is to optimize healing time and to reduce pain symptoms of ulcerative lesions (Oliveira et al, ; Wagner et al, ). Several studies have investigated the development of an ideal healing agent, although there is still no widely accepted or gold standard treatment for the management of oral ulcers.…”

“…Though it may be temporary, the NGF expression almost reached the baseline level and had a rising trend in dorsal horn and DRG respectively, which could be explained by the compensatory expression of NGF. For example, the expression of tumor necrosis factor alpha (TNF-α) increases in rats with diabetes (24), and TNF-α can promote the release of NGF in astrocytes (25). In addition, the hyperalgesia was observed 1, 2, 4, and 8 weeks after diabetes induction, consistent with the time of NGF decrease in DRG.…”
